Deoxyribonucleic acid (DNA) was identified 140 years ago by a Swiss physician Friedrich Miescher. His discovery was fundamental for the development of biochemistry, genetics and molecular biology. Contemporary biology, biotechnology and medicine largely depends on our ability to analyze, synthesize and manipulate DNA. We present highlights of the history of DNA research from the very beginning to the sequencing of human genome.

Opis:
Cytogenetics is complementary to genetic and molecular analysis of genome structure and function. From the beginning it has been mainly used for identification of chromosomes and karyotype construction. Most significant for the progress in cytogenetics was development of chromosome banding techniques and in situ hybridization, especially fluorescent in situ hybridization (FISH) and its different modifications which have become the most important techniques in molecular cytogenetics. FISH allows physical gene mapping and localization of different non-coding DNA sequences on chromosomes and interphase nuclei. Repetitive DNA sequences can generate unique FISH-signal patterns on individual chromosomes valuable for karyotyping and phylogenetic analysis. These studies have important implications for basic research and practical applications. The understanding of the structure, function, organization and evolution of genomes enabled many new cytogenetic applications to both medicine and agriculture, particularly in diagnosis and plant breeding.

Piotr Słonimski, full professor of genetics at University Paris 6 and associate professor on Institute of Biochemistry and Biophysics P.A.S. Born in Warsaw 9.11.1922 died in Paris 25.04.2009. He was full member of French Academy of Science and foreign member of Polish Academy of Sciences, Polish Academy of Arts and Sciences as well as of several others European Academies and dr. hc of many European Universities. He was honored by number of scientific prizes, and was holder of several high decorations, French and Polish. Piotr Słonimski studied medicine during the II World War in Warsaw Underground University, and started after the war his scientific career getting in 1946 his M.D. title at Jagiellonian University in Cracow. Being under occupation the active member of Polish underground and soldier of Warsaw 44 insurrection against Nazis, in Poland incorporated into soviet block he did not escaped the repressions falling on the anti-nazi independence fighters and was arrested by communist secret services. After a few months in prison he was liberated and left Poland for France, were in laboratory of Boris Ephrussi he started his career in CNRS, where from 1971 to 1992 was Director of Centre de Genetique Moleculaire. In line with solid french tradition yeast S. cerevisiae was preferred model of his studies. Here he demonstrated non-standard heritage of mitochondrial traits and become one of co-founders of non-mendelian genetics in Eucatyota. By subtle genetic analysis of selected mitochondrial genes he was first to demonstrate the existence of mosaic genes and pointed towards the regulatory function of information carried on by introns. In nineties Piotr Słonimski was a spiritus movens of European yeast genomics program, acknowledged by special issue of Nature, giving full gene repertoire of the first eucaryotic organism. His scientific interest stayed then in genomes. Here with advanced cryptology methods he was looking for the existence of non-trivial rhythms in coding DNA sequences. His death left in sorrow not only his family but also all the scientist of “yeast community” as well as his comrades in arms from underground and Solidarity movement.

Opis:
François Chapeville (Franciszek Chrapkiewicz), an outstanding Polish bioche- mist, spent his entire adult life in France. After graduating in veterinary and biochemistry there, he completed a postdoctoral fellowship at the Fritz Lipmann laboratory at Rockefeller University in New York. There, he conducted studies that proved that the genetic information encoded in DNA is deciphered in the process of protein biosynthesis via adapter tRNA molecules (adapter hypothesis). In the years 1979–1991 he was appointed director of Jacques Monod Institute of Molecular Biology at the University VII in Paris. He was a great promoter of Polish-French cooperation in the natural sciences. He passed away in Paris at the age of 96. He was buried in his hometown of Strzyżów in the Podkarpacie region.

Opis:
Several aspects of in vitro culture have potential for cereal improvement. This paper focuses on evaluation of somaclonal variation (SV) from immature embryo callus culture, and doubled haploid (DH) production via anther culture in barley. Genetically stable SV was observed for several seedling morphological traits such as albino, yellow, light green and lethal. SV occurred at approximately half the frequency of azide-induced mutagenesis. The potential for widespread application of anther culture-mediated DH production in barley breeding and genetic studies was increased through culture procedure improvements and understanding the inheritance of anther culture response. Methodology improvements included substitution of inexpensive gelrite for expensive ficoll or agarose, ability to grow anther donor plants under field as well as growth chamber conditions and flexibility in cold pretreatment/storage of anther donor spikes for 4-6 weeks prior to anther plating. From diallel analysis, inheritance of anther culture response was complex with additive and dominance effects for embryoid formation, total plant regeneration and green plant regeneration and reciprocal effects (maternal) for green plant regeneration. High x low responder crosses generated F₁’s that were intermediate in response and low x low crosses sometimes produced F₁ heterosis for green plant regeneration. Therefore, some recalcitrant types appear to be usable in anther culture DH production systems within a breeding program.
