Opis:
Zinc-binding proteins from seminal plasma (ZnBPs) originate in the secretions of different accessory sex glands and are implicated in key events associated with sperm-egg fertilization processes. This study describes the isolation and characterization of the ZnBPs of canine seminal plasma. Ejaculates were collected from three crossbred dogs for a 2-week period. The ZnBPs as well as non zinc-binding proteins (nZnBPs) were isolated by zinc-dependent affinity chromatography. The isolated fractions were subjected to native gel electrophoresis (one-dimensional polyacryamide gel electrophoresis, PAGE) and sodium dodecyl sulphate polyacryamide gel electrophoresis (SDS-PAGE), using denaturing and reducing conditions. Zinc-elution profile using affinity chromatography displayed two protein fractions represented by the nZnBPs and ZnBPs, respectively. Using native gel electrophoresis, it was found that both the nZnBPs and ZnBPs occurred in their native state as aggregates, ranging from 140 to 669 kDa. The nZnBPs were disaggregated into 8 protein bands, with molecular weights ranging from 10.7 to 79.7 kDa, following SDS-PAGE analysis. By contrast, SDS-PAGE analysis of the ZnBPs revealed 13 protein bands, with molecular weights ranging from 11.6 to 152.3 kDa. Densitometric analysis showed that 46-48% of nZnBPs could be accounted by protein fractions with molecular weights of 10.7 and 14.2 kDa. Also, 2 protein fractions with molecular weights of 11.6 and 14.3 kDa, were predominant in ZnBPs, accounting for approximately 28-30% of the total proteins. These results demonstrate the zinc-binding capacity of proteins secreted by the canine prostate. The findings of this study indicate that ZnBPs of canine seminal plasma comprise several protein fractions, which might be implicated in the reproductive processes in the dog.

Opis:
Thirty nine canine S. pseudintermedius strains were examined for antibiotic susceptibility and genetic polymorphisms. All strains were methicillin-sensitive S. pseudintermedius (MSSP). Resistance to penicillin was most prevalent (66.6%), followed by resistance to neomycin (56.4%), erythromycin (53.8%), clindamycin (48.7%), chloramphenicol (48.7%), and tetracycline (46.2%). Pulsed-field electrophoresis (PFGE) showed a high genetic polymorphism in the investigated strains.

In the paper the detection of the SSCP polymorphism within the 5’ fragment of bovine beta-lactoglobulin (LGB) gene is described. The 5’ fragment of LGB gene (209 bp) was PCR-amplified and then subjected to electrophoresis allowing the detection of SSCP polymorphism. Among 124 animals (50 cows and 74 bulls) six SSCP patterns were identified and named Rl, R2, R3, R4, R5 and R6, which occured with the frequency of 0.32, 0.51, 0.09, 0.06, 0.01 and 0.01, respectively. The PCR-SSCP method is simple, fast, and relatively inexpensive. The SSCP polymorphism reported in the paper may be useful in looking for the associations between different SSCP patterns and LGB gene expression and milk properties.

Opis:
To saturate barley (Hordeum vulgare L.) genetic maps the linkage relationships of two isoenzyme loci Amp2 (aminopeptydase) and Aat2 (aspartate aminotransferase) with known genetic markers were investigated. Results of the genetic analysis support previous information on the localization of these loci on chromosome 1 and 6, respectively. The following recombination values were estimated: between locus Amp2 and T1-3b translocation break point 13.8 ± 2.1%, between locus Aat2 and translocation T6-7i, 17 ± 3.0% and between locus Aat2 and marker о 24.1 ± 3.0%.

Opis:
The aim of the study was the electrophoretical identification of molecular forms of selected antioxidant enzymes in dog semen. Ejaculates to be studied were chosen from five dogs, aged from two to eight years. Polyacrylamide gel electrophoresis was carried out under non-denaturing conditions and then gels were stained for the activity of the following enzymes: superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Sperm homogenates and all fractions (pre-spermatic, spermatic and post-spermatic) of dog ejaculate demonstrated one protein band with SOD activity characterized by low electrophoretic mobility. Based on the confirmed sensitivity to H₂O₂, it can be assumed that the detected SOD is an enzyme containing ions of Zn²⁺ and Cu²⁺ (Cu,Zn SOD). In sperm homogenates one protein band with GPx activity was characterized by high electrophoretic mobility, whereas in the spermatic and post-spermatic fractions of dog ejaculate three protein bands with different (low, medium and high) electrophoretic mobility were identified. CAT molecular forms were not found in either sperm homogenates or in the analyzed fractions of ejaculate.

Opis:
α-Amylase isozymes from rye endosperm were analysed by means of isoelectric focusing, polyacrylamide gel electrophoresis, immunoelectrophoresis and colorimetric assay. Α-AMY 1 (high pI) group was separated into 13 IEF bands, whereas in group α-AMY2 (low pI) 2 intensive and 6-8 faint bands were found. Two linked (2±1 cM) polymorphic loci and a single locus with two alleles encoding for α-AMY 1 and α-AMY 2 groups, respectively, were identified after genetic analysis of the IEF patterns. All α-amylase isozymes developed on PAGE, were shown to belong to α-AMY 1 group. It was demonstrated that a single PAGE isozyme corresponds to 2-4 separate IEF bands and that most of the IEF bands can be attributed to more than one PAGE isozyme. The activity of α-amylases from PAGE zone I was 2.3 times higher than the activity of zone II isozymes. A strong correlation between the activity and protein amount of particular α-AMY 1 isozymes (r=0.94) was found.

Opis:
This work presents some properties of Sunn-hemp mosaic tobamovirus (SHMV) orginally isolated from bean plants. Virus infected host range and induced symptoms that were typical for SHMV. Following plant species distinquished SHMV from tobacco mosaic tobamovirus (TMV): Phaseolus vulgaris, Pisum sativum, Lupinus albus and Lycopersicon esculentum. In immunoblotting the serum against SHMV did not react with TMV and Tomato mosaic tobamovirus (ToMV). The electrophoretical patterns of whole virions and capsid proteins were characteristic for SHMV and different from that of TMV and ToMV.
W pracy przedstawiono charakterystykę wirusa mozaiki krotalarii (SHMV) wyizolowanego z siewek fasoli wyrosłych z zainfekowanych nasion. Stwierdzono, że zakres roślin gospodarzy badanego izolatu SHMV oraz wywoływane przez niego objawy chorobowe były typowe dla tego wirusa. Gatunki roślin takie jak: Phaseolus vulgaris, Pisum sativum, Lupinus albus i Lycopersicon esculentum różnicują SHMV od TMV. W teście immunoblotingu surowica przeciwko SHMV nie reagowała z TMV-U i ToMV-2., z kolei surowica przeciwko TMV reagowała jedynie z TMV-U₁ i ToMV-2. Obrazy elektroforetyczne zarówno całych virionów jak i białek otoczki wirusowej były charakterystyczne dla SHMV i różne od tych dla TMV-U₁ oraz ToMV-2.
