Opis:
The effect of administration of Methanolic extract of Achyranthsu asper on biochemical parameter of mice were investigated. This drug induced significant reduction in the cholesterol level and amount of RNA. But significant increase in the amount of glycogen at 25 mg/kg and no significant reduction in the amount of glycogen at 50 mg/kg. This drug also induced increase in the level of alkaline phospatese and decrease in the level of acid phosphatase, protein and DNA, but these changes are statistically not significant. Protein 25 mg shows 4.22 ±0.112 and 50 mg shows 4.05 ±0.085. Cholesterol 25mg shows 0.5 ±0.04 and 50 mg shows 0.39 ±0.038. Glycogen 25mg shows 0.288 ±0.011 and 50mg shows 0.248 ±0.177. DNA 25mg shows 0.109 ±0.039 and 50 mg shows 0.085 ±0.035, RNA 25mg shows 0.38 ±0.036 and 50 mg shows 0.25 ±0.031

Malaria is an important health and development challenge in Africa, Animal models most particularly mice, have long been employed to study malaria pathogenesis. This paper describes clinical manifestations due to Plasmodium berghei ANKA infection in juvenile mice as a model for understanding the complications of congenital malaria in neonates. Forty-five juvenile mice (5-7 days old) were acquired from University College Hospital, Ibadan and injected with 2×107 (0.2 ml) Plasmodium berghei ANKA parasitized red blood cells (PRBCs). The mice were then transported to the study site, kept in well-ventilated cages and fed daily with a balanced ration. Post-P. berghei infection, the mice were monitored daily for mortality. Clinical manifestations of experimental cerebral malaria (ECM) were assessed and confirmed if at least ruffled fur, hunching, wobbly gait, limb paralysis, convulsions, or coma was observed. Each sign was given a score of 1. Animals with scores ≥4 were considered to have severe ECM. In the experiment, 20 (44%) mice were lost due to natural cause (i.e. stress) at day 2. Between day 4 and 9, 25 (56%) of the study mice presented clinical signs of ECM. This included: ruffled fur – 25 (100%), hunching - 21 (84%), wobbly gait - 17 (68%), limb paralysis - 20 (80%), convulsions - 25 (100%). Survival rate and severity of ECM in the mice differs, 22 (88.0%) had severe ECM and 3 (12.0%) had mild ECM. This study has shown that parasite establishment and malaria complications can manifest as early as 4 days’ post P. berghei infection in 5-7 days old mice.

The present study was aimed to evaluate the activity of Achyranthes aspera against allergy caused by heavy metal potassium dichromate in albino mice. The potassium dichloromate as induced at a concentration of 20 mg/kg. body weight dissolved in distilled water for 10 days continuously. On 11th day, blood was drawn from mice for serum analysis of IgM and IgG to confirm allergy induction in mice. After allergy induction the mice received petroleum ether, chloroform and methanolic extracts of Achyranthes aspera given 3 doses at concentration of – 200 (low), 400 (medium) and 600 (high) mg/kg to mice by oral route for 7 days after allergy induction. On the 8th day, blood was drawn from mice for serum analysis of IgM and IgG in determine the effect of extracts on serum Titre of mice blood.

Opis:
Acknowledging the immunostimulation effect of ethanol extract white oyster mushroom to animal experiments which were desensitized with red blood cell of lamb. This study is an experimental study, 14 healthy male swiss Webster mices were used. The subjects are randomly divided into two groups. The first group is a control group and the second group is a treatment group. The immunostimulation effect is assessed by specific immune response to desensitized of lamb’s RBC. Humoral immune response was assessed through a hemagglutination test. Cellular immune response was assessed with a delayed type hypersensitivity test by measuring the thickness of edema of the mice’s foot Specific immune response is assessed through total of blood lymphocyte using flowcytometry method. We found an increase of total blood lymphocyte level on the primary sensitization of placebo and Ethanol extract of white oyster mushroom (EEWOM). After secondary sensitization, the total blood lymphocyte level on placebo and EEWOM showed a significant difference. EEWOM increases the total antibody titre significantly compare to placebo. The level of serum gamma-globulin of treatment mice is significantly higher than placebo. On the 24th and 48th hour after the sensitization of lamb’s RBC, the increase of thickness of the mice's feet which were given placebo and EEWOM have significant difference. EEWOM has an immunostimulating effect towards specific immune response.

The present investigation was aimed to study an antiepileptic activity of methanolic extract of Tragia involucrata Linn in mice. In vivo screening models like maximal electroshock-induced convulsion (MES), pentylenetetrazole (PTZ) and picrotoxin (PTX) induced models are used to evaluate the antiepileptic effects of the extracts. The biochemical estimation was done by measuring the lipid peroxidation and reduced glutathione (GSH). In the MES induced convulsion, methanolic extract of Tragia involucrata (METI) at high dose (800 mg/kg body weight), showed high significant inhibition on tonic hind limb extension (THLE, 6.83 ±0.30***) and decrease in duration of stupor period (108.7 ±6.53***). In PTZ and PTX induced model METI (400 mg/kg and 800 mg/kg) showed significant delay on the onset of convulsions, decreased duration of convulsion and reduced mortality significantly. It also showed significant decrease in brain MDA level in lipid peroxidation profile, and increase in the brain glutathione levels in mice against PTZ induced convulsion. The results confirmed that Tragia involucrata Linn possesses dose dependent antiepileptic activity.

The objectives of present work focuses to check the acute and sub acute toxicity studies for the Achyranthes aspera L methanol extract as per OECD guideline in Swiss mice. The dose of 100, 200, 250 gm/kg body weight methanol extract by administering drug intraperitoneally and recorded the growth, body weight, organ weight, general symptoms, morphological and physiological behavior, mortality. These parameters on various organs system in mice were studied. It was considered necessary to assess its potential health hazard in man and to find the safe and effective dose. The observations of changes in body weight, food and water intake as well as cage side observations were reported. There was no abnormality observed in all groups. The whole plant powder methanol extract of Achyranthes Aspera L were found to be nontoxic.

The present study evaluated the protective effect of bergenin on high fat diet (HFD) induced diabetic mice. C57BL/6J mice were segregated in two groups, one fed standard diet (NC) and the other fed HFD for 16 weeks. Mice were fed continuously with high fat diet for 16 weeks and subjected to intragastric administration of bergenin (10, 20 and 40 mg/kg body weight (BW)), metformin (25 mg/kg BW) 9 to 16 weeks. At the end of the treatment nephritic markers, lipid peroxidation product, antioxidant and histopathological examination were carried out to assess the efficacy of the treatment. HFD fed mice showed increased plasma glucose, insulin, altered nephritic markers, antioxidant and histopathological abnormalities. Oral Treatment with bergenin (40 mg/kg BW) showed near normalized levels of plasma glucose, lipid peroxidation product, antioxidants, improved insulin and reduced kidney damage. The effects of bergenin were comparable with standard drug, metformin. These data suggest that bergenin protect kidney from deleterious effect of glucose.

Thespesia populnea Corr. floral extract has been shown to have anti-steroidogenic effect. Bark extract of Thespesia populnea has no toxicity as per the earlier reports of traditional medicine used by the tribal peoples of Indira Gandhi Wild Life Sanctuary, Pollachi, South India. In the present study, anti-steroidogenic effect of Thespesia populnea (Malvacea) bark was evaluated in adult female mice (Mus musculus). Methanolic extract of Thespesia populnea bark (METP) was administered orally at the doses of 100, 250, 400 mg/kg body weight for a duration of 30 days, and normal saline (0.9%) to control group. There was significant reduction in the (P<0.05) wet weight of both ovaries. Cholesterol content in ovaries of METP treated mice was significantly increased (P<0.05). The two ovarian enzymes 3-,17-hydroxy steroid dehydrogenase and serum estrogen and progesterone also decreased (P<0.05) significantly. Administration of METP in mice did not show any variation in the hematological and bio-chemical parameters. A certain number of adult female mice were ovariectomized (OVX) and tested for hormone levels with EDP and with EDP + Extract to confirm the antifertility effect of METP. It is concluded that antifertility effect of METP could be due to its anti-steroidogenic activity.

Hybanthus ennespermus (L.) (Violaceae) has no toxicity asper the earlier reports of fertility traditional medicine used by the tribal people of Grizzled Giant Squirrel Wildlife Sanctuary, Srivilliputur, Southern India. In the present study, fertility effect of Hybanthus enneaspermus (Violaceae) whole part of plant evaluated in adult female mice (Mus musculus). Raw extract of Hybanthus enneaspermus (REHE) was administered orally at the doses of 100 mg/kg body weight for a duration of 30 days and normal salinw (0.9%) to control groups. There were no significance changes in the body weight (P<0.05). The Possibility of marked increase in serum estradiol level (P<0.05) significantly. Administered of REHE in mice did not show any variation in the hematological and biochemical parameters. It is raw extract of Hybanthus enneaspermus (REHE) showed on above result indicated that this alcoholic free extract of Hybanthus ennaespermus confirm the fertility effect of above study.

Nephroprotective effects (NPE) of simple Potentilla reptans - aqueous leaf extract (Pr-ALE) leaves on Paracetamol induced kidney poisonousness in wistar rats. Adult male wistar rats (weight range (WR): 200-220g) were divided into 6 groups (n=6). Paracetamol (PA) and Silymarin (SY) stayed managed intraperitoneally arranged the 5th day to rats in all groups but the normal control. Furthermore, a significant nephroprotective (NP) of the aqueous leaf extract (ALE) and oral dose of PA and SY. Pr-ALE did not mortality or significant changes in the body weight. Progression of nephrotoxicity (NT) induced by PA in rats was interfered by Pr-ALE managed, and these effects were correspond to those managed with SY. This is the first record on NPE of Pr against PA-induced NT.

Resistance training increases strength and stamina without enlarging muscle. Jaras ifg1 responsible for the proliferation and differentiation of muscle tissue in response to the exercise. Post-allocation of adaptation 36 research subjects were randomly allocated to the control group and the test. The purpose of this study to determine the effect of prolonged hanging test of grip strength, muscle mass changes and IGF-1r imunoekspresi the bicep muscles of mice. The uniqueness differences in physiological mechanisms increase the strength after resistance exercise through changes in the dynamics of unknown IGF1 receptors , so this time we did the study increased expression of IGF1 receptors (Igf1r) after resistance exercise, using new method is prolonged hanging test. Grip strength, muscle mass and score imunoekspresi igf1r in bicep muscles will be tested in the mice model (Swiss webster) using pre and posttest design. Examination of grip strength, muscle mass, muscle fiber cross-sectional diameter and imunoekspresi IGF-1r have done at day 28 in the Laboratory of Pathology in Dr. Hasan Sadikin Hospital, Bandung. Our results showed there were effect of prolonged hanging test to grip strength in the final test on mice and igf1r Imunoekspresi on mice. This research concluded that resistance exercise using hanging prolonged test increases grip strength and receptor expression igf1r without increasing muscle mass or muscle fiber cross-sectional diameter.
