Opis:
The aim of the study was to assess the antifungal activity of preparation „fin Candimis” (oregano essential oil) against yeast-like strains belonging to the genus Candida. During the investigation, there were used up nine Candida albicans strains and ten C. glabrata strains isolated from different clinical material, along with one C. albicans demonstration strain ATCC 90028. The oregano essential oil, utilized in the study, was obtained from fresh leaves of Origanum vulgare L. and bore a trade name „fin Candimis”. According to data yielded by its manufacturer, concentration of pure oregano essential oil in preparation „fin Candimis” totals up to 210 mg/ml. The susceptibility of the Candida strains to preparation „fin Candimis” was assessed by means of the disc-diffusion method, upon the Sabouraud solid medium (after a 24-hour incubation of the cultures at temperature of 37 degrees centigrade); the oregano essential oil had been diluted in 1 ml of DMSO, according to the geometrical progression. A measure of the antifungal activity of preparation „fin Candimis” was the minimal inhibitory concentration (MIC), in terms of the fungus growth. Preparation „fin Candimis” is capable of being applied in the prevention and treatment of candidiasis – alone, or as a natural adjunctive agent. The C. albicans strains are more susceptible to preparation „fin Candimis” in comparison to the C. glabrata ones.
Celem pracy była ocena aktywności przeciwgrzybiczej preparatu „fin Candimis” (olejku eterycznego oregano) wobec szczepów grzybów z rodzaju Candida. Do badań wykorzystano 9 szczepów Candida albicans oraz 10 szczepów Candida glabrata, które wyizolowano z różnych materiałów klinicznych, i 1 szczep wzorcowy Candida albicans ATCC 90028 oraz preparat „fin Candimis”, zawierający olejek eteryczny otrzymany ze świeżych liści oregano. Według danych producenta, stężenie czystego olejku eterycznego oregano w produkcie „fin Candimis” wynosi 210 mg/ml. Wrażliwość szczepów z rodzaju Candida na preparat „fin Candimis” oznaczano metodą dyfuzyjno-krążkową na podłożu Sabouraud’a (po 24-godzinnej inkubacji hodowli w temperaturze 37°C); olejek eteryczny oregano rozcieńczano w 1 ml DMSO według postępu geometrycznego. Miarą przeciwgrzybiczego działania preparatu „fin Candimis” było najmniejsze stężenie hamujące wzrost grzyba (MIC). Preparat „fin Candimis” (olejek eteryczny oregano) może mieć zastosowanie w profilaktyce i leczeniu kandydoz – samodzielnie lub jako naturalny środek wspomagający. Szczepy Candida albicans są bardziej wrażliwe niż szczepy Candida glabrata na preparat „fin Candimis”.

In recent years, there has been a rapid increase in the incidence of Candida infections. The different species of the genus Candida vary in their virulence abilities and susceptibility to antifungal agents, depending on several external factors. The result of such modifications may be cross-resistance, which is understood as an acquired resistance to a certain antimicrobial agent after exposure to another drug. The aim of this study was to determine the possibility of cross-resistance between fluconazole, voriconazole, itraconazole, and methotrexate in Candida albicans and Meyerozyma guilliermondii (syn. Candida guilliermondii). Fifteen strains of M. guilliermondii and eight strains of C. albicans, including the standard strains, were tested. For all strains, the minimum inhibitory concentrations (MICs) for fluconazole, voriconazole, and itraconazole were determined before and after stimulation with methotrexate. The median MICs in M. guilliermondii before and after stimulation were 9.333 and 64 mg/L (p = 0.005) for fluconazole; 0.917 and 1.667 mg/L (p = 0.001) for itraconazole, respectively. No significant change in MIC was observed for voriconazole. For C. albicans strains, the median MICs before and after stimulation were 0.917 and 64 mg/L (p = 0.012) for fluconazole; 0.344 and 1.135 mg/L (p = 0.018) for voriconazole, respectively. There was no significant change in MIC values for itraconazole. Thus, this study demonstrates the presence of cross-resistance between voriconazole, itraconazole, fluconazole, and methotrexate for the selected strains. Methotrexate exposure induces different responses when certain drugs are used for various species. Therefore, if a patient was previously exposed to methotrexate, there may be a higher risk of treatment failure with fluconazole than with other azoles such as voriconazole for fungemia caused by M. guilliermondii or itraconazole for C. albicans infection.

Opis:
The paper presents results of geobotanical and taxonomic studies on the distribution and habitat requirements of Nymphaea candida in southern Poland. The researches were conducted in southern Poland in 2003-2009, in the provinces of Lower Silesia, Lublin, Małopolska, Opole, Silesian province as well as, in southern parts of Mazowieckie and Lubuskie. Flowers, leaves and fruits of Nymphaea species were collected from 27 locations. Altogether pollens from 73 populations of N. candida and 18 of N. alba from all the researched area were measured. The trophic level of an ecosystem was evaluated according to the results of the total nitrogen, total phosphorus, chlorophyll a, transparency and biological parameters. As the result of the studies of more than 200 water bodies, 57 localities of N. candida were documented within the investigation area. The populations of N. candida occupy mid-forest water bodies and river ox-bow lakes. A significant number of populations was also found in artificial reservoirs - fish ponds. The most suitable habitat conditions for N. candida occur in shallow waters in the shore zone with the amplitude of the water column vary from 0.5 to 2 m. Regarding the trophy level, N. candida occupies different habitats, mainly mesotrophic and also eutrophic with high content of organic matters. Considering the 15 checked morphological parameters, especially the stigma diameter, the number of carpellary teeth, flower and pollen diameters, the found and collected specimens of N. candida significantly differ from N. alba. The study confirms that N. candida ocurrs in whole lowland Poland without any regional distribution gaps. According to the IUCN guidelines to species assessment the data gathered during the presented study do not allow to classify N. candida as a vulnerable species in Poland. Still existing populations for more than 150 years, numerous stable locations, abundant populations, a habitat accessibility, a biotope extent, an ecological amplitude against the trophy level and direct human impacts suggest, that the species should be regarded as a least concern (LC) taxon.

Despite long-term research, the aquatic genus Nymphaea still possesses major taxonomie challenges. High phenotypic plasticity and possible interspecific hybridization often make it impossible to identify individual specimens. The main aim of this study was to assess phenotypic variation in Nymphaea taxa sampled over a wide area of Eastern Europe and temperate Asia. Samples were identified based on species-specific genome sizes and diagnostic morphological characters for each taxon were then selected. A total of 353 specimens from 32 populations in Poland, Russia and Ukraine were studied, with nine biometric traits being examined. Although some specimens morphologically matched N. xborealis (a hybrid between N. alba and N. Candida) according to published determination keys, only one hybrid individual was revealed based on genome size data. Other specimens with intermediate morphology possessed genome size corresponding to N. alba, N. Candida or N. tetragona. This indicates that natural hybridization between N. alba and N. Candida is not as frequent as previously suggested. Our results also revealed a considerably higher variation in the studied morphological traits (especially the quantitative ones) in N. alba and N. Candida than reported in the literature. A determination key for the investigated Nymphaea species is provided, based on taxonomically-informative morphological characters identified in our study.
