Stem cells exist and can do a lot. For several decades, bone marrow and umbilical cord blood transplants containing haematopoietic stem cells have been used in the treatment of blood diseases. Genetic modifications (gene therapy) of such cells help to cure complex immunodeficiencies and severe anaemias. The limbal stem cells taken from the eye and properly multiplied can regenerate the damaged cornea, and the epidermal stem cells help in the treatment of severe burns and some hereditary, severe skin diseases. Promising experimental research is under way on other uses of stem cells. However, these cells are appropriately selected, having real ability to differentiate into specialized cells whose malfunction is the cause of the disease. Therapeutic applications of stem cells are apparently limited to date. Meanwhile, the Internet is full of advertisements for supposedly miraculous treatments for almost any disease. Stem cells have become a modern synonym of the Holy Grail. A wonderful dish, transforming every drink into elixir of health, youth and long life. Stem cells from a single source, e.g., umbilical cord blood, or so-called cells, although without proven properties of stem cells, are offered in commercial private clinics as a panacea for autism, cerebral palsy, spina bifida, eye diseases, amyotrophic lateral sclerosis and dozens other disorders. Without justification for their action in these diseases, without convincing evidence of safety, but for a high fee. This article discusses stem cells and misunderstandings about including any cells among them. It draws attention to the real possibilities and confirmed uses of stem cells and presents the problems, doubts and dangers for patients associated with commercial offers of treatments using “stem” cells. The author cites the positions of scientific institutions and societies warning against premature commercialization of unjustified and potentially dangerous therapies

Aluminium recycling is cost-effective and beneficial for the environment. It is expected that this trend will continue in the future, and even will steadily increase. The consequence of the use of recycled materials is variable and difficult to predict chemical composition. This causes a significant reduction in the production process, since the properties of produced alloy are determined by the inicrostructure and the presence of precipitates of other phases. For this reason, the type and order of formation of precipitates were systematically investigated in recent decades. These studies involved, however, only the main systems (Al-Cu. Al-Mg-Si, Al-Cu-Mg, Al-Mg-Si-Cu), while more complex systems were not analysed. Even trace amounts of additional elements can significantly affect the alloy inicrostructure and composition of precipitates formed. This fact is particularly important in the case of new technologies such as laser surface treatment. As a result of extremely high temperature and temperature changes after the laser remelting large amount of precipitates are observed. Precipitates are nanometric in size and have different morphology and chemical composition. A full understanding of the processes that occur during the laser remelting requires their precise but also time effectively phase identification, which due to the diversity and nanometric size, is a major research challenge. This work presents the methodology of identification of nanometer phase precipitates in the alloy AlSi9Cu, based on the simultaneous TEM imaging and chemical composition analysis using the dispersion spectroscopy using the characteristic X-ray. Verification is performed by comparing the simulation unit cell of the identified phase with the experimental high-resolution image.

Opis:
Cellular therapy, as a part of regenerative medicine, implies to the treatment of human disorders with cells as a medical product, so called – “living drugs”. Usually such therapy is applied when other alternative efficient pharmacological therapies are not available. Stem cells of different origin: 1) tissue specific e.g. hematopethic, epithelial, neuronal, limbal; 2) mesenchymal stem cells (MSC) harvested from variety of tissues; 3) pluripotent stem cells: embryonic stem cells (ESC) and induced pluripotent stem cells (iPSC) – serve as a source of cells for regenerative medicine application, depending upon disease and application re- quirements. Currently MSC are the type of stem cells that are most frequently used in registered regenerative medicine clinical trials. In this paper we provide the information on the application of cell therapy in orthopedics, hematology, ophthalmology, dermatology, gastrology and neurology. The influence of origin of MSCs and iPSCs on their mode of action as therapeutic, regenerative agents are discussed. Advantages and disadvantages of application of different cell types for cell therapy are underlined. Last, but not least current low regulations in Poland and requirements of European regulatory bodies for cell therapy are pointed out and discussed.

The aim of this article of this paper is to present issues related to characterization of nanometric-sized carbides, nitrides and/or carbonitrides formed during tempering of carbide-steel cermets. Closer examination of those materials is important because of hardness growth of carbide-steel cermet after tempering. The results obtained during research show that the upswing of hardness is significantly higher than for high-speed steels. Another interesting fact is the displacement of secondary hardness effect observed for this material to a higher tempering temperature range. Determined influence of the atmosphere in the sintering process on precipitations formed during tempering of carbide-steel cermets. So far examination of carbidesteel cermet produced by powder injection moulding was carried out mainly in the scanning electron microscope. A proper description of nanosized particles is both important and difficult as achievements of nanoscience and nanotechnology confirm the significant influence of nanocrystalline particles on material properties even if its mass fraction is undetectable by standard methods. The following research studies have been carried out using transmission electron microscopy, mainly selected area electron diffraction and energy dispersive spectroscopy. The obtained results and computer simulations comparison were made.

Opis:
The development of innovative materials for clean energy systems and aeronautics requires use of modern research methods to characterize the structure on the level from micro- to nanoscale. Modern two-dimensional imaging techniques recently available in electron microscopes allow use of tomographic methods to characterize the structure of materials. Application of modern three dimensional imaging techniques such as electron tomography allows accurate qualitative and quantitative measurement of the structure elements in the micro- and nanoscale. The electron tomography studies have been carried out for three-dimensional visualization and metrology of different materials for clean energy systems and aeronautics. Electron tomography results provided quantitative data about shape, size and distribution of the particles, complementary to those obtained by means of quantitative TEM metallography.
Rozwój innowacyjnych materiałów dla niskoemisyjnych systemów energetycznych i lotnictwa wymaga zastosowania nowoczesnych metod badawczych w celu scharakteryzowania struktury na poziomie od mikro- do nanoskali. Nowoczesne dwuwymiarowe techniki obrazowania od niedawna dostępne w mikroskopach elektronowych, umożliwiają zastosowanie metod tomograficznych do scharakteryzowania struktury materiałów. Zastosowanie nowoczesnych technik trójwymiarowego obrazowania, takich jak tomografia elektronowa umożliwia dokładny jakościowy i ilościowy pomiar elementów mikrostruktury w mikro- i nanoskali. Badania metodami tomografii elektronowej przeprowadzone zostały w celu trójwymiarowej wizualizacji i metrologii elementów mikrostruktury róznych materiałów konstrukcyjnych dla systemów niskoemisyjnej energetyki i lotnictwa. Badania metodami tomografii elektronowej dostarczaja danych ilościowych dotyczacych kształtów, wielkości i rozkładu przestrzennego cząstek i są one uzupełnieniem do danych uzyskiwanych za pomocą metod ilościowej analizy metalograficznej przy wykorzystaniu transmisyjnej mikroskopii elektronowej.

In July 2013, symptoms of stem rot were observed in the Dracaena sanderiana cuttings in greenhouses of Mahallat County, Markazi Province, Iran. The symptoms first appeared as severe wilting. Later, leaves became brown and necrotic. Symptoms on the cuttings were observed as rotted areas on the middle of the stems. The cortical tissues of the plants showed a distinct brown discoloration. Eventually, the infected plants died. The pathogen was isolated from Dracaena stems and identified as F. solani by a fragment of the translation elongation factor 1-alpha (EF-1α) gene. Fusarium solani was confirmed by a pathogenicity test, and the causal agent was re-isolated from infected D. sanderiana plants. To the best of our knowledge, this is the first report of stem rot caused by F. solani on the cuttings of D. sanderiana.

This study was undertaken to investigate the effect of severe plastic deformation (SPD) by extrusion combined with reversible torsion (KoBo) method on microstructure and mechanical properties of Al-5Cu and Al-25Cu alloys. The extrusion combined with reversible torsion was carried out using reduction coefficient of λ = 30 and λ = 98. In this work, the microstructure was characterized by light microscopy (LM), scanning electron microscopy (SEM) and scanning transmission electron microscopy (STEM). Compression test and tensile test were performed for deformed alloys. The binary Al-5Cu and Al-25Cu alloys consist of the face cantered cubic (FCC) α phase in the form of dendrites and tetragonal (C16) θ-Al2Cu intermetallic phase observed in interdentritic regions. The increase of Cu content leads to increase of interdentritic regions. The microstructure of the alloys is refined after applying KoB deformation with λ = 30 and λ = 98. Ultimate Tensile Strength (UTS) of Al-5Cu alloy after KoBo deformation with λ = 30 and λ = 98 reached about 200 MPa. UTS for samples of Al-25Cu with λ = 30 and λ = 98 increased compared to Al-5Cu alloy and exceed 320 MPa and 270 MPa respectively. All samples showed increase of plasticity with increase of reduction coefficient. Independently of reduction coefficient, the compressive strain of Al-5Cu alloys is about 60%. The Al-25Cu alloy with λ = 98 showed the value of compressive strain exceed 60%, although for this same alloy but with λ = 30, the compressive strain is only 35%.

The samples of the CuCr0.6 alloy in the solution treated and additionally in aging states were severely plastically deformed by compression with oscillatory torsion (COT) method to produce ultrafine – grained structure. The samples were processed by using process parameters as: frequency of torsion (f = 1.6 Hz), compression speed (v = 0.04 mm/s), angle torsion (α = ±6°), height reduction (Δh = 7 mm). The total effective strain was εft = 40. The microstructure has been analyzed by scanning transmission electron microscope (STEM) Hitachi HD-2300A equipped with a cold field emission gun at an accelerating voltage of 200 kV. The quantitative microstructure investigations as disorientation angles were performed using a FEI INSPECT F scanning electron microscope (SEM) equipped with a cold field emission gun and a electron backscattering diffraction (EBSD) detector. The mechanical properties were determined using MST QTest/10 machine equipped with digital image correlation (DIC). The COT processed alloy previously aged at 500°C per 2h shows high mechanical strength, ultimate tensile strength UTS: 521 MPa and yield tensile strength YS: 488 MP attributed to the high density of coherent precipitates and ultrafine grained structure. The aim of this work was to investigate the microstructure and chemical composition of the transition zone between 16Mo3 steel and Inconel 625 weld overlay coating produced by the Cold Metal Transfer (CMT) method. Investigations were primarily carried out through transmission electron microscopy (TEM) on thin foils prepared by FIB (Focus Ion Beam). The chemical analysis demonstrated that the amount of certain elements (Fe, Ni, Cr, Mo, Nb) in the transition zone between the base material and the weld overlay changes quickly, from the composition of the steel to the composition of the composite zone. STEM and TEM investigations revealed that two areas are clearly visible in the transition zone. In the narrow band close to the fusion boundary where plates are clearly visible and the Ms temperature is higher than room temperature, electron diffraction analyses show reflections of martensite and austenite. Moreover, the crystallographic relations between martensite and austenite can be described by the Kurdjumov-Sachs (K-S) relationship {110}α’ || {111}γ <111>α’ || <110>y ). The microstructure of the part of the transition zone with an Ms temperature lower than room temperature as well as that of the composite zone is austenite. The investigations proved that the width of the martensitic area can be significantly limited by using the CMT technique for weld overlaying.

Opis:
Allvac 718Plus (718Plus) is a high strength, corrosion resistant nickel- based superalloy used for application in power generation, aeronautics and aerospace industry. The 718Plus microstructure consists of a γ matrix with γ’-Ni3(Al,Ti) and some δ- Ni3Nb phases as well as lamellar particles (η-Ni3Ti, η*-Ni6AlNb or Ni6(Al,Ti)Nb) precipitated at the grain boundaries. The primary strengthening mechanism for this alloy is a precipitation hardening, therefore size and distribution of precipitates are critical for the performance of the alloy. The aim of this study was to characterize precipitates in the 718Plus superalloy using Scanning Transmission Electron Microscope combined with Energy Dispersive X-ray Spectroscopy (STEM-EDX) and Focused Ion Beam Scanning Electron Microscope (FIB-SEM). The STEM-EDX and FIB-SEM tomography techniques were used for 3D imaging and metrology of the precipitates. Transmission electron microscopy and EDX spectroscopy were used to reveal details of the 718Plus microstructure and allow determine chemical composition of the phases. The study showed that electron tomography techniques permit to obtain complementary information about microstructural features (precipitates size, shape and their 3D distribution) in the reconstructed volume with comparison to conventional particle analysis methods, e.g. quantitative TEM and SEM metallography.
