In spite of the enormous achievements of medical and biological sciences, parasites still constitute important medical and veterinary problem. One of the main obstacles in the treatment and eradication of human and animals parasitic diseases is drug resistance of parasites. This phenomenon has more and more frequently been described both in medical and veterinary practice. The application of numerous molecular biology techniques in modern parasitological laboratories have enabled the investigations of parasites at the DNA level. The results of these studies have revolutionised our knowledge and made possible the recognition of not only the reasons and extent of genetic differentiation of parasites but also the recognition of genetic and molecular mechamisms of drug resistance. The results of many studies showed interspecific, interpopulational, and even intrapopulational heterogeneity of parasites in their sensitivity to antiparasitic drugs. Moreover, it has been found that in various parasites there often exist different molecular bases of the drug resistance. The review concerns different molecular mechanisms of drug resistance in selected parasite taxa (Schistosoma spp., Haemonchus contortus, Leishmania spp., Plasmodium falciparum, Entamoeba histolytica).

Resistance in the hematophagous arthropods - problem on the rise. I. Resistance mechanisms to insecticides. Hematophagous arthropods are pesky but can also be very dangerous to human health due to their ability to act as vectors to many viral, bacterial or parasite-related diseases. The common application of chemicals to control pests leaded to the increasing resistance to pesticides among both insects and ticks. The causes underlying the appearance of the resistance and the mechanisms involved are presented in this article. The description of mechanisms is presented starting from decreased penetration of the pesticide, to an increase in metabolism of pesticide, to the reduced sensitivity of the pesticide's target.

In this review we summarize the great amount of recent information on the apoptosis in aspeets of the host-parasite interaction. Although apoptosis is a form of programmed cell death which plays a pivotal role in normal tissue development a plethora of pathogens including parasitic protista and helminths are able to modulate host apoptosis pathways to their own advantage. Here in we present and discuss new research data and results describing the phenomenon as a process have been controlled by gene expression, biochemical reactions and receptor-ligand interactions at the cell membrane surface. Section | describes apoptosis as ongoing process in normal tissue development. Section 2 analyzes the role of apoptosis in outcome of infection and pathogenesis of several disorders evoked by viruses and bacteria. The cellular mechanisms of cell death during infection with unicellular parasites such as Leishmania sp. and Plasmodium sp. are described in Section 3. In the next paragraph the potency of parasitic protista and helmiths for modulation host apoptosis pathways to their own advantage is discussed. The involvement of apoptosis in immunoregulation of the host immune function was proposed as a one of possible mechanism in creation of the host-parasite relationship. The molecular and cellular mechanisms of parasite-induced immune response via apoptosis pathways are discussed. We conclude that novel strategies for the management of the host-parasite relationships need to be explained into the mechanisms by which parasites induced apoptosis in contribution to the activity of immune system of the host.

The mechanisms of discrimination between self and non-self are presented in detail, including characterization of interacting components of host (cellular and humoral factors) and parasite (a sporocyst surface) origin. The phenomena of molecular disguise and molecular mimicry are discussed in the context of the compatibility betweeen a host and a parasite. The possible ways of killing parasite are indicated, with special regard to production of reactive oxygen intermediates by hemocytes during the process of killing.

In mycoses congenital – nonspecific innate as well as acquired immunity (involving neutrophiles, monocytes, macrophages, dendritic cells and lymphocytes) both play important roles in host defence. Th1 lymphocytes release cytokines (IL-2, IL-12, IFNg) and stimulate cytotoxic cells and neutrophiles to destroy fungal cells. Th2 lymphocytes, on the other hand, suppress cellular immunity by releasing the cytokines IL-4, IL-6 and IL-10 which counter regulate the secretion of IL-2, IL-12, IFNg and depress the activity of macrophages. Cellular mechanisms play essential roles in host responses to fungal infections. Dysfunction of T lymphocytes and a reduction in their number are typically observed in patients with mycotic diseases. There occurs a reduction of both T lymphocyte populations and the T-helper to T-suppressor cell number ratio, and these are of critical importance in explaining the diminished IgA production and enhanced adhesion of fungal cells to the surface of host cells as well as in facilitating the intrusion of fungi throughout the skin and mucous membranes. The specific immunological reaction, associated with the synthesis of antibodies against fungal cell wall or cytoplasmic antigens, is of little significance in protective immunity, but nevertheless has a rather important role to play in diagnosis as well as in supporting phagocytosis by inhibition of fungal cell adherence. In patients with mycoses, typically low blood serum level of the immunoglobulin class G and A and low sIgA in saliva are observed. A detailed understanding the nature and function of the immune system in mycoses is necessary to enable improvements in pharmacotherapy with antifungal antibiotics and chemotherapeutics, as well as to treatments based on immunotherapy and vaccination.

As can be seen from many studies protozoan and helminth parasites appear to have one or more anti-oxidant enzymes able to scavenge or quench the reactive free-radicals, and there is strong evidence that such enzymes may play crucial role in protecting against host response. Presented there review illustrates that methods to block anti-oxidant protection of parasites may be new field in the search for improved ways to inhibit parasitic survival.

Nitric oxide (NO) has been shown to play a crucial role in various physiological and pathological conditions. NO plays a role in the immunoregulation and it is implicated in the host non-specific defence in a variety of infections. Abundant evidence indicates that NO contributes to the host defence function of macrophages. High levels of NO are mediated by up-regulated expression of the iNOS gene in response to the activating signals, in particular to the secretion of pro-inflammatory cytokines (IFN-y, TNF-a, IL-1, IL-2) by Thl cells. In this review, the role of NO during a number of parasitic infections has been summarized. Up to now, enhanced levels of NO production and expression of iNOS gene have been described in such infective diseases as malaria, toxoplasmosis, leishmaniosis, trypanosomosis and schistosomosis. During these infections, the preferential production of pro-inflammatory cytokines predisposes to the increased synthesis of NO, which mediates host protection through either direct parasite killing or by limiting parasite growth. The evidence presented in this review supports the conclusion that NO plays an important role in the majority of parasitic infections.
