The study attempted to analyse and characterize bacteria of the genus Staphylococcus isolated from the surface and contents of quail eggs, taking into account their phenotypic properties, biochemical reactions, antibiotic sensitivity patterns, and PCR to test for the presence of the mecA gene, which is responsible for resistance to methicillin. The study included 45 strains of the genus Staphylococcus isolated from the whites, yolks and shells of table quail eggs. The results obtained indicate that a fairly high percentage of the retail quail eggs tested were contaminated with Staphylococcus bacteria. Among the species isolated (11 in total), the most frequently occurring strains were of Staphylococcus hominis (26.7%), followed by Staphylococcus aureus (15.6%), S.xylosus and S. lentus (13.3% each), while percentages of the other Staphylococcus species were under 10%. The Staphylococcus strains tested had highly differentiated biochemical and enzymatic properties. As many as 7 biotypes were distinguished among the 7 S. aureus isolates, 6 biotypes within the species S. xylosus (6 strains tested), 5 biotypes among the 6 strains of S. lentus, but only 4 biotypes among the 12 S. hominis strains. The antibiotic sensitivity testing showed 15.5% of the strains to be resistant to one or more of the therapeutic agents tested. Moreover, some isolates exhibited intermediate sensitivity to the drugs, particularly to gentamicin (24.4%), neomycin (31.1%), streptomycin (46.7%) and Linco-Spectin (48.9%).

Opis:
Genistein (GEN), an isoflavonoid phytoestrogen, is one of the potent estrogenic compounds derived from plants that can cause disrupting effects on sex organ development in non-mammalian and mammalian species. The present study revealed effect of genistein on germ cell number in the genital ridges during gonadogenesis. Genistein (16 and 24 μg/g egg) was injected into the egg yolk prior to incubation. Effect of genistein on quail-primordial germ cells (PGCs) number was examined by counting the number of Wisteria floribunda (WFA)-positive cells localized in both left and right genital ridges compared with the control group. Both concentrations of genistein resulted in significant decrease of PGC number compared with the control group. Percentages of the sterility rate of the embryo treated with 16 and 24 μg of genistein/g egg were 19% and 23%, respectively. These results provide evidence that genistein may be a germ cell toxicant causing sterility later in life of adult birds. This is the first report on the effect of genistein on PGC number in the genital ridges of the avian embryo.

Coccidiosis is the most predominant parasitic disease affecting Japanese quails (Coturnix coturnix japonica) in commercial farms. Coccidiosis as a subclinical infection is difficult to diagnose without parasitological examinations. Oocysts of two Eimeria species, E. bateri and E. tsunodai, were determined in the analysed quail flock. Infected birds were administered Baycox 2.5% at the dose of: group I - 7 mg toltrazuril/kg BW per day provided in drinking water (1.5 ml/0.5 l H₂O) that was available 24 h for 2 days, group II - 14 mg/kg BW (3 ml/0.5 l H₂O), and group III - 24.5 mg/kg BW (5 ml/0.5 l H₂O); in groups II and III, the solutions were available 8 h/24 h for 2 days. After the first day of the treatment, the number of excreted oocysts (OPG - oocysts per gram) increased, a steady decrease in oocyst counts began on the second day of Baycox administration and lasted until a three-day period when no oocyst were determined in faecal samples. Regardless of the dose applied, toltrazuril (Baycox) completely eliminated E. bateri coccidia and led to a highly significant reduction in the number of E. tsunodai oocysts. The results suggest that the effectiveness of toltrazuril varies depending on coccidia species and developmental stages of the parasite. From the clinical point of view, the treatment applied significantly reduces the number of coccidia oocysts in commercial flocks of Japanese quails.

The activity of aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase, total protein, albumin and cholesterol levels were determined in the blood serum of Japanese quails infected with coccidia and treated with Baycox (active ingredient: toltrazuril). Lower levels of AST and ALT activity were noted in treated birds regardless of the applied Baycox dose. The biochemical changes observed in the blood serum of Japanese quails point to coccidia-induced damage of digestive system tissues despite an absence of pronounced clinical symptoms. Significantly lower levels of AST activity and higher levels of LDH activity in treated birds indicate that coccidiosis treatment with toltrazuril contributed to the regeneration of digestive system tissues. An insignificant increase in cholesterol levels was noted, whereas the other serum biochemical parameters remained within the reference ranges.

Anther and somatic tissue culture in combination with mutagenesis were carried out to evaluate the efficiency of different mutagenic treatments of various in vitro culture materials, and to obtain some promising variants for rice improvement. Results indicated that in japonica rice radiation treatment of dry seeds and young panicles influenced the percentage of green plantlets regeneration from anther culture. Both treatments increased significantly the percentage of regenerated green plantlets in comparison with the control. Irradiation with 30 Gy of rice callus increased also the percentage of regenerated green plantlets. For indica rice, the combination of the suitable dose of gamma rays irradiation on seeds and an improved medium, increased the percentage of callus induction. This approach made it possible to use anther culture in indica rice breeding. Somatic tissue cultures combined with radiation-induced mutagenesis led to the development of a number of promising mutants including some new cytoplasm-nucleus interacting male-sterile lines with almost 100% stigma exsertion. Their development would be of practical significance for increasing the genetic diversity for production of hybrid rice.

Tolerance to a new herbicide, pyributycarb, was evaluated both at the plant and cellular levels. Several highly or moderately tolerant strains chosen at the plant level, showed a parallel relation of to tolerance at the cellular level. However, on the whole, correlation between total tolerance indices and survival rates of calli was not significant in 18 out of the 80 studied strains. As a result of somaclonal selection for two herbicides, lines NB-200 and NM-100 were regenerated from the tolerant calli screened with benthiocarb at 200 ppm, and molinate at 100 ppm, respectively. In the R₂ generation, both the lines displaned a stable tolerance both at the plant and cellular levels. Thus the highly tolerant mutant lines were developed from a moderately tolerant line, N-61, via in vitro selection. To achieve a short-cut method in the interspecific genetic exchange, a series of techniques related to cell fusion were established in rice and related species. Two kinds of somatic hybrids between the cultivar Kitaake and tetraploid Oryza species, O. punctata and O. officinalis, were successfully produced. Among the somatic hybrid plants, a wide range of chromosomal variation was observed. Aneuploid plants with a chromosome number around 2n = 72 (hexaploid), which are expected from a symmetric fusion between diploid and tetraploid strains, were obtained showing mixoploidy within a plant. Most of the somatic hybrids were characterized by intermediate features of plant-type showing high sterility, shattering of spikelets and reduced plant height. As an exception, a diploid plant, which was identified by RFLP analysis using the rDNA gene probe, closely resembled Kitaake and produced viable seeds. A tetraploid hybrid plant was also promising for the introduction of economically important characters through the reduction of chromosome numbers by doubled haploids. Gametoclonal variation and gamma radiation was applied to Kitaake. The mutation frequency was prominently increased by gamma ray treatment, especially at high doses of 200 Gy or 300 Gy. In the M₃R₂ or M₄R₃ generations, most of the variants showed unfavourable characters. Most of the mutant characters were governed by single or double recessive genes. Several mutants such as short culm and early flowering time might be used for rice breeding.

The green plant bug (Apolygus lucorum Meyer-Dür) is a key pest of Bt cotton in China. Along with biological control, chemical control is one of the most important strategies in A. lucorum Integrated Pest Management (IPM). The goal of this study was to evaluate the toxicity of eight conventional insecticides to A. lucorum and to assess the susceptibility of two generalist predators Chrysopa sinica (Jieder) and Propylaea japonica (Thunbery) to insecticides that are commonly used in A. lucorum management. Via glass-vial and leaf-dip bioassay, toxicity tests with selected insecticides at two different life-stages of A. lucorum indicated significant differences between the LD50 or LC50 values for these compounds within different insecticidal classes. Phenylpyrazole fipronil had the highest toxicity to 4th-instar nymphs and adults of A. lucorum, whereas neonicotinoid imidacloprid had the lowest toxicity among the insecticides. Females were more tolerant to insecticides than were males, as shown by the higher LD50 values for females. Furthermore, laboratory tests showed that endosulfan had the highest selectivity to C. sinica and P. japonica: the selective toxicity ratios (STRs) were superior to other tested insecticides, particularly imidacloprid, and were 5.396 and 4.749-fold higher than baseline STRs, respectively. From this study, we conclude that fipronil can potentially be used to efficiently control A. lucorum. An alternative control agent worth consideration is endosulfan, owing to its relative safety to non-targeted natural enemies.
