This discusses the prospects for understanding the genetic basis of few selected traits in humans. Phylogenetic analysis provides a path to identify genes and biological processes underlying any trait influenced by inheritance. We suggest that different experimental approaches should be undertaken for traits caused by common genetic variants versus those arising from rare genetic variants. We discuss the intellectual foundations of genetic of Mendelian traits in humans by examining the degree of freedom of four particular factors , namely toung rolling (1.307), hitchhiker’s tumb (3.22), widow’s peak (44.825) and mid digital hair (92.825).

Opis:
Emerging of dengue infection has increased dramatically in Sri Lanka in recent years. Initial infection with a particular serotype is known as primary infection which is usually asymptomatic or mild disease manifestations. Although cross reactive T cells and cross reactive antibodies have been shown to contribute to disease pathogenesis, these mechanisms alone do not explain the immune - pathological mechanisms leading to severe infection. However, this study has suggested that antigen antibody detection and viral RNA detection in host blood contribute to the occurrence of disease. Therefore, it is important to further investigate the quality of dengue specific immune responses and confirmation of dengue RNA content in patients with acute severe dengue and asymptomatic dengue infection.

The MTHFR gene within the human genome, codes for the synthesis of Methylenetetrahydrofolate Reductase enzyme, which reduces 5,10-Methylenetetrahydrofolate to 5-Methyltetrahydrofolate, which in turn, is the major circulatory form of folate in the blood. Folate, in this form, among it’s other functions, is involved in reducing the homocysteine levels in the blood, whose elevated levels lead to Hyperhomocysteinemia, causing various major disorders. Mutations within the gene lead to impairment of gene function, in turn causing the homocysteine levels to rise. The C677T and A1298C mutations are the main causative agents for MTHFR gene disruption. During the course of the project, a total of 79 samples were analyzed for the presence of these mutations. The blood samples were first subjected to PCR, giving two separate DNA fragments each responsible for either of the conditions. The fragments were then subjected to RFLP analysis to detect the mutations. The results were finally given with respect to the risk factor faced by each individual based on a molecular diagnostic point of view.

The quality assurance (QA) and quality control (QC) programmes of imaging equipment are key ingredients of a quality system. This study is aimed at measuring X-Ray machine parameters and comparing them with the American Association of Physicists in Medicine (AAPM) report No. 74. Also, this study made comparison with similar articles locally and internationally. The study was conducted in 12 X-Ray unit (A-L) located in Delta State. A MagicMax Universal Basic Unit was used alongside an XR Multidetector (silicon photodiode) to measure dose, dose rate, practical peak voltage (PPV), exposure time, current in milliampere (mA) and quantity (Q) in milliampere seconds (mAs). The XR Multidetector was positioned at 100 cm from the focus to image distance (FID) to make most measurements. Six (6) X-Ray units were above 20 years, accounting for 50% of the total X-Ray units used. 30% of X-Ray units > 20 years failed the QC test, which was 2.3 times higher than X-Ray units < 10 and 10-20 years respectively. On the average, 55% pass mark was achieved in the 12 X-Ray units across the state, while 26.11% of the test could not be done due to bad knobs and other challenges. Two (2) X-Ray units with an average age of 3 years, passed all the test. The study showed that many of the X-Ray units were old and certain machine parameters could not be assessed. The study also reveal that most X-Ray specifications manual were either misplaced or missing. Continuing QC programme is highly recommended in the studied areas.

Opis:
Yellow head virus is a common viral pathogen which infects cultured Penaeus monodon world-wide, resulting in immature larvae and the growth retardation. Therefore, several detection methods are recommended by the Office des Epizootics (OIE) for use in Asia including Sri Lanka. This study was attempted to evaluate the RT-PCR diagnostic method is the appropriate and accurate YHV detection method which is recommended by the OIE comparing with histological identification and haemolymph Smear method. A total of 150 individuals of Penaeus monodon, post larve samples obtained from different shrimp hatcheries located in Northwestern province were screened for the presence of YHV and the results were comparatively analyzed with three different protocols, namely wet mount observation, histopathological observation and RT- PCR detection. Highest prevalence was recorded with wet mount observations (78%) and the lowest was recorded using histopathology (22%). Comparison of PCR tests generated consisted of positive samples 31/51 (2010), 38/65(2011) and 18/34 (2012) respectively. The results obtained with the IQ2000 commercial kit, were the most reliable and it indicated 56.6% average prevalence of YHV.

A total of 60 Russ 2011 two weeks old broiler chicks with an average weight of 215.33+4.55g were fed with diets compounded with sweet potato leaves (SPL) for 56 days. The diets consist of T1 (0% SPL), T2 (5% SPL), T3 (10% SPL) and T4 (15% SPL). The proximate composition of the SPL was determined in the laboratory. The birds were randomly allocated to four treatments using completely randomized design (CRD). Each treatment was subdivided into three replicates of five chicks each. At the end of the experiment, all data collected were subjected to one way analysis of variance and difference in means was separated using Duncan’s New Multiple Range Test. The results showed no significant difference (p>0.05) in the average body weight gain, feed intake, feed conversion ratio (FCR) and protein efficiency ratio (PER). However, birds on T2 (32.77g) had the best daily body weight gain followed by T4 (30.36g), T3 (30.06g) and T1 (29.21g) while in terms of daily feed intake, T4 (98.20g) consumed most followed by T2 (98.17g) and T3 (96.59g) whereas, T1 (92.38g) consumed the least feed. The birds on T2 (3.00) demonstrated the best FCR followed by T1 (3.16), T3 (3.21) and T4 (3.23). Also T1 (1.59) showed the best PER closely followed by T1 (1.53), T3 (1.46) and T4 (1.43). There was, however, a significant difference (p<0.05) in Hb concentration when T2 (11.62g/dl) and T3 (12.48g/dl) were compared with T1 (10.34g/dl) and T4 (10.21g/dl). In the PCV, a statistical difference (p<0.05) also was observed between T1 (24.73%) and others (T2 (28.18%), T3 (28.97%)and T4 (27.14%)) while similar trend (p<0.05) was seen in the RBC count when T4 (9.72x106/ul) was compared with T1 (10.21 x106/ul), T2 (10.81x106/ul) and T3 (11.43x106/ul) and no significant difference (p>0.05) among all the treatments with respect to WBC (T1 (21.23x103/ul), T2 (19.18x103/ul), T3 (21.26x103/ul) and T4 (22.63x103/ul)). Although the haematological parameters fell within normal physiological ranges, there was a gradual increase in these values as the inclusion levels progressed till T3 (10% inclusion) and then a decrease. It is therefore recommended that inclusion of SPL up to 5% in the diet of broilers is good for enhanced body weight gain, FCR and PER and up to 10% for enhanced haematological parameters.

Medical exposure for abdomen radiography is associated with higher doses compared to X-Rays of the chest and other extremities. The study aims to determine the mean entrance skin dose (ESD) for 104 adult patients between 20-89 years with 2 X-Ray units (A and B) in Aba, South-East Nigeria and to determine the ESD at the 75th percentile to estimate the local diagnostic reference levels (LDRLs). This study also determined the effective dose (E), the dose area product (DAP) and the relationship between absorbed dose (DT) and other parameters. This study will also compare its findings with relevant articles where necessary. The study used 2 functional floors mounted X-Ray units. A total of 208 annealed thermoluminescent dosimeters (TLDs) were used made of Lithium Fluoride, doped with Magnesium and Titanium (LiF: Mg, Ti). Two TLD chips were used per patient. The chips were positioned at the anterior and posterior end of the patient respectively for a given beam area for abdomen radiography. After exposure, a calibrated RadPro TLDcube 400 reader (Freiberg Instrument, Germany) was used to estimate individual patient doses. This was done by multiplying the TLD counts by a pre-determined calibration factor (CF). The mean/75th percentile ESD for facilities A and B was 2.92/4.12 and 3.01/3.67 mGy. The E for facilities A and B was 0.73 and 0.82 mSv respectively. There was a good relationship between the DT with ESD, exit dose (ED) and DAP for facility A, but no relationship was seen with other parameters. The mean ESD was lower compared to the National Council on Radiation Protection and Measurements (NCRP 172) and the American Association of Physicists in Medicine (AAPM) reports respectively. The study proved useful and could serve as a reference point to initiate LDRLs within the South-East zone in Nigeria for abdomen radiography.

Opis:
White spot syndrome is a viral infection of Penaeus monodon shrimp. It is highly lethal, contagious and killing shrimps and doesn’t have a specific anti-viral treatment for this infection. This important infection affects on shrimp farms in Sri Lanka and leads to the loss of economy. Most of shrimp products are export oriented. Therefore this study aimed to determine the factors, detection and prevalence of WSSV in shrimp farms, Sri Lanka. The prevalence rate of WSSV among the collected samples (n = 100) is 58.05% (2011), 59.06% (2012) in Sri Lanka respectively. Therefore the infection and resistance of virus has increased among the shrimp population annually. Further studies need to be identified the WSSV aggressive viral strains and prevention strategies to evade from this sever infection in shrimp farming.

Cervical cancer is the second commonest cause of cancer among women. According to the World Health Organization (WHO) data, in 2006 there were 1.4 million women who were suffering from cervical cancer and 80% of deaths among them were of women from developing countries. Genital infection with Human Papilloma Virus (HPV) is one of the major etiological factors for developing cervical cancers in almost all countries (World Health Organization, 2005). Papilloma viruses were first identified, cloned and sequenced from cervical tumor specimens and were subsequently established as important causative agents for the development of cervical cancer. Differences in prevalence rates are observed worldwide. Infection is more common in sexually active young women of 18-30 years and a reduction in acquisition of infection is observed after 30 years. The situation of Cervical cancer in Sri Lanka is identified as the second most common cancer among women in Sri Lanka and approximately 7.74 million women are included in the risk category. It is reported that nearly 850 die from cervical cancer each year.

In the current study we attempt to identify the relationship of among geckos species that are surviving in Adam’s peak Sri Lanka. A ten month survey was undertaken to document the diversity and abundance of geckos at the Adam’s peak in Sri Lanka. By using molecular data, the aim was to genetically characterize the species of geckos occurring along directions in Adam’s peak and phylogenetically relate these to other different parts of the world. A total of 79 individuals of gecko’s tails and eggs were captured in four different areas in Adam’s peak. We use 373 bp of RAG1 to detect the gene location and DNA sequence data to recover phylogenetic relationships and elucidate a biographic pattern and close relationship among geckos. According to the morphological identification suggested that gecko’s tail specimens belonged to several species and the molecular detection method confirmed the gene pool is differentiated among the population. Analysis of sequences of the RAG1 region from gecko species confirmed that the samples belonged to family Gekkonidae with the different gene population.

The reagent NaHMICdt•2H2O has found to be a good chelating agent for the spectrophotometric determination of various metal ions. The present investigation deals with spectrophotometric determination of Pd(II) by using NaHMICdt•2H2O. The reagent reacted with Pd(II) to give yellow complex extractable into toluene. Pd (II) was extracted quantitatively in the pH range 0.5-2.0. Pd(II)-HMICdt complex showed λmax at 435 nm. The extracted species has a 1:2 (M:L) stoicheometry. Beer’s law is obeyed in the range of 0.2-0.8 μg ml-1 of Pd (II). The molar absorptivity and the sandells sensitivity were found to be 0.754 x 104 lmol-1cm-1 and 0.0140 μg mol-1 cm-2 respectively. The effect of divers ion on the extraction of Pd(II) has been investigated. The proposed method is highly sensitive, selective, and simple and rapid has been applied for accurate determination of palladium in synthesis mixtures and real samples.

A fashion trend is emerging in Islamic culture based on the concept of modest fashion all over the world. Significant brands of the Western fashion world have already launched their Hijab and Abaya lines. The objective of the present research is to investigate the important factors of modest fashion in the Islamic culture and to detect the future world fashion trends of Islamic fashion and then to appliance the concept of modesty to the fashion market of Sri Lanka. This investigative study is based on the qualitative and questionnaire-based methods. The study is confined to questionnaires which were distributed among a sample of 150 Muslim women with diverse backgrounds to gather data and statistics around Colombo and Kandy. Secondary data were collected from books, articles, web sites and visual data such as pictures and photographs which facilitated to forward a successful research. Examination of the influence of culture on Muslim women’s dress etiquettes led to understand what Muslim women expect from their dress and led to identify potential opportunities in the contemporary market of modest fashion.

El-Oued is known for some diseases caused by fluoride concentration in drinkable water. To reduce it, we have chosen a sample with the highest content of fluoride among many sources in order to coagulate it with Al2(SO4)3ź18H2O, Fe2(SO4)3źH2O and FeSO4. In order to get better reduction yield of fluoride, a study has been done on the influencing parameters (concentration, pH, temperature) to choose the best conditions. The remove of fluoride is favorable at low concentration of Al2(SO4)3, at room temperature and normal acidity.
