Opis:
An optical tomograph in which a tested object is illuminated from five directions has been presented in the paper. The measurements of luminous intensity after changing into discrete signals (0 or 1) in the detectors equipped with 64 optical sensors were subjected to reconstruction by means of the matrix algorithm. Detailed description of the measuring sensor, as well as the principles of operation of the electronic system, has been given in the paper. Optical phenomena occurring at the phase boundary while transmitted through the sensor wall and phenomena inside the measuring space have also been taken into account. The method of the sensor calibration has been analysed and a way of technical solution of the problem under consideration has been discussed. The elaborated method has been tested using objects of the known shape and dimensions. It was found that reconstruction of the shapes of moving bubbles and determination of their main parameters is also possible with a reasonable accuracy.

In this study, mycelial compatibility grouping (MCG) of Sclerotinia sclerotiorum (Lib.) de Bary was determined in 15 infected potato fields in Bahar and Lelehjin, Hamadan, Iran. Among 193 isolates, 37 MCGs were identified and 67% were represented by single isolates observed at single locations. Within Bahar fields, 29 MCGs were identified and 15 MCGs were recognized in Lalehjin samples. MCGs 2, 6, 18 and 25 were collected at high frequency from multiple locations. The efficacy of Potato Dextrose Agar (PDA) amended with activated charcoal for identifying MCGs in this fungus was tested in doubtful reactions and proved to be an effective medium. The activated charcoal produced a black reaction area in incompatible interactions as well as a red reaction line in PDA amended with McCormick's red food coloring.

Opis:
A temperature dependent model is necessary for the generation of hysteresis loops of ferromagnetic materials. In this study, a physical model based on the Jiles-Atherton model has been developed to study the effect of temperature on the magnetic hysteresis loop. The thermal effects were included through a model of behavior depending on the temperature parameters Ms and k of the Jiles-Atherton model. The temperaturedependent Jiles-Atherton model was validated through measurements made on ferrite material (3F3). The results have been found to be in good agreement with the model.

The efficacy of three types of mineral spray oil (MSO), and Abamectin plus MSO to control the citrus leafminer Phyllocnistis citrella Stainton were compared with the conventional broad spectrum pesticides, Confidor, and Dursban, in two citrus orchards in northern Iran. Differences were found among the various concentrations of MSO in the number of mines and live larva per leaf, sampled 5 days after the application of 3 sprays at an interval of 5 days. Mineral spray oil applied at a rate of ≥ 0.65% in water, showed no significant differences when compared with Confidor, and Dursban. Abamectin plus MSO at a rate of 0.02% plus 0.50% in water respectively, provided the highest level of control and reduced the population of larva up to 85%. Generally, a concentration of ≥ 0.65% MSO in water caused a significant damage reduction. However, by reducing the MSO concentration, the rate of control was decreased. In another field experiment conducted in mature citrus orchards, the effect of two pest management types (applying just MSO vs. usage of synthetic pesticides) on the activity of P. citrella, was evaluated. The comparison results indicated that there is more P. citrella damage in orchards under pressure of synthetic pesticides than in orchards in which the synthetic pesticides were eliminated for years.

Arbuscular-mycorrhizal internal structures (i.e. total root colonization, arbuscules, vesicles) and external structures (i.e. spore density), and Glomeromycota spore morphotypes, were evaluated in wheat severely infected with Mycosphaerella graminicola - the causal agent of Septoria leaf blotch. Plots in which the infection was controlled with a commercial fungicide at recommended field doses, were also examined. The commercial fungicide used was an admixture of trifloxistrobin and tebuconazole. No negative effects of the fungicide application on arbuscular-mycorrhizal fungi (AMF) were found. The M. graminicola fungicidal treatment actually favoured the formation of arbuscules and AMF spores, as there was a selective increase in the density of spores belonging to the glomoid morphotype. Arbuscular-mycorrhizal fungi have an absolute dependence on the carbon provided by the plant. A severe foliar disease leading to a diminished carbon supply to the roots would generate decreases in carbon availability. Such decreases would strongly affect mycorrhizal associations and development. Furthermore, the change in the green-leaf area produced by a severe foliar disease and/or a reversal of that condition through fungicide treatment could result in shifts in the composition of the AMF community so as to favour glomoid morphotypes. Glomoid species have been previously considered as r-strategists.

Opis:
Human growth hormone (somatropin) is one of the most widely used recombinant proteins that stimulates growth, cell reproduction, and cell regulation in humans. Synthetic production of this protein normally results in low yields and inclusion body formation. To overcome these difficulties, the production of somatropin along with two common signal peptides, namely TorA and SufI, in co-expression with a cytosolic chaperone, GroEL/ES, was evaluated in the present study. The target protein and the two signal sequences (TorA and SufI) were synthesized and cloned into an expression plasmid (pET-22) by using Nde l and Xho l endonucleases. The expression vector (pGro7) containing chaperone proteins (GroES/EL) and one of the expression vectors containing the signal sequence (and the target protein) were co-expressed in the BL21 DE3 expression host. The results showed that although some of the expressed proteins exit the cytoplasm and enter the periplasmic space, there is also an accumulation of proteins (probably as inclusion body) inside the cytoplasmic area. Western blot analysis showed that the inclusion of a signal sequence in the cassette containing the target protein could help to secrete the protein in the periplasmic space and culture media when compared with control groups. The result of these experiments show that the TAT secretion system promotes transportation of the target protein out of the cytoplasm. This secretory system completes folding of the protein structure and transfers the mature protein to the periplasmic space.

Bovine Rotavirus and Bovine Coronavirus are the most important causes of diarrhea in newborn calves and in some other species such as pigs and sheep. VP8 subunit of rotavirus is the major determinant of the viral infectivity and neutralization. Spike glycoprotein of coronavirus is responsible for induction of neutralizing antibody response. Studies showed that immunoglobulin of egg yolk (IgY) from immunized hens has been identified to be a convenient source for specific antibodies for using in immunotherapy and immunodiagnostic to limit the infections. In this study, chimeric VP8-S2 gene was designed using by computational techniques. The chimeric VP8-S2 gene was cloned and sub-cloned into pGH and pET32a (+) vectors. Then, recombinant pET32a-VP8-S2 vector was transferred into E. coli BL21 CodonPlus (DE3). The expressed protein was purified by Ni-NTA chromatography column. Hens were immunized with the purified VP8-S2 protein three times. IgY was purified from egg yolks using polyethylene glycol precipitation method. Activity and specificity of anti-VP8-S2 IgY were detected by dot-blotting, Western-blotting and indirect ELISA. We obtained anti-VP8-S2 IgY by immunizing hens with the recombinant VP8-S2 protein. The anti-VP8-S2 IgY was showed to bind specifically to the chimeric VP8-S2 protein by dot-blotting, Western-blotting analyses and indirect ELISA. The result of this study indicated that such construction can be useful to investigate as candidates for development of detection methods for simultaneous diagnosis of both infections. Specific IgY against the recombinant VP8-S2 could be recommended as a candidate for passive immunization against bovine rotavirus and bovine coronavirus.

The role of the tea commodity in the economy of Indonesia is quite strategic. Various types of microorganisms in nature have been known to increase the benefit of the root function, suppress disease, and accelerate plant growth. This study aimed to determine the potential of indigenous bacteria (Azoto II-1, Acinetobacter sp., bacteria Endo-5, bacteria Endo-65 and Endo-76) on the growth of tea plants and their potential in increasing resistance to blister blight disease. The test of microbes’ potential effect on growth and blister blight was conducted in Gambung, West Java in an experimental field using a randomized block design (RBD) with six treatments and each treatment was replicated four times. The composition of the treatments was: A) Endo-5; B) Endo-65; C) Endo-76; D) Azoto II-1; E) Acinetobacter sp.; and F) control (without microbes). Bacterial suspension was applied directly to the soil at a dose of 2 l · ha−1. The bacterial suspension was applied six times at 1 week intervals. The results of field observations indicated that the intensity of blister blight decreased in all treatments but did not significantly differ from the control. Meanwhile, the results of Acinetobacter sp. treatment in tea shoots was 17.26% higher than the control.

The granary weevil, Sitophilus granarius (L.), is one of the most important internal feeders of stored grain. Nanotechnology has become one of the most promising new approaches for pest control in recent years. In our screening program, laboratory trials were conducted to determine the effectiveness of silica nanoparticles (SNPs) and zinc nanoparticles (ZNPs) against the larval stage and adults of S. granarius on stored wheat. Nanoparticles of silica and zinc were synthesized through a solvothermal method. They were then used to prepare insecticidal solutions of different concentrations and tested on S. granarius. Silica nanoparticles (SNPs) were found to be highly effective against S. granarius causing 100% mortality after 2 weeks. ZNPs were moderately effective against this pest.

To formulate an efficient and eco-friendly strategy for the management of cotton seedling disease complex, pot experiments were conducted and the efficiency of eight Bacillus strains against seven fungi involved in the disease were determined. A greenhouse evaluation of the interaction between fungal isolates and Bacillus strains was carried out. The evaluation revealed a very highly significant Bacillus strains x fungal isolates interaction for all the following parameters: preemergence damping-off, postemergence damping-off, survival, plant height, and dry weight. This interaction implies that a single strain of the Bacillus sp. can be highly effective against a fungal isolate, but may have only minimal effects on other fungal isolates. The results of the present study demonstrated that Bacillus circulans and B. coagulans were the most effective strains in controlling cotton seedling disease. Therefore, strains of Bacillus spp. should be tested against as many fungal isolates as possible. The testing will improve the chance of identifying Bacillus strains effective against several fungal isolates.
