Opis:
Ceratopteris thalictroides is a water fern very sensitive to both dehydration and low temperature. This study focuses on the cryopreservation of this species by encapsulation-dehydration technique, in particular on the effects of pre-culture step, alginate bead size and the physical conditions of culture on the cryopreservation efficiency. Encapsulated and non-pre-cultured gametophytes did not survive cooling with liquid nitrogen. When cryopreservation was preceded by a 2-week period of pre-culture, regrowth reached 42.1%. Reduction in the size of the alginate bead, and culture in total darkness resulted in improved gametophyte regrowth capacity (75.5% or 81.7%, respectively). The best results (91.3%) were obtained when all factors tested occurred simultaneously. The gametophytes recovered very quickly and sporophytes were formed within 4 weeks after rewarming. These simple improvements can be used, not only for the cryopreservation of gametophytes in cryptogams but also for some recalcitrant species of seed plants.

Opis:
Cryopreservation in liquid nitrogen (–196℃, LN) is the method of long−term conservation of plant tissues, which has been evolving for many of seed of woody plant species. During cryopreservation protocol to LN in plant cells take place the supercooling tissue water or vitrification, which allows the successful storage. Water content was a significant determining factor with survival of cryostored embryonic axes of woody plants. The protocols of cryopreservation include some steps during preparation of plant material, which could be a source of oxidative stress, phase transition of membranes and destruction in cells.

Opis:
Shoot tips from in vitro plants of four rose species were cryopreserved by the droplet vitrification method. Optimized conditions involved exposure to loading solution for 20 min, then treatment with plant vitrification solution (PVS2) for 20 min (Rosa agrestis, R. canina and R. dumalis) or 30 min (R. rubiginosa) followed by freezing in liquid nitrogen. Survival rate ranged from 78.3 to 95.1%, depending on the species. Regrowth rate of shoot tips was 50.5% for R. agrestis, 63.2% for R. rubiginosa, 71.4% for R. dumalis and 78% for R. canina. The preculture of donor plants in a medium with 0.25 цЫ sucrose facilitated the isolation of shoot tips and increased regrowth rate after cryopreservation. Plant regeneration was carried out in Murashige and Skoog medium with 1 цЫ 6-benzylaminopurine, 1.5 цЫ gibberellic acid and 0.087 M sucrose. Plants regenerated from cryopreserved shoot tips did not display morphological alterations in comparison with non-cryopreserved shoot tip - derived plants.

Opis:
The purpose of the research is to prepare a mathematical and numerical model for the phenomenon of heat transfer during cryopreservation. In the paper, two popular methods, slow freezing and vitrification, are compared. Furthermore, the basic model of thermal processes is supplemented by the phenomenon of phase transitions. To determine the temperature distribution during cryopreservation processes, one uses the heat transfer equation proposed by Pennes. An integral part of the energy equation is the substitute thermal capacity (STC) performed according to the concept named one domain method (fixed domain method), The numerical model is developed using the finite difference method (FDM) connected with directed interval arithmetic. The final part of the article contains the results of numerical simulations.

The Oomycete pathogen is frequently sampled, isolated to pure cultures, stored, and char-acterized. Cryopreservation (storage in liquid nitrogen, –196°C) is a technique that ensures safe, long−term conservation of plant species with recalcitrant seeds, vegetatively propagated species and biotechnology products such as somatic embryos, cell lines and genetically transformed material. The aim of cryostorage is to keep plant tissue in a safe way, which does not cause changes during storage at neither phenotypic, cytological and histological, biochemical, genetic nor epigenetic level. Therefore, before cryostorage will be applied for long−term preservation it should be thoroughly assess if it does not cause any injuries in plant material.

The aim of this study was to evaluate the influence of sucrose concentration in the preculture medium, as well as the duration of osmotic dehydration on the efficiency of chrysanthemum ‘Richmond’, ‘Lady Orange’ and ‘Lady Salmon’ cryopreservation by encapsulation-dehydration technique. For all cultivars tested, the best regrowth of cryopreserved shot tips was recorded with 0.25 M sucrose concentration and 10 µM ABA during a two-week preculture, followed by a 4- or 7-day osmotic dehydration. The survival rate ranged from 56.8–58.0% (the Lady group) to 63.6% (‘Richmond’). However, the ability to grow was smaller and reached 18.2–50.7%. It was found that higher sucrose concentration during the preculture slowed the growth of chrysanthemum shoot tips and led to an increased formation of multiple shoots (by activating axillary buds) or deformed adventitious shoots (incapable of further growth). The frequency of tissue hyperhydricity also increased, while the rhizogenesis efficiency decreased when higher sucrose concentration in the preculture medium was applied. The influence of osmotic dehydration duration on the explants morphogenetic response was cultivar-dependent.

Opis:
The discovery of conifer somatic embryogenesis (SE) and the subsequent development of SE protocols for a range of genera and species have opened new research opportunities to forest biotechnologists and a means towards mass clonal propagation for the forest industry. This paper provides a general description of the conifer SE process, followed by a review of protocols developed specifically for several conifer species in the Pinaceae family for which production of somatic trees has been demonstrated. Additionally, future research needs, including approaches for developing markers to optimize SE process and the production of high quality embryos, are discussed.

Opis:
The aim of the study was to assess the susceptibility of small-leaved lime (Tilia cordata Mill.) seeds to drying and freezing in liquid nitrogen (-196°C). Seed samples were frozen in liquid nitrogen for 24 h at 11 different levels of seed moisture content (m.c.), ranging from 3.1% to 22.8% (fresh weight basis). All samples, including unfrozen control samples, were subjected to scarification with concentrated sulphuric acid (Tylkowski 1998) either before or after freezing. Seed pre-treatment before germination (at 3~15°C/16~8h) involved cold stratification at 3°C without substrate. Seed drying to 3.1% m.c. significantly reduced their germinability (to 63%), as compared to the high germinability (82-88%) of seeds with 5.2-20.9% m.c. Thus seeds of this species can be assigned to the ‘suborthodox' category. Such a high germinability (79-87%) was preserved after freezing in liquid nitrogen in samples dried to 9.0-17.4% m.c. if scarification was performed before freezing, and in samples dried to 9.1-16.2% m.c. if scarification was performed after freezing. The highest percentage of seedlings emerged after freezing in liquid nitrogen from seeds dried to 11.1-20.1% m.c. (emergence 65-75%) if scarification was performed before freezing, and from seeds dried to 7.3-17.8% m.c. (emergence 53%-71%) if scarification was performed after freezing.
