Investigations into the abundance of adult D. reticulatus in relation to the effect of climatic conditions (temperature, humidity) on host-seeking behaviour were conducted during the autumn (September-October) and spring (May) activity peaks in 2008–2009 in eastern Poland (51°25’N). The study was conducted in the same habitat where similar examinations were performed in 1999–2000. A comparative analysis revealed that the abundance of D. reticulatus had almost doubled within 10 years. Analysis of data on the humidity and temperature conditions prevailing during tick collection, and on tick abundance in the respective study periods in 1999–2000 and 2008–2009, as well as absence of climatic changes over many years, indicate that the increase in the numbers of ticks may have been a result of a multitude of other factors, e.g. weather or environmental conditions favourable for ticks and their hosts. The substantial differences in D. reticulatus abundance observed during the autumn activity peak (an increase from 126 and 128.6 specimens per collection in 1999 and 2000, respectively, to 247.3 in 2008) demonstrate the considerable effect of the biotic and abiotic conditions prevailing during the development of young and adult stages on the abundance of this tick species. The activity of adult D. reticulatus ticks in the autumn was 2.7-fold higher in comparison to that observed during the spring collection; the difference was statistically significant (p<0.0001). Females dominated in both seasonal activity peaks. The ratio between females and males during the spring and autumn peaks was 3.31:1 and 1.05:1, respectively. The increase in the abundance of the D. reticulatus population implies a higher risk of transmission of tick-borne diseases in the study area, and the necessity to develop and implement effective prevention methods and tick control programmes.

Dermacentor reticulatus is widely distributed dangerous tick that usually lives in the river valleys, boggy forests, meadows, and wooded pastures. Tick populations from various regions may exhibit morphological differences. In our study we compared morphometric features of tarsus in larvae D. reticulatus from Polish and Slovakian populations. I tarsus width, III tarsus length, and length of dorsal setae of I tarsus were significantly higher in Polish populations. Indices of width to length of tarsus I and tarsus III were also significantly different in both populations. The other examined morphologic features were similar, what may result from the same environmental conditions of both populations.

Opis:
A total of 684 Ixodes ricinus ticks (321 nymphs, 184 males, and 179 females) were collected by fl agging lower vegetation in 6 forest districts located on the territory of Lublin province (eastern Poland). Ticks were examined by polymerase chain reaction (PCR) method for the presence of Anaplasma phagocytophilum DNA with two pairs of primers: EHR521/EHR747 for detecting 16S rRNA gene, and LA6/LA1 for detecting ankA gene. To study the relationship between infection in ticks and people occupationally exposed to tick bite, blood serum samples of 261 forestry workers employed in the same forest districts were examined by immunofl uorescence method for the presence of specifi c antibodies against A. phagocytophilum. A total of 70 ticks out of 684 examined (10.2%) showed the presence of A. phagocytophilum 16S rRNA gene. The prevalence of infection was signifi cantly dependent on tick’s stage (χ2=49.2, p<0.00001) and geographical locality (χ2=34.4, p<0.00001). The percentage of I. ricinus females infected with A. phagocytophilum (24.6%) was signifi cantly greater compared to males (6.5%) and nymphs (4.4%) (p<0.00001). Only 19 ticks out of 684 examined (2.8%) showed the presence of A. phagocytophilum ankA gene, signifi cantly less compared to 16S rRNA gene (p<0.00001). The prevalence of infection demonstrated by the presence of ankA gene was also signifi cantly dependent on tick’s stage (χ2=23.6, p<0.00001) but not on locality (χ2=9.8, p=0.082). A signifi cant correlation was found between the presence of A. phagocytophilum 16S rRNA gene in I. ricinus female ticks from the particular forest districts and the serologic response to A. phagocytophilum of forestry workers employed in the same districts (p<0.05). No signifi cant correlation was found between the presence of A. phagocytophilum ankA gene in I. ricinus ticks and serologic response of exposed workers. In conclusion, detection of A. phagocytophilum infection in ticks by PCR with the use of EHR521/EHR747 primers detecting 16S rRNA gene is signifi cantly more sensitive compared to LA6/LA1 primers detecting ankA gene.
