Introduction. Substance abuse significantly influences human health and may induce problems with social functioning worldwide. Numerous genetic and environmental risk factors, as well as their interactions, accelerate the development of drug addiction. Etiologically, the dopaminergic mesocorticolimbic reward pathways are related to psychoactive substance addiction, and the reward properties of heroin are connected with changes in the mesolimbic dopaminergic system. Objective. The aim of this study is a haplotypic analysis of subjects addicted to polysubstance. However, with the knowledge that this is not a homogenous subgroup, it was decided to separate and analyze homogenous subgroups of subjects in order to find specific haplotypic variants among them. The subjects in the subgroups were addicted to heroin, and subjects with more than two relapses in the past two years. Materials and method. The study group comprised of 301 polysubstance addicted rural male subjects. From this group, 2 homogenous subgroups of subjects were isolated and additionally analyzed: (1) a group of heroin addicted subjects (n=61), and (2) a group of heroin-addicted subjects with at least two relapses in the last two years (n=21). The group consisting of all polysubstance addicted rural subjects and both homogenous subgroups were analyzed against a control group of non-addicted subjects (n=300), matching gender and age. Five polymorphisms in the DRD2/ANKK1 region were analyzed: rs1076560, rs1800498, rs1079597, rs6276 in the DRD2 gene, and rs1800497 in the ANKK1 gene. Results. A statistically significant haplotype association was found in analysis of the heroin addicted subjects, compared to controls, and two possible trends – when comparing the whole group of addicted subjects to controls, and in relapse subgroups, compared to the controls. Conclusion. The results obtained showed that haplotypes indicate a part of the biological component of addiction.

In the paper, the development and performance of an optical sensor for detection of neurotransmitters (dopamine) is presented. The concentration of dopamine is measured basing on fluorescence quenching of graphene quantum dots. In the sensor, the dopamine molecules coat the graphene quantum dots surface – in result, the quenching of fluorescence occurs due to the Förster resonance energy transfer. The changes in fluorescence correspond to specific concentrations of the neurotransmitter in tested samples, so it is possible to accurately determine the concentration of dopamine in the sample.

Molybdenum-containing enzymes, aldehyde oxidase and xanthine oxidase, are important in the oxidation of N-heterocyclic xenobiotics. However, the role of these enzymes in the oxidation of drug-derived aldehydes has not been established. The present investigation describes the interaction of eleven structurally related benzaldehydes with guinea pig liver aldehyde oxidase and bovine milk xanthine oxidase, since they have similar substrate specificity to human molybdenum hydroxylases. The compounds under test included mono-hydroxy and mono-methoxy benzaldehydes as well as 3,4-dihydroxy-, 3-hydroxy-4-methoxy-, 4-hydroxy-3-methoxy-, and 3,4-dimethoxy-benzaldehydes. In addition, various amines and catechols were tested with the molybdenum hydroxylases as inhibitors of benzaldehyde oxidation. The kinetic constants have shown that hydroxy-, and methoxy-benzaldehydes are excellent substrates for aldehyde oxidase (Km values 5×10-6 M to 1×10-5 M) with lower affinities for xanthine oxidase (Km values around 10-4 M). Therefore, aldehyde oxidase activity may be a significant factor in the oxidation of the aromatic aldehydes generated from amines and alkyl benzenes during drug metabolism. Compounds with a 3-methoxy group showed relatively high Vmax values with aldehyde oxidase, whereas the presence of a 3-hydroxy group resulted in minimal Vmax values or no reaction. In addition, amines acted as weak inhibitors, whereas catechols had a more pronounced inhibitory effect on the aldehyde oxidase activity. It is therefore possible that aldehyde oxidase may be critical in the oxidation of the analogous phenylacetaldehydes derived from dopamine and noradrenaline.

John Dewey, as Sidney Hook characterized him, was the philosopher of science and freedom. Dewey, as Larry Hickman has demonstrated, was also a philosopher of technology. And, as most people familiar with Dewey know, he was a philosopher of education and democracy. The complex of technology, science, freedom, education and democracy requires re-examination, not only because of our contemporary cultural political situation but also because of our growing insights into the human condition thanks to the technosciences of life, especially human life. Dewey’s philosophical method of reconstruction, equipped with insights from evolutionary neuroscience and ecological psychology, offers means of reconceiving and thus reevaluating our conception of tools and technology within our cultural context. I begin to take up Mark Tschaepe’s challenge to neuropragmatism to counter what he calls “dopamine democracy” – Plato’s critique of democracy resurrected in neural garb coupled with a critical examination of how social media and other so-called “smart” technologies undermine healthy democratic life. Central to this neuropragmatist approach are cultural affordances – opportunities for action humans have created initially for specific purposes and later retrofitted for other ends-in-view. Dewey’s reconstruction – as method as well as the reconstruction of technology, science, freedom, education and democracy as an entangled complex – is thus imagined as our best strategy for achieving the culture of creative democracy.

Opis:
Progress in elucidating neuroimmune connections has created new opportunities for improving the treatment of Parkinson’s disease (PD). In recent years, mind-body therapies have been shown to have positive effects on the immune and nervous systems, but interactions at the molecular level have not been tested. Thus, the main aim of the present study was to investigate the effects of therapy based on visualization and suggestion on the concentrations of IL-6, CRP, DA, BDNF, CoQ10, and TAC in patients with advanced PD. Eight patients with PD and 8 elderly healthy men (control group) were enrolled in the study. The therapy lasted 19 days and consisted of three parts: individual sessions with the therapist, mixed (therapist and audio-file), and self-training (audio-file). Blood samples were taken before training and at the end of each part. The expected changes in the investigated markers was observed during therapy: the serum concentration of IL-6 and CRP decreased, whereas DA and BDNF increased, however, this change was observed only after the first part of intervention when the therapy was conducted by the therapist. In the subsequent stages, the levels returned to the baseline. Noteworthy, after the therapy, we observed a significant increase in the motor and intellectual skills of the PD patients. No such changes were observed in the control group. Mind-body therapy based on visualization and suggestions aimed at changing the concentrations of signaling molecules, which are crucial in the development or treatment of PD, may be an effective element in supporting pharmacological therapy.

Security of a society is related to the level of its health which largely depends on the level of professional education of medical staff whose task is to help a person in life-threatening situations. Life-threatening situations include among others acute cardiac failure or renal failure. One of the methods to improve health condition of patients with these medical problems is intravenous treatment with dopamine administered by an infusion pump. Medical staff is responsible for proper programming of a pump so that it gives a patient the appropriate dose. Introduction of modern technology to training programme can facilitate the process of learning how to programme an automatic infusion pump. The article describes an example of a simple computer programme created on the basis of free software which is GeoGebra and shared as HTML pages. The prepared learning resource does not require from its receivers any programming skills or special software.

Introduction. Royal Jelly (RJ) is a popular bee-derived product used widely in European and Asian traditional medicine. RJ has some pharmacological activities to support health and longevity as well as prevent ageing. Objectives. To evaluate whether a short-term 6-day Royal Jelly administration is able to induce behavioural and neurochemical effects in aged rats. Materials and method. RJ (previously chemically characterized by GC-FID and GC–MS) was given to 18-month-old male Wistar rats (100 and 500mg of powder/kg b.w./day) in subcutaneous injection for 6 days. Spatial memory was assessed in a water maze. Afterwards, the level of neurotransmitters, their metabolites and turnover in the selected brain regions were estimated by HPLC. Results. Short-term RJ administration did not change spatial memory in aged rats in the water maze, although it was sufficiently active to modify most of all the serotonergic and dopaminergic transmission in the prefrontal cortex and hippocampus. Conclusion. The obtained results indicate that Royal Jelly is able to affect very quickly the neurotransmission in the brain structures responsible for cognitive performance; however, short-term administration is not sufficient to exert behavioural consequences.

Opis:
The significant progress made in recent years, especially in the field of imaging techniques and immunohistochemistry, enables an earlier diagnosis and the treatment of clinically non-functioning pituitary adenomas (NFPAs). In spite of this, these tumours still remain an important medical issue for a large number of specialities. The aim of this article is to provide an overview of the etiopathogenesis, diagnosis and treatment of clinically non-functioning pituitary adenomas with special reference to the development of highly advanced methods in surgery as well as radiotherapy and, especially, in pharmacotherapy.
Istotny postęp, jaki dokonał się w ostatnich latach, zwłaszcza w zakresie rozwoju diagnostyki obrazowej i badań immunohistochemicznych, umożliwia wcześniejsze rozpoznawanie, a następnie leczenie nieczynnych hormonalnie gruczolaków przysadki (NFPAs). Mimo to guzy te dalej pozostają istotnym problemem klinicznym dla lekarzy wielu specjalności. Celem pracy jest szczegółowe przedstawienie poglądów na temat etiopatogenezy, obrazu klinicznego, diagnostyki i metod leczenia klinicznie nieczynnych hormonalnie gruczolaków przysadki ze szczególnym uwzględnieniem postępów w technikach chirurgicznych oraz rozszerzających się możliwości radioterapii, a szczególnie farmakoterapii.
