Opis:
A PR10 gene (ThPR10) was cloned from Tamarix hispida and characterized. Real-time RT-PCR was employed to evaluate gene expression levels. ThPR10 was expressed in both leaves and roots of T. hispida under normal growth conditions, and can be highly induced in both leaf and root tissues by abiotic stresses including NaCl, PEG, cold, CdCl2, and ABA (abscisic acid) treatments. Our results indicated that ThPR10 is involved in the abiotic stress response, and regulated by an ABA-dependent signaling pathway. Subsequently, ThPR10 was localized at the subcellular level. The gene was fused with the GFP N-terminal driven by CaMV35S promoter and transiently expressed in onion epidermal cells. This strategy localized the ThPR10 protein to the nucleus of onion epidermal cells, suggesting that the pathogenesis-related proteins play a functional role in the cell nucleus.

Opis:
The friction and wear properties of 201HT aluminum alloys and the corresponding competitive coupons were tested on an electro-hydraulic servo face friction and wear testing machine (MM-U10G). The microstructures of the competitive coupons were investigated by scanning electron microscopy (SEM) and consequently the corresponding friction and wear mechanisms were studied. The results demonstrated that: (1) the best competitive material of friction and wear performance of the 201HT was the 201HTC. (2) the 201HTC modified by carbon following the initial mill for oil storage of the micro-groove to be produced, increased the corresponding lubrication performance reduced the friction coefficient and wear rate effectively. (3) the 201HT-201HTC could obtain both better friction and wear mainly due to the initial process of grinding following the 201HT plastic deformation occurred in the surface and the formation of a series of re-melting welding points, whereas the 201HT material hardness would be similar to the 201HTC material hardness, which led into the competitive material friction and wear performance improvement.

Opis:
Generally, gross errors exist in observations, and they affect the accuracy of results. We review methods to detect the gross errors by Robust estimation method based on L1-estimation theory and their validity in adjustment of geodetic networks with different condition. In order to detect the gross errors, we transform the weight of accidental model into equivalent one using not standardized residual but residual of observation, and apply this method to adjustment computation of triangulation network, traverse network, satellite geodetic network and so on. In triangulation network, we use a method of transforming into equivalent weight by residual and detect gross error in parameter adjustment without and with condition. The result from proposed method is compared with the one from using standardized residual as equivalent weight. In traverse network, we decide the weight by Helmert variance component estimation, and then detect gross errors and compare by the same way with triangulation network In satellite geodetic network in which observations are correlated, we detect gross errors transforming into equivalent correlation matrix by residual and variance inflation factor and the result is also compared with the result from using standardized residual. The results of detection are shown that it is more convenient and effective to detect gross errors by residual in geodetic network adjustment of various forms than detection by standardized residual.

Opis:
Toxoplasma gondii is a parasite that has been extensively studied due to its medical and veterinary importance in terminating pregnancies. Consequently, a satisfactory vaccine is required to control its adverse effects on pregnant animals. The microneme protein, MIC3, is a major adhesion protein that binds to the surface of host cells and parasites, and is therefore a potential vaccine against T. gondii. The viability of MIC3 as a vaccine is investigated in this study. Sheep were injected twice, intramuscularly, with plasmids containing DNA encoding for the mature form of MIC3 protein formulated into liposomes. Control sheep were injected with an empty vector or received no injections. The injection of sheep with DNA plasmids encoding for MIC3 elicited an immune response after the first and second injections as indicated by antibody responses and the production of IFN-γ. The immune response, as measured by the IgG2 and IgG1 serum levels, was boosted after the injection of the MIC3 DNA vaccine together with high anti-MIC3 antibodies. The results demonstrate that the intramuscular injection of sheep with a plasmid containing DNA coding for MIC3 protein induces a significant and effective immune response against T. gondii.
