Background. Candida is a strain of fungi that is found on the surface of healthy mucous membranes of the oral cavity. Yet, Candida may also be considered opportunistic microorganisms. The pathogenicity of Candida spp. depends on strain-specific characteristics including, invasiveness, adhesion, and production of biofilm abilities as well as the production of enzymes allowing the colonization of tissue. The aim of the paper was to evaluate the frequency of Candida spp. occurrence in the oral microbiota of healthy adults, and its relation with risk factors and colonization. Material and methods. This study consisted of 72 healthy participants (from the Lubelskie Voivodeship between 19 and 73 years of age). Medical history was collected to determine potential relationships to risk factors affecting oral microbiota (past oral cavity diseases, chronic diseases, and smoking). The participants who were qualified had no oral cavity infections during the study. The collected clinical samples (oral cavity swabs) were disseminated on the chromogenic base for isolation and differentiation of Candida spp. Results. The frequency of Candida spp. occurrence was at the level of 63.8%. C. albicans was the most frequently identified species (56.5%). C. glabrata (45.6%) and C. tropicalis (23.9%) were identified less often. In 21.7% of swabs, two species of Candida were identified and in 2.2% of swabs three species identified. The most common colonization risk factor was associated with smoking (32.6%). Conclusions. Yeasts of Candida species are part of the healthy microflora of the oral cavity in people of different ages and may occur as single species or coexist with other species. The coexistence of chronic diseases, propensity for oral cavity infections and smoking significantly influence the Candida colonization. This may result in future clinical consequences, for example in cases of immunodepression.
Wprowadzenie. Grzyby z rodzaju Candida mogą w warunkach fizjologicznych występować na powierzchni błony śluzowej jamy ustnej, jednak równocześnie uznawane są za drobnoustroje oportunistyczne. Chorobotwórczość Candida spp. zależy od właściwości szczepu, inwazyjności, zdolności do adhezji i tworzenia biofilmu oraz wytwarzania enzymów umożliwiających kolonizację tkanek. Celem pracy była ocena częstości występowania Candida spp. w mikrobiocie jamy ustnej u osób dorosłych w warunkach zdrowia oraz określenie związku pomiędzy czynnikami ryzyka a kolonizacją. Materiał i metody. Grupę badaną stanowiły 72 zdrowe osoby (mieszkańcy województwa lubelskiego, w wieku od 18 do 73 lat), od których zebrano wywiad odnośnie obecności czynników wpływających na stan mikrobioty jamy ustnej (występowania w przeszłości chorób jamy ustnej, istnienia chorób przewlekłych oraz palenia papierosów). Do udziału w badaniach zakwalifikowano osoby, które w trakcie ich prowadzenia zadeklarowały brak infekcji w obrębie jamy ustnej. Pobrany materiał kliniczny (wymazy z jamy ustnej) posiano na podłoże chromogenne do izolacji i różnicowania Candida spp. Wyniki. Częstość występowania Candida s pp. odnotowano na poziomie 63,8%. Najczęściej i dentyfikowano C. albicans (56,5%), rzadziej C. glabrata (45,6%) i C. tropicalis ( 23,9%). W 21,7% w ymazów rozpoznano po dwa gatunki Candida, a w 2,2% wymazów trzy. Najczęstszym deklarowanym czynnikiem ryzyka kolonizacji było palenie tytoniu (32,6%). Wnioski. D rożdżaki z rodzaju Candida są obecne w mikroflorze jamy ustnej w warunkach zdrowia u osób w różnym wieku i mogą występować jako pojedyncze gatunki lub współistnieć obok siebie w różnych konfiguracjach. Współistnienie chorób przewlekłych, skłonności do infekcji w obrębie jamy ustnej oraz palenie papierosów wpływają istotnie na kolonizację jamy ustnej drożdżakami, co może w przyszłości skutkować konsekwencjami klinicznymi w przypadku spadku odporności.

Opis:
The purpose of this study was to determine the in vitro susceptibility to 6 antifungal agents of 143 strains of Candida species isolated from 545 patients with suspicion of fungal infection. Test ATB Fungus (bioMerieux) was used. Among all Candida isolates 91.6% were susceptible to tested polyenes and 32.9% to all azoles. Less susceptible to polyenes isolates were noted in low percentage (0.7 to 4.2%) and to azoles more frequcntly (32.9 to 41.2%). Candida isolates resistant to antifungal agents tested were dependent from the kind of clinical samples and species of fungi. Monitoring of the susceptibility to antifungal agents by ATB commercial test seems to be uscful tool for therapeutic purposes.

In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Opis:
Early detection and correct diagnosis of fungal infection is important for further therapy and prophylaxis. Currently, there are a number of proposals related to rapid diagnostic tests. To those we can include chromogenic media, such as CHROMagar® Candida (GRASO). This medium has been developed to differentiate four species of genus Candida. Its use in laboratory has opened up possibilities for growing not only the genus Candida, but also other fungi. The study suggests that medium CHROMagar® Candida can provide an excellent pre-test, or be used as complementary to biochemical tests. Establishing a full template of colours would enable more efficient and fuller use of the medium properties, especially in the discrimination of the teleomorphic form from anamorphic.

We have evalueated the activity of acidous asparagine protease in 48 strains of Candida isolated from homogenates of palatine tonsils from 116 patients after tonsilectomy. Strains most frequently isolated were C. albicans (66.7±6.8 %) and C. tropicalis (14.6±5.09 %). The range of proteolysis zone for Candida strains was 4.6 to 12.5 mm. Over 60% of the studied strains showed high and very high proteolytic activity (>6.6 mm) and the remaining strains with showed moderate activity (4.6-6.5). There were no strains with low medium activity (to 4.5 mm zone). The range of the number of rosettes E formed by lymphocytes obtained from the studied tonsils was 0-268.3. Lymphocytes isolated from 70 % of tonsils infected with Candida formed only 0-130 rosettes E; in the remaining cases the number ranged between 131 and 220. The number of lymphocytes T forming rosettes E isolated from palatine tonsils from patients not infected with fungi was 2.5 times higher. There was no correlation between the size of proteolysis zone and the number of rosettes E.

The incidence of Candida spp. infections has increased in recent years due to the widespread use of broad-spectrum antibiotics and the growing numbers of immunocompromised individuals. Despite the predominance of Candida albicans, non-albicans Candida species, such as Candida glabrata, Candida tropicalis, Candida guilliermondii, Candida dubliniensis, Candida parapsilosis, and Candida krusei are emerging as both, colonizers and pathogens, that can cause superficial and systemic infections. Currently, there are various methods for identifying yeasts from clinical samples. Conventional methods for identification of Candida species are based on morphological and physiological attributes. Although phenotypic identification methods are cost effective and practical procedures for routine discrimination of isolates in the clinical microbiology laboratory, the accurate identification of all isolates. Moreover, given the variability in the phenotypic characteristics and other irregularities in the taxonomy of the genus Candida, identification based on traditional methods has become less reliable. Important progress has been made with various studies on the phenotypic characteristics of these species using commercially available identification systems. The aim of this review is to summarize the methods available for identification of Candida species. The first part of the article presents the characteristics of phenotypic methods and quick identification techniques, including systems based on the use of chromogenic media.

The objective of the study was the analysis of occurrence and number of Candida in the alimentary tract of 209 patients with intestinal amoebiasis considering their state after the treatment. Amoebiasis was diagnosed on the grounds of Entamoeba histolytica present in the faeces, what has just recently been recognised equal to the presence of E. histolytica complex. In all patients serological investigations were performed using indirect haemagglutination (IHA) test. The cases with relapses or with the intestinal ailments after elimination of the parasite from the body at the simultaneous isolation of Candida in the faeces were focused on. In the compass of several years a wide number of qualitative and quantitative mycological tests were performed comprising 39 patients with relapses after symptomatic amoebiasis and 42 with ailments which had appeared after the treatment form asymptomatic amoebiasis. Observation of the dynamics of number of Candida cells revealed most often their increment in subsequent examinations. It occurred in 69% people examined after the treatment of asymptomatic antoebiasis and in 40,9% after the treatment of symptomatic amoebiasis. The differences obtained were statistically significant. The number of yeast remained at the same high level in 23,8% patients after asymptomatic amoebiasis and in 36% after symptomatic amoebiasis. Drop, of Candida cells number was noted in 7,1% people after asymptomatic amoebiasis and in 11,4% after symptomatic amoebiasis. In the light of the results obtained it turns possible that suffering from amoebiasis could even in the long run affect Candida tendency to proliferate or the tendency to remain at the same high level. It may be presumed that the growing number of yeast was an important reason for the intestinal ailments after the elimination of E. histolytica. The results of mycoserological investigations also were taken into consideration.

Candida is a yeast species recognized as the most frequent etiological agent of systemic and invasive thrush in humans. Invasions can affect all tissues, organs and systems of human in various stages of development. In the last 10 years Candida infections have increased 15 times. The purpose of our study was to determine the sensitivity of four antibiotics belonging to three different groups of antifungal agents against clinical and food-borne Candida strains. Our studies showed that of all tested strains, 7% was resistant to nystatin, 32% to fluconazole, 23% to voriconazole, and no strains grew in the presence of caspofungin. Despite the differences in biochemical profiles of clinical and food-borne isolates of Candida, a group of strains showing resistance to antibiotics include both types of isolates. At the same time circulating of antibiotic-resistant strains outside the hospital environment and the yeast infection via food is possible.
