Opis:
Microbial surfactants or biosurfactants are surface active amphiphilic macromolecules that are produced by a number of microorganisms (bacteria, yeast and fungi). These compounds have extensive application in various industries especially in food, pharmaceutical and oil industry. The aim of this paper is to optimize the culture conditions for the biosurfactant production from olive oil by a novel halophilic isolate microorganism. The Taguchi experimental design methodology based analysis of olive oil as carbon source, yeast extract as nitrogen source and KH2PO4 as phosphorus source revealed that the olive oil and yeast extract signifi cantly affect biosurfactant production in high salt conditions. Maximum biosurfactant (E24= 40%) produced in the presence of 4% (v/v) olive oil, 0.2% (w/v) yeast extract, and 0.002% (w/v) KH2PO4 . In conclusion, halophilic archaeon Haloarcula sp. IRU1 could be a potential microorganism for the production of biosurfactant from olive oil as carbon source in high salt conditions. The optimal parameters obtained during the optimization process were: olive oil 4%, yeast extract 0.4% and KH2PO4 0.004%.

Opis:
The separation and purification of rebaudioside A from Stevia rebaudiana crude extracts (Steviosides) by macroporous resin were optimized by Taguchi orthogonal array (OA) experimental design methodology. This approach was applied to evaluate the influence of five factors (adsorption temperature, desorption time, elution solution ratio, adsorption volume and type of resin) on the rebaudioside A yield. The percentage contribution of each factor was also determined. The results showed that elution solution ratio and adsorption volume made the greatest (59.6%) and the lowest (1.3%) contribution, respectively. The results showed that the Taguchi method is able to model the purification of rebaudioside A process well (R2 > 0.998) and can therefore be applied in future studies conducted in various fields. Adsorption temperature 35°C, desorption time 60min, elution solution ratio 3, adsorption volume 200ml and HPD-400 as resin were the best conditions determined by the Taguchi method.

Opis:
The photooxidative destruction of C. I. Basic Red 46 (BR46) by UV/S2O82- process is presented. Central Composite Design (CCD) was employed to optimize the effects of operational parameters on the photooxidative destruction efficiency. The variables investigated were the initial dye and S2O82- concentrations, reaction time and distance of the solution from UV lamp. The predicted values of the photodestruction efficiency were found to be in good agreement with the experimental values (R2 = 0.9810, Adjusted R2 = 0.9643). The results of the optimization predicted by the model showed that the maximum decolorization efficiency (>98%) was achieved at the optimum conditions of the reaction time 10 min, initial dye concentration 10 mg/l, initial peroxydisulfate concentration 1.5 mmol/l and distance of UV lamp from the solution 6 cm. The figure-of-merit electrical energy per order (EEo) was employed to estimate the electrical energy consumption and related treatment costs.
