The aim of our study was to evaluate and compare antioxidant and antimicrobial activity of aqueous, methanol and chloroform extracts of leaves of Allium ursinum (A. ursinum). Total phenol (TPC) and flavonoid (TFC) content in extracts and antioxidant activity using DPPH (1,1- diphenyl-2-picrylhydrazyl) assay were determined spectrophotometrically. In vitro antimicrobial activity was tested by microdilution method. The highest TPC and TFC were observed for chloroform extract. The extracts showed different degrees of antimicrobial activity. The intensity of antimicrobial action varied depending on the group of microorganism and the type of extracts. Our results demonstrated rational basis for the traditional uses of A. ursinum in alleviation of oxidative stress and against various pathogenic microorganisms.

SSixteen ethanolic extracts were obtained from seven different bee bread samples. The total flavonoid content in the extracts was determined by aluminium-chloride method and was in the range of 8.3 mg/L±6.24% to 195.3 mg/L±1.35% and 28.8 mg/L±19.33% to 603.3 mg/L±4.64% with reference to quercetin and rutin, respectively. The relative standard deviations (RSD) for parallel measurements for the calibration curves of quercetin dehydrate and rutin trihydrate were in the range of 0.51% to 9.39% and 5.02% to 19.91%, respectively. The RSD for parallel measurements for the extracts with reference to quercetin dihydrate and rutin trihydrate were in the range of 0.23% to 11.64% and 4.64% 19.33%, respectively. The total flavonoid content mainly depended on a ratio of bee bread to 50% ethanol and technology of obtaining bee bread. The significant differences between results were statistically confirmed. The best antibacterial activity of bee bread extracts was found against Bacillus cereus CCM 2010, Clostridium perfringens CCM 4435, and Staphylococcus aureus subsp. aureus CCM 4223. The activity of the bee bread extracts against Gram negative bacteria, Aspergillus and Penicillium genera was lower with moderate anticandidal activity. The obtained results indicated that it was very important to employ extracts with a high content of bee bread in 50% ethanol (1:5, 1:10). According to the results of this study, bee bread is a product which is rich in flavonoids and with good antibacterial activity against Gram positive bacteria and can be considered as a raw material for development of diet supplements and antimicrobial medicinal products

The study analyzed colour intensity, the total phenolic and flavonoid contents, as well as antioxidant and antibacterial activities of honeys from Serbia. All the tested honeys contained considerable levels of phenolics. The highest phenolic content was obtained in forest honey - 1389.71 mg gallic acid equivalents/kg. High levels of phenolics and flavonoids were also determined in honeydew, meadow and oregano honeys. The radical scavenging activity measured by DPPH method in linden and forest honey was 0.45 and 2.75 mmol Trolox equivalents/kg, respectively. The ferric reducing potential was the greatest in forest honey - 6.04 mmol Fe2+/kg. Darker honeys had a greater phenolic content and antioxidant activity. The antimicrobial activity of honeys against six bacterial strains isolated from human material were studied using a microwell dilution assay. The minimal inhibitory concentration of honeys ranged from 6.25% w/w (for oregano honey against Enterococcus faecalis and honeydew honey against Streptococcus pneumoniae) to >50% w/w (meadow honey against Staphylococcus aureus). The minimal bactericidal concentration ranged from 12.5% w/w (honeydew and bee pollen enriched honey against E. faecalis) to >50% w/w. Higher values of antimicrobial activities were found in honeydew, oregano, and forest honeys, while meadow honey showed the lowest antimicrobial activity. Analyzed honeys exhibited a strong antioxidant and broad-spectrum antibacterial activity against multidrug-resistant pathogenic bacteria. Phenolic compounds, especially in dark amber forest and honeydew honey, as well as in amber oregano honey, may contribute to their efficacy in therapeutic administration.

On account of the interesting pharmacological properties of hydantoin and 2-thiohydantoin derivatives, and other purposes such as textile printing, catalysts for polymerizations, their use in the production of resins and plastics, they have been the subject of chemical and biological studies. In this review article, we focused our interest on the most important methods for the synthesis of hydantoin and 2-thiohydantoin derivatives, different reactions and different application of them. We explored their antitumor, antimicrobial, antiviral, anxiolytic, antidiabetic, anticonvulsant activities, their utilities in treatment of vascular dysfunction, using them as an inhibitor of fatty acid amide hydrolase, using in hyperpigmentation applications, and utilities of them in the production of antimicrobial polyurethane coatings.

Some new isoxazole derivatives 3a-d were synthesized via the reaction of 3-(dimethylamino)-1-(5-methyl-1-(3-nitrophenyl)-1H-1,2,3-triazol-4-yl)prop-2-en-1-one (1) with different hydroximoyl chlorides derivatives 2a-d. From these new isoxazoles 3a-d a new series of isoxazolopyridazines 4a-d was derived using hydrazine hydrate. In addition, enaminone 1 was reacted with ethyl acetoacetate to afford the corresponding ester derivative 6, the latter was submitted to react with different chemical reagents to obtain a variety of bioactive substituted pyridine derivatives. The azido derivative 14, was used as the key molecule for the synthesis of new urea and aryl carbamate derivatives upon its reaction with different amines and phenol through Curtius rearrangement. The chemical structures of all new compounds were investigated from their spectral and microanalytical data. The synthesized compounds were tested for their pharmacological potency as, anti-hepatic cancer and anti-microbial agents. Most of the tested compounds showed good anti-hepatic cancer results comparing with the standard drug doxorubicin especially when their toxic effects on the normal cell lines were studied. Referring to the anti-microbial test most of the compounds showed strong effects.

Opis:
Macroalgae from Fucus genus are a valuable source of bioactive components as they are abundant in complex polysaccharides, fatty acids and polyphenols. In this work, the biological activity and chemical composition of extracts and fractions obtained from endemic Fucus virsoides J. Agardh species collected in the summer and in the fall were investigated. From dichloromethane:methanol (1:1) extract three fractions were made: petroleum-ether, ethyl-acetate and n-butanol. The aim of the study was to examine the influence of the seasonal variations on algal composition and activity. The significant seasonal variation in content and biological activity of Fucus virsoides samples was found. Fall extract and fractions exerted higher cytotoxic effects on cancer cell lines in comparison with summer extract and fractions. The examined extracts and fractions showed higher cytotoxic activity towards cancer cells compared to normal fibroblast MRC5 cells. Morphological evaluation and cell cycle distribution analysis demonstrated their proapoptotic activity in human cervical adenocarcinoma HeLa cells. Fall extract and fractions better suppressed the migration and tube formation of EA.hy926 cells in comparison with summer extract and fractions. Fall extract and fractions were more potent in inhibition of α-glucosidase enzymatic activity. Ethyl-acetate fractions, from both seasons, exhibited the best antibacterial and antifungal activity on all tested bacteria and fungi. In conclusion, the two fall fractions ethyl-acetate and petroleum-ether rich in polyphenols and polyunsaturated fatty acids, were the most active and exhibited prominent anticancer and anti-α-glucosidase activities.

Human neutrophil peptides (HNPs, alpha-defensins) are a group of six defensins being considered as new antimicrobial drugs because of their multifunctional efficiency against bacteria, viruses and fungi. Regardless of the unique biological properties alpha-defensins are unstable compounds and their activity depends on many physical and chemical factors as well as on the kind of the used cultivation media. This leads to research difficulties and obstructions in their therapeutic application. The purpose of this study was to determine antibacterial activity of alpha-defensin 2 dissolved in pharmacopeia buffers and in parallel to develop selective and accurate analytical tests for identification and assay of alpha-defensin 2 in the course of study. The antibacterial effect of alpha-defensin 2 was determined against the strain Escherichia coli (ATCC 25922). It was found that 10 mg/l of Human neutrophil peptide-2 (HNP-2, alpha-defensin 2) dissolved in pH 9.0 buffer caused 90% inhibition of the bacterial respiratory activity. This buffer was considered as a suitable environment for deploying the antibacterial activity of the alpha-defensin. А selective MS analysis method for identification of alpha-defensin 2 in sample mixtures was developed. Also HPLC method with alternative selectivity was elaborated for identification and assay of mixtures containing alpha-defensin 2 and aminoacids in Mueller Hinton broth. The procedure includes development of system suitability test determination.

This study analyzed chemical composition of the essential oils (EOs) of Thymus vulgaris L., harvested in two regions of northwest Algeria (Tlemcen and Mostaganem), and evaluated their antimicrobial and cytotoxic activity. Fifty one compounds representing 99.99% and 100% of total thyme oil of Mostaganem (EO.TM) and Tlemcen (EO.TT), respectively were identified by GC-FID and GC-MS. Carvacrol was a dominant compound in both oils (89.15 and 82.42%). The antimicrobial activity, demonstrated by disc diffusion and broth macrodilution methods, showed a strong inhibitory effect of the oils on seven reference strains tested and particularly on Bacillus cereus ATCC 10876 and Candida albicans ATCC 10231. A cytotoxicity assay for two normal and two cancer cell lines indicated the highest sensitivity of canine B-cell lymphoma cell line (CLBL-1) for which IC50 values were 35.39 ± 9.0 (EO.TT) and 84.51± 3.2 (EO.TM). Murine fibroblast cell line (3T3), with IC50 116.97 ± 2.7 for EO.TT and 146.29 ± 0.5 for EO.TM was the most resistant line.
