Wprowadzenie i cel pracy. Kleszcze są zaliczane do obligatoryjnych ektopasożytów kręgowców lądowych, w tym również i człowieka. Stanowią rezerwuar, niekiedy wektor wielu drobnoustrojów chorobotwórczych (bakterii, wirusów i pierwotniaków) zagrażających zdrowiu zwierząt i ludzi. Rodzaje Theileria i Babesia stanowią duże zagrożenie głównie dla zwierząt, powodując m.in. gorączkę wschodniego wybrzeża, tropikalną tejleriozę, babeszjozę.
Introduction and objective. Ticks are classified into obligatory eco-parasites of terrestrial vertebrates, including humans. They constitute a reservoir, sometimes a vector of many pathological microorganisms (bacteria, viruses and protozoa) threatening the health of humans and animals. The primary objective of the study was an attempt to evaluate whether ticks of the species Ixodes ricinus and Dermacentor reticulatus, commonly occurring in Poland, may constitute a potential vector and reservoir for the protozoa: Babesia, Theileria, and Toxoplasma gondii, and at the same time, play a role in the circulation of these pathogens in nature. Brief description of the state of knowledge. The genera Theileria spp., Babesia spp, and as has been found in recent years, Toxoplasma gondii belong to the group of protozoa occurring in ticks. The genera Theileria and Babesia create a great risk, mainly for animals, causing, among others, east coast fever, tropical theileriosis and babesiosis. In the case of Toxoplasma gondii, this protozoan is especially dangerous for pregnant women, and may cause permanent and irreversible foetal defects. Studies to-date confirm the presence of these protozoa in some species of ticks, as well as the possibility of co-occurrence of some of them in one individual tick. Summing up. From the aspect of public health, studies concerning the potential role of ticks in the transmission of parasitic protozoa may contribute to the assessment of the actual risk of infection in the case of tick bites. The results concerning the role of ticks as an alternative route of transmission of T. gondii – the etiologic factor of toxoplasmosis, would be of a special importance for pregnant women.

Opis:
For the first time in the world literature, the presented study describes oral-anal contact between two sympatric Ixodes ricinus and Dermacentor reticulatus ticks, which are the vectors of many pathogens in Europe. Among the specimens collected in nature, an I. ricinus male was found with the hypostome and chelicerae inserted in the anal aperture of a D. reticulatus female. Given the biological and physiological traits of both species and microorganisms colonising these arthropods, it seems that the interspecific oral-anal contacts between ticks may be an unknown alternative route of transmission of pathogenic and symbiotic microorganisms present in ticks’ alimentary tract, faeces, body surface, and in the haemolymph and organs, taking place when the male’s mouthparts are inserted into the female’s body. The authors suggest that interspecific contacts between ticks in natural conditions can contribute to the spread of some microorganisms among different tick species and their hosts.

A total of 305 ticks (21 larvae, 243 nymphs, 19 females and 22 males) were collected by fl agging of vegetation in suburban woods of Pisárky Park (city of Brno) from July to October 2002. The midgut of each tick was dissected out and transferred individually into BSK-H medium. After cultivation, all specimens were examined by dark-fi eld microscopy (DFM) for the presence of borreliae. Out of 305 tick samples, 45 were (14.8%) DFM positive. The following polymerase chain reaction (PCR) then revealed 37 (12.1%) samples positive for the presence of Borrelia burgdorferi sensu lato DNA. All 37 samples were further analysed by restriction fragment length polymorphism (RFLP) method. PCR-RFLP analysis revealed 14 strains of B. afzelii (37.8%), 15 strains of B. garinii (40.5%) and 2 strains of B. burgdorferi sensu stricto (5.4%). Four samples (10.8%) showed a mixed population of these genospecies. Two samples produced atypical RFLP pattern which were detected by sequence analysis as B. valaisiana (5.4%). Isolation attempts resulted in 21 spirochaetal strains (including two stains of B. valaisiana). The results show the diversity of B. burgdorferi s.l. in tick population and refer the risk of infection by pathogenic borreliae in Brno.

Opis:
Ticks are known as vectors of a wide range of pathogens of medical and veterinary importance; some of them of zoonotic concern constitute a hazard for the emergence of tick-borne diseases shared between humans and domestic animals and becoming a part of the ‘One Health’ concept. Canine and feline tick-borne diseases have emerged in recent years, performing an extensive geographic distribution and enlarged global prevalence. The present review focuses on the recent epidemiological studies on the emergence of tick-borne bacterial pathogens in dogs and cats, and the discussion whether pet ownership increases the risk of tick-borne diseases. A lot of data provide confirmation that dogs and cats themselves may substantially contribute to the circulation of the ticks and tick-borne bacterial pathogens in the environment. Molecular diagnostics of tick-borne pathogens infections generates a lot of problems like the choice of molecular methods and molecular markers for the detection of bacterial genomic DNA, but play an important role in the diagnosis of infections. The study provides some insight into molecular diagnostic techniques and new potentially recognized bacterial pathogens of this group. Protecting human and companion animal health from vector-borne infections requires controlling vector populations, containing development of novel, practicable strategies that will limit vectors and transmission of vectorborne disease pathogens.

Opis:
Introduction. Ticks (Acari: Ixodida) are vectors and/or reservoirs of many pathogens, i.e. Borrelia burgdorferi sensu lato, Anaplasma phagocytophilum and Babesia microti. These pathogens are ethiological agents of such diseases as Lyme borreliosis, human granulocytic anaplasmosis and human babesiosis. Objective. The aim of the study was to evaluate the role of the Ixodes ricinus in the transmission of Borrelia burgdorferi sensu lato, Borrelia afzelii, Borrelia garinii, Borrelia burgdorferi sensu stricto, Anaplasma phagocytophilum and Babesia microti in Opolskie Province in Poland. Materials and method. DNA from 222 ticks was isolated by the ammonia method. The pair of primers specific to the flagelline gene was used to detect of B. burgdorferi s. l. To detect of genospecies of this spirochete, three pairs of internal primers were used. In turn, two pairs of primers specific to the 16S rDNA gene and the 18S rRNA were used, respectively, for the detection of A. phagocytophilum and B. microti. Borrelia burgdorferi s. l., A. phagocytophilum, and B. microti were detected in 4.5%, 2.7% and 5.4% of examined ticks, respectively. Results and conclusions. Of the ten ticks infected with B. burgdorferi s. l., B. afzelii was found in seven, undefinied genospecies in two, and mixed infection with B. afzelii and B. burgdorferi s. s. in one. The study demonstrated the potential risk of exposure of humans and animals to infections of B. burgdorferi s. l., A. phagocytophilum and B. microti in the examined area of Poland.

Opis:
A total of 684 Ixodes ricinus ticks (321 nymphs, 184 males, and 179 females) were collected by fl agging lower vegetation in 6 forest districts located on the territory of Lublin province (eastern Poland). Ticks were examined by polymerase chain reaction (PCR) method for the presence of Anaplasma phagocytophilum DNA with two pairs of primers: EHR521/EHR747 for detecting 16S rRNA gene, and LA6/LA1 for detecting ankA gene. To study the relationship between infection in ticks and people occupationally exposed to tick bite, blood serum samples of 261 forestry workers employed in the same forest districts were examined by immunofl uorescence method for the presence of specifi c antibodies against A. phagocytophilum. A total of 70 ticks out of 684 examined (10.2%) showed the presence of A. phagocytophilum 16S rRNA gene. The prevalence of infection was signifi cantly dependent on tick’s stage (χ2=49.2, p<0.00001) and geographical locality (χ2=34.4, p<0.00001). The percentage of I. ricinus females infected with A. phagocytophilum (24.6%) was signifi cantly greater compared to males (6.5%) and nymphs (4.4%) (p<0.00001). Only 19 ticks out of 684 examined (2.8%) showed the presence of A. phagocytophilum ankA gene, signifi cantly less compared to 16S rRNA gene (p<0.00001). The prevalence of infection demonstrated by the presence of ankA gene was also signifi cantly dependent on tick’s stage (χ2=23.6, p<0.00001) but not on locality (χ2=9.8, p=0.082). A signifi cant correlation was found between the presence of A. phagocytophilum 16S rRNA gene in I. ricinus female ticks from the particular forest districts and the serologic response to A. phagocytophilum of forestry workers employed in the same districts (p<0.05). No signifi cant correlation was found between the presence of A. phagocytophilum ankA gene in I. ricinus ticks and serologic response of exposed workers. In conclusion, detection of A. phagocytophilum infection in ticks by PCR with the use of EHR521/EHR747 primers detecting 16S rRNA gene is signifi cantly more sensitive compared to LA6/LA1 primers detecting ankA gene.
