Seed-borne diseases of wheat such as Fusarium head blight (FHB), a fungal disease caused by several species of Fusarium, results in reduced yield and seed quality. The aim of this study was to identify the Fusarium species, the effect of Fusarium-infected seeds on germination and vigor indices and to determine the location of Fusarium spp. in seeds, as well as to investigate the pathogenicity and variability of aggressiveness of the isolates obtained from pre-basic seeds wheat fields in Iran. According to morphological and molecular characters, the species F. graminearum, F. culmorum, F. avenaceum and F. poae were identified. Among the isolates, F. graminearum was the predominant species with the highest frequency and relative density of 92.9% and 70.9%, respectively. We observed that germination and vigor indices were decreased due to increased Fusarium-infected seeds. Results indicated significant differences among cultivars and seed-borne Fusarium levels. While a higher infection level of Fusarium spp. most commonly occurred in the seed coat, only F. graminearum was observed in embryos. Our study about pathogenicity showed that 77.3% of the Fusarium spp. isolates were not pathogenic and 22.7% isolates of Fusarium spp. were pathogenic or weakly pathogenic. Our results indicated that variability in aggressiveness among isolates of a species and positive correlation may be determined by pathogenicity tests. This is the first time the location of Fusarium spp. in seeds has been identified. It is also the first time that Fusarium-infected seeds in pre-basic seeds wheat fields of Iran have been evaluated.

We used germination tests to assess the frequency of polyembryony in 9 asparagus cultivars with a high propensity to produce double embryos with different ploidy levels: Alpha, Andreas, Boonlim, Cipres, Eposs, Helios, Limbras, Ravel and Sartaguda. Twin embryos inside a single seed were found in 3 cultivars: Eposs 2n, Ravel 2n and Sartaguda 2n, at 0.60% frequency (15 seeds with twin embryos out of 2500 seeds). Of 30 obtained seedlings, 14 were separated diploid-diploid twins, 6 were conjoined diploid pairs, 8 were separated diploid-haploid and 2 were diploid-haploid pairs conjoined in the hypocotyl region. Some embryos showed unilateral dominance of one embryo (size and shape). The haploid status of the smallest embryo was confirmed by chromosome number (n=x=10) and flow cytometry (nuclear C DNA amount 1.95 pg). The haploid obtained in this manner possessed enough vegetative vigor to undergo chromosome doubling.

Serine inhibitors have been described in many plant species and are universal throughout the plant kingdom. Trypsin inhibitors are the most common type. In the present study, trypsin and chymotrypsin inhibitory activity was detected in the seed flour extracts of four Egyptian varieties of soybean (Glycine max). The soybean variety, Giza 22, was found to have higher trypsin and chymotrypsin inhibitory potential compared to other tested soybean varieties. For this reason, Giza 22 was selected for further purification studies which used ammonium sulphate fractionation and DEAE-Sephadex A-25 column. Soybean purified proteins showed a single band on SDS-PAGE corresponding to a molecular mass of 17.9 kDa. The purified inhibitor was stable at temperatures below 60°C and was active at a wide range of pH, from 2 to 12 pH. The kinetic analysis revealed a non-competitive type of inhibition against trypsin and chymotrypsin enzymes. The inhibitor constant (Ki) values suggested that the inhibitor has higher affinity toward a trypsin enzyme than to a chymotrypsin enzyme. Purified inhibitor was found to have deep and negative effects on the mean larval weight, larval mortality, pupation, and mean pupal weight of Spodoptera littoralis. It may be concluded, that soybean protease inhibitor gene(s) could be potential targets for those future studies which are concerned with developing insect resistant transgenic plants.

Proteases are one of the most important digestive enzymes in the midgut of Hyphantria cunea Drury. Proteases are responsible for protein digestion. In the present study, we evaluated the efficiency of some plant inhibitors on proteases in the gut of the H. cunea. Last instar larvae were collected from mulberry trees. The digestive system of the larvae was used as an enzyme source. The total proteolytic and trypsin activity were assessed by the hemoglobin and BApNA, respectively, as the substrate. The evaluation of the total proteolytic and trypsin activities in various pHs showed the highest relative activity at a pH of 11. Also, the inhibitory effect of inhibitors extracted from Alhagi maurorum Medik., Lathyrus sativus L., Vicia faba L., Prosopis farcta (Banks & Sol.) Eig., and Panicum miliaceum L. on the digestive protease of the fall webworm was measured. Protease inhibitors extracted from A. maurorum, P. farcta and P. miliaceum showed negligible inhibition but L. sativus was able to inhibit 34.72% and 100% of the total activity of proteolytic and trypsin, respectively. Also, the total proteolytic and trypsin activities were inhibited by the inhibitor from V. faba, at 22.27% and 100%, respectively. The zymogram pattern of trypsin with nitro-cellulose membranes showed 2 isoforms in the gut of H. cunea. The inhibitor from L. sativus completely inhibited both isoforms. Gel electrophoresis of proteolitytic activity revealed at least 6 isoforms the inhibitor extracted from L. sativus; completely inhibiting some of them. The inhibitor from L. sativus was purified by ammonium sulfate precipitation and gel-filtration. The molecular mass of the inhibitor was determined as 45 kDa. The highest inhibition of trypsin activity by the inhibitor from L. sativus occurred at a pH of 10. The stability of the inhibitor from L. sativus was evaluated at different pHs and temperatures. The results showed that the inhibitor from L. sativus was stable at a pH of 11.0, and showed 45% inhibition on trypsin activity at a pH of 11. Also, this inhibitor revealed stability up to 50°C.

The article presents an experimental-theoretical analysis of fluidised-bed drying of poppy seeds directed on minimisation of energy. The analysis was performed for a complete drying node incorporating a heat exchanger and a fan. Two complementary factors were used in the exergetic evaluation: exergy efficiency and unit consumption of exergy. An analysis of drying in stationary bed was carried out for comparison purposes. Results of the exergetic analysis can become a basis for innovative works focused on decreasing energy consumption of a technological node being analysed, e.g. by the use of recirculation of fluidising-drying medium.

Shoot tips excised from shoot culture of Salvia officinalis were encapsulated in 2% or 3% (w/v) sodium alginate and exposed to 50 mM calcium chloride for complexation. Immediately or after 6, 12 or 24 weeks of storage at 4°C, the synthetic seeds were cultured for 6 weeks on half-strength MS medium supplemented with indole-3- acetic acid (IAA) (0.1 mg/l) and solidified with 0.7% agar. The frequency of shoot and root emergence from encapsulated shoot tips was affected by the concentrations of sodium alginate and additives in the gel matrix (sucrose, gibberellic acid, MS nutrient medium) as well as duration of storage. The frequency of shoot and root induction of non-stored synthetic seeds was highest with shoot tips encapsulated with 2% sodium alginate containing 1.5% sucrose and 0.5 mg/l gibberellic acid (GA3). Shoot tips maintained their viability and ability to develop shoots even after 24 weeks of storage when they were encapsulated in 3% alginate with 1/3 MS medium, sucrose (1.5%) and GA3 (0.25 mg/l). Root formation tended to decrease with storage time. Overall, 90% of the plantlets derived from stored and non-stored synthetic seeds survived in the greenhouse and grew to phenotypically normal plants. This procedure can enable the use of synthetic seed technology for germplasm conservation of S. officinalis, a plant species of high medical and commercial value.

On wheat seeds harvested in Upper Silesia (south-western Poland) a large number of species of Fusarium were recorded. Fusarium avenaceum and F. culmorum were most commonly isolated. Flooding of fields for 1-7 days caused the increase of pathogenic Fusarium spp. in seed samples, especially F. culmorum. The infection of seeds caused a rapid decrease of their viability, and negatively affected health status of emerging seedlings in field conditions, in spite of performed chemical treatment.
W artykule przedstawiono wyniki analizy mikologicznej ziarna pszenicy ozimej z terenów objętych powodzią na Górnym Śląsku (południowo-zachodnia Polska). Zalanie upraw pszenicy wodą przez okres 1-7 dni spowodowało wzrost udziału w mikoflorze ogólnej ziarna patogenicznych gatunków Fusarium, głównie F. culmorum. Procentowy udział gatunku F. avenaceum w grupie patogenicznych gatunków z tego rodzaju był wyższy na ziarnie z terenów niezalanych wodą. Stwierdzono negatywny wpływ wysokiego zasiedlenia ziarna przez gatunki Fusarium na jego zdrowotność, co wyrażało się znacznym obniżeniem siły kiełkowania w czasie jego przechowywania oraz obniżeniem zdrowotności siewek.

Secondary plant substances - glucosinolates and saponins reduced seed germination and seedling growth of lettuce. Saponins showed greater inhibitory effect on both germination and growth of lettuce seedlings than glucosinolates. The effect depended on the concentration of these compounds in the ground. Tested compounds caused abbreviation of roots, browning their tips and prevented root hair development.
Testowano wpływ roślinnych metabolitów wtórnych: saponin lucerny i glukozynolanów rzepaku na kiełkowanie nasion sałaty. Stwierdzono hamujący wpływ obu badanych związków na kiełkowanie. Znacznie silniejsze działanie wykazywały saponiny, przy czym efekt ten zależny był od ich stężenia w podłożu. Badane związki wpływały nie tylko na liczbę skiełkowanych nasion, ale również na wygląd siewek (skrócenie korzonków, brunatnienie ich końców, brak włośników).

The effect of W92/72 transgenic flax seeds taken from a variety of Linola on the production of SCFA, ammonia and methane by bacteria inhabiting rabbit caecum was studied. The in vitro method was used where caecal contents from rabbits was incubated with W92/72 transgenic or Linola flax seeds, or without any additives (control samples).The total concentration of SCFA was higher in samples with the addition of flax seeds than in the control samples. The increase in concentrations of acetic, propionic and butyric acids was the highest in samples with Linola seeds added. A higher percentage of propionic and butyric acids was observed in the contents incubated with addition of flax seeds as compared to the control samples. This increase was the result of a percentage decrease in acetic acid. No differences were observed in the concentration of ammonia between fermented samples. Moreover, the addition of flax seeds resulted in slight decrease of pH in incubated samples. In gas samples, the methane level was higher in samples with flax seeds added, although the highest level was found in samples with transgenic seeds.In addition, gas pressure was significantly higher in samples with flax seeds added as compared to control samples, and this may indicate a higher intensity of microbiological fermentation processes. These studies suggest that neither Linola nor W92/72 flax seeds have any unfavorable effect on the caecal microflora activity of rabbits. A beneficial influence of flax seeds on the microbiological fermentation process in rabbit caecum was observed, based on an increase in percentage ratio of propionic acid in samples with flax seeds added.
