In this study we report the usefulness of nested PCR for screening of the persistent B. microti infections in rodent hosts. Female BALB/c mice were inoculated with 100 μl of donor blood infected with B. microti. Infections were detected using microscopic examination of Giemsa-stained blood smears. To determine whether B. microti DNA was present in blood and/or spleen tissue, nested PCR was performed targeting a specifi c fragment of the gene encoding the 18S rRNA. Blood was sampled every 10 days post-infection (dpi) until day 30, after which mice were sampled every 30 days until the end of experiment at 360 dpi. The most extensive parasitaemia (39% of infected erythrocytes) was observed at 10 dpi. Between 20-60 dpi, less then 1% of infected erythrocytes were detected in blood smears, and from 90 dpi onwards, infected erythrocytes were no longer observed. B. microti DNA was successfully amplifi ed from the blood of mice from 10 dpi until 180 dpi, as well as from spleens of infected mice at 10 and 20 dpi. The presented results show that nested PCR is the method of choice for monitoring infections of B. microti in the blood of rodent hosts, and could therefore be a tool for environmental monitoring of naturally infected rodents which are the predominant source of infection for tick vectors.

The effects of long-term administration of a standardised extract of Curcuma longa on the explorative activity in aged 24-month-old male Wistar rats were estimated in a hole-board test. The animals received the extract orally for two months in rodent chow at doses of 10 and 50 mg/kg/day. The correlations between concentration of some neurotransmitters and amino acids in selected brain regions, as well as the level of corticosterone in plasma (described previously), and the parameters of exploration were calculated. The antioxidant processes (GSH, GST, CAT and MDA levels) in heart and skeletal muscles were also investigated. The results suggested that the explorative activity of plant extract-treated rats was enhanced; however, no correlations between brain neurotransmitter concentration or plasma corticosterone level and the parameters of explorative activity were observed. In biochemical investigation, chronic C. longa extract administration influenced antioxidant processes (CAT, GST and MDA levels) in skeletal muscles of aged rats, but not in the heart muscle.

Introduction and objective. Deficits of vitamin resources constitute a significant public health problem, especially among the elderly population. The aim of the research was to determine the level of vitamin 25 (OH) D and vitamins from group B in the chronically ill elderly in domiciliary care, depending on functional capacity and coexisting diseases. Materials and method. The pilot study included 137 patients staying in long-term domiciliary care. Samples of the participants’ venous blood was obtained for laboratory tests. Centrifuged serum was used to determine the level of the following biochemical parameters: vitamin 25 (OH)D, B12, folic acid and total protein, albumin, triglycerides, total cholesterol and HDL cholesterol. Assessment of the functional status of patients was made by using the Barthel scale. Results. More than 3⁄4 of the patients with functional deficit (according to Barthel’s score 0–85 points) were deficient in vitamin 25 (OH)D, while folic acid values were below the reference values in more than half of the patients. Respondents with lower functional efficiency were characterised by a reduced average value of vitamin 25 (OH)D and folic acid. Conclusions. The studied group of the chronically ill elderly was characterised by a deficiency of vitamin D3 and folic acid. Subjects with a functional impairment deficit show a reduced mean value of vitamin 25 (OH)D and folic acid in the blood serum, compared to the group of patients with higher mobility.
