Opis:
During 2015, samples from 22 apple trees showing proliferation symptoms were collected in southwest Bulgaria and Central and South Poland and tested for phytoplasma presence. ‘Candidatus Phytoplasma mali’ was identified in 18 samples based on results of restriction fragment length polymorphism (RFLP) analysis of the 16S rRNA gene amplified in nested PCR using primer pair P1/P7 followed by R16F2n/R16R2 and F1/B6 primer pairs. The nitroreductase and rhodonase-like genes and ribosomal protein genes rpl22 and rps3 were then analyzed using PCR-RFLP technique to study the genetic variability of the phytoplasma strains. Two restriction profiles, P-I or P-II, were obtained from fragments of 16S rDNA plus 16S-23S spacer region digested with HpaII enzyme. Restriction fragment length polymorphism analysis of nitroreductase and rhodonase-like genes using digestion with HincII endonuclease revealed that all ‘Ca. P. mali’ strains belonged to the subtype AP-15. Analysis of rpl22 and rps3 ribosomal protein genes digested with AluI enzyme resulted in classification of detected phytoplasma strains to rpX-A subgroup.

Opis:
The effect on cell viability and content of PhC of three grapes varieties – Moravian Muscat, Blue Burgundy and Lemberger is presented. The effect of polyphenols from wine and grapes was studied for many times, but the effect of pomace, the by-product of wine production, was neglected. Thus study is devoted to compare the effect of berries and pomace on cell viability in context of their utilization as source of bioactive compounds. Effect on viability of human keratinocytes (HaCaT) was investigated in vitro using following concentrations of PhC in cultivation medium: 25, 50, 75 and 100 µg·ml–1. The results show that the content of PhC in berries and pomace was similar and the cell viability decreased with increasing concentrations of PhC, in most cases. The impact on cell viability also depends on individual variety of grapes.

Opis:
A protocol was developed for high frequency and low cost of in vitro shoot proliferation and ex vitro rooting of Eustoma grandiflorum (Gentianaceae) on solid medium. Shoot tips as explants were cultured on Murashige and Skoog (MS) medium enriched with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) (0.00, 0.01, 0.10 and 1.00 mg l–1) and 6-benzylaminopurine (BAP) (0.00, 0.50, 2.00 and 5.00 mg l–1). Three culture media systems (solid, liquid and double-phase) were applied. None of the explants cultured on liquid and double-phase media resulted in live plant production. Maximum axillary shoot number (54.45) was recorded in the plantlets treated with 0.10 mg l–1 2,4-D in combination with 5.00 mg l–1 BAP. Treatment of 0.01 mg l–1 2,4-D along with 0.50 mg l–1 BAP produced maximum node number and internode length. Some shoots produced on medium containing plant growth regulators (PGRs) were rooted in soil. The largest number (5.50/plantlet) and longest length of root (7.75 cm/plantlet) were obtained in ex vitro condition on the base of shoots produced in culture medium enriched with 0.10 mg l–1 2,4-D along with 0.50 mg l–1 BAP. The combination of 1.00 mg l–1 2,4-D and 0.50 mg l–1 BAP was found to be the most suitable PGRs for obtaining the highest callus weight. The most fresh weight was calculated from plantlets grown on the medium containing 0.10 mg l–1 2,4-D along with 5.00 mg l–1 BAP. Maximum dry weight was obtained in free-PGRs medium. About 90% of the rooted plantlets were established successfully in cultivation beds. Acclimatized plants were morphologically similar to the mother plants.
