Opis:
Aspergillus glaucus is a fungus able to tolerate low water activity of the environment. Its dense growth and sporulation were found on basaltic stones with epilithic lichens after 14 years of storage at a temperature of 4–7°C and relative humidity of 14–18%. Dust and soil particles deposited on the lichen thalli and dissolved in the water condensed on the stones during the storage period, apparently served as a nutrient source for the fungus. Probably, strongly xeric water regime on basaltic stones suitable for A. glaucus did not allow mesophilic fungi to develop and prevented the xerotolerant fungus from competition with other microfungi for nutrient sources. It is also possible that specific cellular mechanism associated with the production of chaotropic compounds (such as glycerol) supported germination and development of A. glaucus at low temperatures, which were considered non-optimal for the fungus.

Opis:
The growth of Topospora myrtilli isolates, obtained in the years 2001-2003, was studied on PDA medium at –10, +2, +6, +12, +18, +22 and +28oC. The growth and sporulation of the isolates were observed at +22oC on the following culture media: PDA, maltose – MA and mineral – MSA, as well as on two versions of oat media, one containing 20 g of oat fl akes per litre (OA-20), and the other containing 50 g of oat fl akes per litre (OA-50). The observations of the linear growth of colonies of the studied strains were carried out during a period of 21 days, whereas the formation of morphological structures until the 52nd day of cultivation. It was found that T. myrtilli can grow and sporulate in a wide range of temperature, even at +2oC. The pathogen growth activates with the increase of temperature, up to the optimal temperature, i.e. from +18oC to +22oC. The increase of temperature to +28oC did not favour the mycelium growth and prevents pathogen sporulation. PDA, OA-20, OA- 50 and MA media can be considered to be the most suitable for the growth and development of T. myrtilli, due to the intensive growth and formation of typical macro- and microscopic features. It is necessary to emphasise the great usefulness of oat medium, especially OA-50, for the cultivation of Topospora myrtilli, in the aspect of sporulation of the fungus. It was found that mineral medium is unsuitable due to the formation of mycelium with an untypical structure and colouration as well as late conidial sporulation.
Wzrost izolatów Topospora myrtilli, uzyskanych w latach 2001-2003 badano w temperaturze: – 10, +2, +6, +12, +18, +22 i +28oC na pożywce PDA. Ponadto w temperaturze +22oC obserwowano wzrost i zarodnikowanie izolatów na pożywkach: PDA, MA, MSA oraz dwóch pożywkach owsianych: jednej o zawartości 20 g płatków owsianych na 1 litr – OA – 20, drugiej 50 g płatków owsianych na 1 litr – OA – 50. Obserwacje wzrostu liniowego kolonii badanych szczepów prowadzono przez 21 dni, a tworzenie struktur morfologicznych do 52-ego dnia hodowli. Ustalono, że T. myrtilli może rosnąć i zarodnikować w szerokim zakresie temperatury. Wskazuje na to możliwość wzrostu wegetatywnego grzyba nawet w temperaturze +2oC. Wraz ze wzrostem temperatury uaktywnia się rozwój patogena, aż do temperatury optymalnej od +18oC do +22oC. Temperatura +28oC nie sprzyja rozwojowi grzybni i uniemożliwia zarodnikowanie patogena. Ze względu na intensywny wzrost i tworzenie typowych cech makro- i mikroskopowych za najodpowiedniejsze podłoża dla wzrostu i rozwoju T. myrtilli można uznać pożywki: PDA, owsianą OA-20, owsianą OA-50 oraz maltozową. Spośród wymienionych należy podkreślić dużą przydatność do hodowli T. myrtilli pożywki owsianej, a zwłaszcza OA-50 w aspekcie zarodnikowania grzyba. Pożywka mineralna okazała się nieodpowiednia, ze względu na tworzenie grzybni o nietypowej strukturze i zabarwieniu oraz późne zarodnikowanie konidialne.

Sixteen identified and three unidentified species belonging to six genera of zoosporic fungi were isolated from forty water samples which were collected from different fish and fish hatcheries farms at Abbassa city, Sharkiya governorate, Egypt, using sesame seeds baiting technique at 20±2°C. Saprolegnia and Achlya contributed the broadest spectra of species diversity amongst the other genera of zoosporic fungi. Saprolegnia diclina and Aphanomyces sp. were the most prevalent species of zoosporic fungi. The abundance of zoosporic fungal species in these aquacultures was correlated with some physicochemical characteristics of the water samples. The two dominant species of zoosporic fungi were tested for their tolerance of NaCl solution and its impact on some morphological and metabolic activities of these fungi. Saprolegnia diclina tolerated concentrations of NaCl solution till 12000 μg/ml whereas the maximum resistance of Aphanomyces sp. was 8000 μg/ml. The examined morphological aspects of the two studied fungal species, which included the colony diameters, the vegetative hyphae, zoosporogenesis, zoospores discharge, sexual reproductive structures and gemmae formation, were generally affected depending upon the tested fungal species and the applied dose of NaCl solution. The low treatments of NaCl solution were significantly stimulative compared with the control for protease production by S. diclina but higher doses were significantly suppressive. A significant decline in protease activity at all applications was found when Aphanomyces sp. was treated with NaCl solution. The total free amino acids and total protein content of S. diclina and Aphanomyces sp. mycelia were almost significantly increased relative to untreated controls at the low dose of NaCl solution and they were significantly dropped at the higher concentrations by the two zoosporic fungi.

Opis:
Growth and sporulation of 20 single-spore isolates of Drechslera avenae grown on different agar media were investigated at 24oC and 11oC. Four different agar media were used – water agar (WA, 20 g agar x l-1), maltose-peptone agar (MPA, 20 g maltose from Difco, 2.5 g peptone, 20 g agar x l-1), potatodextrose agar (PDA, from Difco, 39 g x l-1), and rye agar (RA). The isolates were evaluated in respect of growth, colony colour, and formation of morphological structures. The study showed differences between isolates in respect of colony growth, depending on culture conditions. Comparing macroscopic features of all D. avenae isolates tested, three groups of strains formed equal colonies and four individual strains were separated. Generally, most strains, except strains number: 2, 4, 7, 9, 10, 13 and 17, grew faster on all agar media at 24oC than at 11oC. In case of D. avenae isolates, hyphal growth was the fastest on MPA medium and the slowest on WA. All strains investigated produced typical conidiophores and conidia both at 24oC and 11oC, but at 24oC earlier sporulation was observed. MPA and RA were the best media for the sporulation of D. avenae strains. Sporulation of this species was the latest on WA.
