    We aimed to quantify the gene expression changes of the potent orexigenic melanin-concentrating hormone (MCH) in chicken (Gallus gallus) hypothalamus with quantitative real-time polymerase chain reaction (qPCR), and for the first time determine peptide concentrations with a novel radioimmunoassay (RIA) under different feeding status. Three different experimental conditions, namely ad libitum feeding; fasting for 24 h; fasting for 24 h and then refeeding for 2 h, were applied to study changes of the aforementioned target and its receptor (MCHR4) gene expression under different nutritional status. The relative changes of MCH and MCHR4 were also studied from 7 to 35 days of age. Expression of PMCH and MCHR4 along the gastrointestinal tract (GIT) was also investigated. We found that expression of both targets was significant in the hypothalamus, while only weak expression was detected along the GIT. Different nutritional states did not affect the PMCH and MCHR4 mRNA levels. However, fasting for 24 h had significantly increased the MCH-like immunoreactivity by 25.65%. Fasting for 24 h and then refeeding for 2 h had further significantly increased the MCH peptide concentration by 32.51%, as compared to the ad libitum state. A decreasing trend with age was observable for both, the PMCH and MCHR4 mRNA levels, and also for the MCH-like immunoreactivity. Correlation analysis did not result in a significant correlation between MCH peptide concentration and abdominal fat mass in ad libitum fed birds. In conclusion, MCH peptide concentration altered in response to 24 h fasting, which indicated that this peptide may take part in feed intake regulation of broiler chickens.

    Rosnąca skala i częstotliwość występowania zagrożeń, jakie mogą dotknąć gospodarkę i społeczeństwo na poziomie krajowym wymaga zmiany podejścia do zarządzania ryzykiem regulacyjnym. W artykule postuluje się dokonanie przełomu w mechanizmie zarządzania ryzykiem regulacyjnym poprzez sformułowanie na nowo celu i kryteriów optymalizacji działań regulatorów. Referat składa się z czterech logicznie powiązanych części. Na początku uzasadnia się rolę i istotę zarządzania losowym ryzykiem regulacyjnym, następnie dokonuje się przeglądu przesłanek przemawiających za zarządzaniem ryzykiem regulacyjnym, która jest koncepcją od niedawna lansowaną m.in. przez instytucję OECD. Następnie określa się główne obszary tematyczne wchodzące w zakres zarządzania losowym ryzykiem regulacyjnym. W ostatniej i najważniejszej części referatu podejmuje się próbę uzasadnienia przeformułowania celu i kryteriów zarządzania ryzykiem regulacyjnym.
    Regulatory Impact Assessment (called shortly in Polish as OSR) is an instrument improving the law. It have been developing very quickly in Poland since our country became the member of OECD (November 1996) and of the European Union (May 2004). Poland build special institutional infrastructure to evaluate new regulations. This year about 1000 of administrative people will be trained in applying the rules of OSR. It is observe that part of regulations in force includes information about the results of the OSR. The purpose of this paper is to change practice in the methodological issue aiming to choose the best law alternative for the society. The author recommends two main solutions. First proposal is to implement in evaluation of law projects risk issues. The second is to change aim of the regulation risk. Recently, in practice conservative approach to the risk regulation dominates. This approach leads to minimizing not only the value of losses but also costs of reducing or avoiding risk. In the article it is stated, that better solution is to add also direct and indirect benefits of risk reduction activities. As a result, the new aim is to minimize differences between losses and all identified benefits in long horizon.

    Aplikacje RIA zyskują popularność w szerokim świecie Internetu. Ich główną zaletą jest bardzo duży stopień przystępności dla użytkownika, przede wszystkim ze względu na eliminację konieczności "przeładunku" strony po dokonaniu pewnych jej modyfikacji. RIA jest idealną technologią do współpracy z środowiskiem CMS. System CMS umożliwia zarządzanie stronami w bardzo łatwy sposób, nawet dla osób z relatywnie małą znajomością języka HTML. Technologia RIA jest wymarzonym rozwiązaniem dla właścicieli małych firm, które nie mogą sobie pozwolić na zatrudnienie dodatkowej osoby tylko do zarządzania stroną internetową. Skomplikowane systemy, takie jak CMS, stają się nawet prostsze niż pisanie strony w zwykłym - klasycznym kodzie HTML. Do głównych celów realizowanych w powyższym systemie można zaliczyć uproszczenie wszystkich skomplikowanych operacji oraz zapewnienie przyjaznego dla użytkownika interfejsu, który "poprowadzi" za rękę użytkowników. System CMS musi być bardzo elastyczny, aby dostosować się do użytkowników. Innym bardzo ważnym aspektem jest bieżące utrzymanie systemu, gdyż każdy błąd zagrażający bezpieczeństwu danych musi zostać wyeliminowany. Dla środowiska rozwijającego oprogramowanie ważnym wątkiem jest również rejestracja wszystkich błędów, tak aby w przyszłości móc go odtworzyć w zależności od kierunku rozwoju oprogramowania.
    RIA applications are gaining popularity over the internet. They are very user friendly because we are eliminating page reloading after making actions. It's ideal technology for making backend for CMS. A CMS system makes managing of website very easy for people with zero HTML skills. It's perfect for owners of small business which can't afford for hire another person, just for managing website. But such complex systems as CMS are very complicated, even if it's simpler than writing website in plain HTML it still may be too hard to understand for people. And it is developer's work to simplify all complex operation, provide user friendly interface which will take users hand and bring him safely to the other side. CMS systems must be also very flexible to adjust themselves to users. We must also remember about keeping system up to date, because every bug which is threaten for data safety must be eliminated. For developer it's also important to log all errors, so in future he or she can recreate the error on develop environment.
