The objective of the study was to determine the effect of increasing manganese concentrations, 2, 4, 8 and 16 times higher than the standard content in Murashige and Skoog (1962) medium, on morphological traits and on the accumulation of iron and magnesium in the roots and shoots of Dendrobium kingianum Bidwill. For micropropagation of orchid plants we used shoots placed on solid MS medium supplemented with 1.0 mg dm-3 kinetin and 0.5 mg dm-3 NAA, as well as different concentrations of manganese (as MnSO4· 4H2O): 22.3 (control), 44.6, 89.2, 178.4 and 356.8 mg dm-3. After 18 months of in vitro culture, the increased concentrations of manganese, from 44.6 to 356.8 mg dm-3 in the MS medium, had no stimulatory effect on orchid development in the in vitro culture. The greatest number and length of shoots and roots and the greatest fresh weight of plantlets were noted in the control. At the high manganese concentrations of 178.4 and 356.8 mg dm-3 in the MS medium, the number and length of these organs and the fresh weight of entire plantlets were statistically significantly lower than in the control. Spectrophotometric analysis (ASA) showed that as the manganese concentrations in the medium increased, there was an increase in the manganese content and a decrease in the iron and magnesium content in the orchid shoots and roots. Moreover, the iron accumulation in roots was 2-3 times greater than in shoots, while the magnesium accumulation was only 15-30% greater. In contrast to these metals, the accumulation of manganese was 2-3 times greater in the orchid shoots than in the roots.

The study focused on the influence of an increased copper content in the Murashige and Skoog (1962) solid medium on the in vitro plant growth and development of Dendrobium kingianum Bidwill. Sterile explants of pseudobulbs were used for micropropagation of orchid plants on the MS regeneration medium supplemented with 0.5 mg dm-3 NAA and 1.0 mg dm-3 kinetin. Copper (as CuSO4 × 5H2O) was added to all the combinations in concentrations of 0.025 (control), 0.625, 1.25, 2.5 and 5.0 mg dm-3. The results showed that the treatments with 1.25 and 2.5 mg dm-3 stimulated the orchid growth and development in in vitro culture. After eight months of growing in in vitro culture, the highest number of shoots, the longest roots and the heaviest fresh weight of plantlets were obtained in these treatments. In medium with the highest copper concentration (5.0 mg dm-3), a negative influence of the metal on the length of roots and fresh weight of orchids was noted. Spectrophotometric analysis (ASA) showed that the copper and iron accumulation increased in both shoots and roots with the increase in the external Cu level, whereas the zinc and calcium accumulation in these organs decreased. The copper and zinc accumulation in the roots was about 1.5-2.5 times higher than in the shoots, but the iron accumulation was about 3-3.5 times higher. The calcium accumulation in roots was only 5-12% higher than in shoots.

Iodine deficiencies in the human diet have created the need to search for new sources of food enriched with this element. The aim of biofortification of plants with iodine is to increase the iodine content in plants while ensuring that food products are safe for health. Seedlings enriched with iodine may be an alternative source of this element. Iodine is not an essential nutrient for plants, and its role and effects on plants are not fully known, particularly in the early stages of development. The aim of the study was to determine the effect of applying KI on the iodine biofortification of lettuce seedlings of the variety Michalina. The seedlings were grown on Petri dishes lined with filter paper. The doses of KI used for biofortification were 0 (control), 0.5, 1, 2.5 and 5 μM dm-3. Iodine concentration was determined in the seedlings by ICP-MS, while K, Na, Mg, Ca, Fe, Zn, Mn and Cu were assayed by atomic absorption spectrometry. In respect of the biological quality of the iodine-enriched seedlings, we determined their length, biomass yield and chlorophyll content by spectrophotometry following extraction with acetone. A significant increase in the iodine content in seedlings was obtained after each dose of KI and we can claim that even the lowest dose, 0.5 μM dm-3 KI, is sufficient for biofortification. In comparison with the control, KI reduced the length of seedlings but did not affect their biomass or the chlorophyll content. Seedlings growing in the presence of 1, 2.5 and 5 μM dm-3 KI contained more potassium and less sodium and manganese than the control seedlings. The magnesium concentration was lower than in the control seedlings following the application of 2.5 or 5 μM dm-3 KI. No significant differences in the content of calcium, zinc, iron or copper were noted in the biofortified seedlings.
