In this study, the effects of salicylic acid to antioxidative activity and photosynthetic characteristics in waterlogging stress of two peony cultivars (‘Fengdanbai’ and ‘Mingxing’) were investigated. 4-year-old peony grown in different levels of waterlogging stress and then different concentration prepared SA (0.0, 0.1, 0.5 and 1.0 mmol L⁻¹) sprayed on fresh leaves of peony. The antioxidative enzymes activities include superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), chlorophyll content, relative conductivity and MDA content were measured in leaves about different waterlogging treatment, the photosynthetic characteristics were also measured using photosynthetic measurement system. The results showed that waterlogging stress decreased the chlorophyll content in all peony cultivars leaves, but with SA treatment can inhibit the decrease of chlorophyll content. Relative conductivity increased as the extension of waterlogging time in two cultivars. SA treatment could effectively inhibit the increase of relative conductivity, and 0.5 mmol L⁻¹ of SA was the most suitable concentration. SOD, POD, CAT activity increased first and then decreased in different waterlogging condition, SA significantly increased the activity of various enzymes. MDA content was increase as the expansion of waterlogging time in two cultivars. SA inhibits the increase of MDA content. Of all concentration of SA, 0.5 mmol L⁻¹ was the best concentration to inhibit the waterlogging stress. For the photosynthetic characteristics, the net assimilation rate (Pn), stomatal conductance (Gs), transpiration rate (Tr) and intercellular CO₂ (Ci) were decreased under different waterlogging condition. SA treatment can increase Pn, Gs, Tr and Ci of peony.

The enzyme strictosidine synthase (STR, EC: 4.3.3.2) plays a key role in the biosynthetic pathway of terpenoid indole alkaloid (TIA). It catalyzes the condensation of the tryptamine and secologanin to form 3α(S)-strictosidine, which is the common precursor of all TIAs. In this paper, a STR gene designated as DoSTR (GenBank: KX068707) was first cloned and characterized from Dendrobium officinale with rapid amplified cDNA ends method (RACE). DoSTR has a length of 1380bp with 1179bp open reading frame encoding 392 amino acids. BlastP analyses showed that its amino acid sequence was classified into Str_synth superfamily. qRT-PCR showed that DoSTR was expressed in all tissues tested, with a significantly higher level in flower and the lowest in stem. Four different treatments with MeJA, SA, ABA and AgNO₃, respectively, could induce the DoSTR expression to a different extent. And the effect of MeJA was the most obvious and transcript level of DoSTR induced by MeJA was 20.7 times greater than that of control at 48 hours after treatment. Furthermore, it was found that DoSTR was localized in vacuole through transient expression in tobacco. The characterization and expression of DoSTR can help in further studying the role of DoSTR in the biosynthesis of TIAs in D. officinale. This study may throw light on the alkaloid biosynthesis pathway of D. officinale.

Paeonia sect. Moutan is a wide known ornamental plant in the world. The objective of this study was to provide the theoretical basis for scientific preservation and utilization of tree peony resources of Hunan province of China. Simple sequence repeat (SSR) markers were applied to reveal the genetic diversity and relationship of 21 tree peony resources and 45 domestic and foreign tree peony cultivars. Clear bands, the size of which ranged from 115 bp to 379 bp, were detected with 14 primers. In total, 90 alleles were detected and the number of alleles detected with one primer varied between 5 and 13; the number of effective alleles ranged from 1.183 to 2.070; the polymorphism ratio of each locus was 100%. The observed heterozygosity, which ranged from 0.120 to 0.851 with an average of 0.532, was larger than the expected one, which ranged from 0.090 to 0.470 with an average of 0.300. Shannon index ranged from 0.137 to 0.695 and fixation index ranged from −0.332 to −0.869. The results show abundant genetic diversity in tree peony of Hunan province and SSR markers distinguishing homonymous tree peony resources successfully.
