Due to interesting role of dihydropyridines in cardiovascular diseases and drug resistance studies and lack of a fluorine-18 labeled imaging agent for L-type calcium channel studies, this study was designed. [18F]Dimethyl 2-(fluoromethyl)-6-methyl-4-(2-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylate 2 was prepared in no-carrier-added (n.c.a.) form from a starting brominated compound in one step at 80°C in Kryptofix2.2.2/[18F]. Compound 2 was administered to normal rats via their tail veins for preliminary biodistribution studies and the ID/g% of the labeled compound was determined up to 3 h post injections. Coincidence images were obtained in rats 5 to 120 min. Radiofluorination on bromo precursor gave a fluorinated compound in 95% radiochemical purity and a 8% yield shown by RTLC and HPLC. Biodistribution studies showed that the tracer is accumulated in the heart in the first few minutes, followed by metabolism resulting in very soluble 18F-containing metabolites eliminated through the urinary tract. In coincidence images, the target organ was shown to be the heart. Lung had high accumulation possibly due to the presence of Ca2+ channels and/or hydrolyzing enzymes showing a significant myocardial uptake at 120 min. The data demonstrates a significant agreement with the reported L-type calcium channels throughout the animal body. To our knowledge, this is the first example of 18F-DHPs in the literature.

Abstract Bleomycin (BLM), labeled with radioisotopes, is widely used in therapy and diagnosis. In this study, BLM was labeled with 62Zn for oncologic PET studies. The complex was obtained at pH = 2 in saline at 90°C in 25 min. Radio-TLC showed an overall radiochemical yield of 95 97% (radiochemical purity > 97%). Stability of complex was checked in vitro in mice and human plasma/urine. Preliminary in vivo studies were performed to determine complex stability and distribution of 62Zn BLM in normal and fibrosarcoma-bearing mice. 62Zn BLM accumulated significantly in induced fibrosarcoma tumors in mice according to biodistribution/imaging studies. 62Zn BLM can be used in PET oncology studies due to its suitable physicochemical properties as a diagnostic complex in vitro and in vivo. Further studies should be performed for evaluation of the complex behavior in larger mammals.

The 18O isotopic water permeation and separation characteristics of a hydrophobic PTFE membrane using Air Gap Membrane Distillation (AGMD) and Vacuum Enhanced Membrane Distillation (VEMD) were investigated. Permeation fluxes were measured by weighing the collected membrane-permeated water vapor. 18O/16O of each water sample was analyzed by the Tunable Diode Laser Absorption Spectroscopy (TDLAS). We observed the effects of the air filled membrane pores and the temperature gradient applied to the membrane surfaces on the vapor permeation flux and the oxygen isotope separation for the first time. For both AGMD and VEMD, the permeation flux and the degree of 18O separation increased as the membrane interfacial temperature gradient increased. Even though, oxygen isotope separation and the permeation flux for VEMD is slightly higher than AGMD, the latter may be more efficient from the system's operational point of view.

In this paper, we present prospects for using the Jagiellonian positron emission tomograph (J-PET) detector to search for discrete symmetries violations in a purely leptonic system of the positronium atom. We discuss tests of CP and CPT symmetries by means of ortho-positronium decays into three photons. No zero expectation values for chosen correlations between ortho-positronium spin and momentum vectors of photons would imply the existence of physics phenomena beyond the standard model. Previous measurements resulted in violation amplitude parameters for CP and CPT symmetries consistent with zero, with an uncertainty of about 10−3. The J-PET detector allows to determine those values with better precision, thanks to the unique time and angular resolution combined with a high geometrical acceptance. Achieving the aforementioned is possible because of the application of polymer scintillators instead of crystals as detectors of annihilation quanta.

Copper-61 (T1/2 = 3.33 h) produced via the 64Zn(p,alfa)61Cu nuclear reaction, using a natural zinc target, was separated from the irradiated target material by two ion exchange chromatography steps and was used for the preparation of [61Cu]-diethylenetriaminepentacetate ([61Cu]DTPA) using freshly-prepared DTPA cyclic dianhydride. An electroplated natural zinc layer on a gold-coated copper backing was irradiated with 22 MeV protons (22 12 MeV on the target, 180 miA irradiation, 3.2 h, final activity 220 GBq of 61Cu2+, RCY > 95%, radionuclidic purity > 99%, 60Cu as the only radionuclidic impurity; T1/2 = 23.7 min). Colorimetric methods showed that traces of chemical impurities in the product were below the accepted limits. The solution of [61Cu]DTPA was prepared with a radiochemical yield of more than 80% starting with 61CuOAc ligand at room temperature after 30 min. RTLC showed the radiochemical purity of more than 99%. The specific activity obtained was about 9.1 TBq/mmol. The tracer was shown to be stable in the final product and in the presence of human serum at 37°C up to 3 h.

Positron Emission Tomography (PET) is a method for determining biochemical and physiological processes in vivo in a quantitative manner. The most commonly used radionuclides are 11C, 13N, 15O and 18F, with respective half-lives of approximately 20 min, 10 min, 2 min, and 110 min. 18F labeled FDG (fluoro-2-deoxy-D-glucose) is now the most frequently used radiopharmaceutical and finds its application prominently in the field of oncology. Originally, the production of these radionuclides was performed with the existing accelerators, designed for nuclear physics, but with increasing interest in the PET methodology specially designed PET-production cyclotrons became available. The nuclear reactions involved are (p,n), (d,n), (p,a) and (d,a) and the thresholds for the nuclear reactions involved are 5 to 6 MeV. Based on these values and on other parameters, a proton 15 to 20 MeV cyclotron is often chosen. Since the half-life of a radionuclide limits the production time, the maximum beam current is an important parameter, together with the target construction, for the ultimate yield obtainable. In the development of special PET production cyclotrons, attention has also been paid to improve the extraction efficiency and the possibility of multiple extractions by designing negative ion cyclotrons. Commercial cyclotrons can often be acquired as an easy to operate integrated radionuclide production unit including targetry and some units. Regional FDG factories are nowadays being created to fulfil the demand for PET radiopharmaceutics. The possible choices in commercially available cyclotrons for the production of PET radionuclides will be discussed.

68Ga is an important positron-emitting radionuclide for positron emission tomography. In this work 68Ga was produced via the 68Zn(p,n)68Ga nuclear reaction. 68Zn electrodeposition on a copper substrate was carried out by alkaline cyanide baths. 68Zn target was irradiated with a 15 MeV proton beam and a 150 mi A current. The production yield achieved was 136 mCi/ mi Aźh (5.032 GBq/mi Aźh). 68Ga was separated from zinc and copper by a combination of cation exchange chromatography and liquid-liquid extraction methods.

The main aim of this work is to investigate the use of new lanthanum bromide (LaBr3) in whole body PET (WB-PET) scanners and to compare the obtained results with those for BGO and LSO crystals which are currently used in conventional and dedicated PET systems. Our results show that there is a gain in noise effective count rate (NECR) arising from the reduced scatter and random fractions in a LaBr3 scanner. The spatial resolution of the WB-PET with LaBr3 is slightly worse than the LSO and BGO crystals because of its low density and effective atomic number. But our study showed the increased NECR, excellent energy resolution and low decay time of LaBr3 which promise a significant improvement in WB-PET performances especially for narrow coincidence windows.

Opis:
The Jagiellonian Positron Emission Tomograph (J-PET) collaboration is developing a prototype time of flight (TOF)-positron emission tomograph (PET) detector based on long polymer scintillators. This novel approach exploits the excellent time properties of the plastic scintillators, which permit very precise time measurements. The very fast fi eld programmable gate array (FPGA)-based front-end electronics and the data acquisition system, as well as low- and high-level reconstruction algorithms were specially developed to be used with the J-PET scanner. The TOF-PET data processing and reconstruction are time and resource demanding operations, especially in the case of a large acceptance detector that works in triggerless data acquisition mode. In this article, we discuss the parallel computing methods applied to optimize the data processing for the J-PET detector. We begin with general concepts of parallel computing and then we discuss several applications of those techniques in the J-PET data processing.

The J-PET detector being developed at the Jagiellonian University is a positron emission tomograph composed of the long strips of polymer scintillators. At the same time, it is a detector system that will be used for studies of the decays of positronium atoms. The shape of photomultiplier signals depends on the hit time and hit position of the gamma quantum. In order to take advantage of this fact, a dedicated sampling front-end electronics that enables to sample signals in voltage domain with the time precision of about 20 ps and novel reconstruction method based on the comparison of examined signal with the model signals stored in the library has been developed. As a measure of the similarity, we use the Mahalanobis distance. The achievable position and time resolution depend on the number and values of the threshold levels at which the signal is sampled. A reconstruction method as well as preliminary results are presented and discussed.

Opis:
A new automated target for high yield production of aqueous [18F] fluoride at high beam currents has been constructed for a cyclotron at NRCAM in Karaj (Cyclone 30). It consists of one small golden sphere inside another target chamber, mounted into a special holder, which provides rapid cooling by water flow around and inside the sphere. The target is irradiated with 28 MeV protons. The incident energy on the target chamber is 18 MeV. This target is operated without external overpressure and has been tested for beam currents up to 60 ěA. The measured target yield is 80% of the theoretically calculated yield of 18F, which is used for the synthesis of [18F] 2-fluoro-2-deoxy-D-glucose.

Instytut Chemii i Techniki Jądrowej jest jedną z nielicznych placówek w Polsce, w których prowadzone są badania nad otrzymywaniem nowych radiofarmaceutyków. Prace w tym zakresie prowadzi Pracownia Badań i Syntezy Radiofarmaceutyków pod kierownictwem prof. dr hab. Aleksandra Bilewicza. Tematyka prowadzonych badań jest ukierunkowana na opracowanie nowych metod produkcji radionuklidów oraz sposobów ich dostarczania do miejsc zmian patologicznych w organizmie człowieka. W artykule przedstawiono opisy badań realizowanych przez doktorantów tej Pracowni w ramach prac doktorskich. Wśród tych prac należy wymienić nanonośniki dla emiterów promieniowania alfa, badania nad inhibitorami cholinoesteraz, nowe metody wytwarzania 99mTc i 43Sc czy produkcję radiofarmaceutyków teranostycznych.
The Institute of Nuclear Chemistry and Technology is one of the few institutions in Poland, where research on the acquisition of new radiopharmaceuticals is being conducted. The work in this area is carried out by the Laboratory of Research and Synthesis of Radiopharmaceuticals under the supervision of prof. dr hab. Aleksander Bilewicz. The research is focused on the development of new methods of radionuclide production and methods of their delivery to pathological changes in the human body. The article presents descriptions of research carried out by doctoral students as part of their theses. These include the nanocarriers for alpha-emitters, the study of cholinesterase inhibitors, new methods for the production of 99mTc and 43Sc, as well as the production of teranostatic radiopharmaceuticals.

Opis:
The increasing numbers of PET studies for routine diagnosis creates a real hazard to radiation workers. The aim of this study is to estimate the annual whole-body and finger radiation dose to technologists working with 18F-FDG in a PET center with high patient potential. In our PET center, the number of PET imaging has increased almost to 5000 studies per year. Our standard dose for tumor imaging is 518 MBq of 18F-FDG. Five technologists performing all steps of 18F-FDG imaging (5 patients per technologist in a day) were officially involved round the week for handling and injecting 18F-FDG to patients. Whole-body and finger dose measurements with TLDs were performed for two different time periods: i) before shielding precautions during the first 6 months (without a shielding for sterile syringe and without a lead container for shielded syringe) and ii) after shielding precautions during the next 6 months (with a shielding for sterile syringe and with a lead container for shielded syringe). The average annual whole-body radiation dose for one technologist before shielding precautions was 7.82 mSv and after shielding precautions was 5.76 mSv. On the other hand, while the average annual finger radiation doses for one technologist before shielding precautions were 210.36 and 293.72 mSv for the left and right hand, after shielding precautions were they 158.16 and 217.58 mSv for the left and right hand, respectively. According to our results, if one technologist performs the whole-body PET imaging of 5 patients per day, the annual radiation dose to this technologist will not exceed the recommended limits by ICRP.

Preliminary design of a spherical brain PET (SBPET) using liquid xenon (LXe) as detector is considered in this research work. The major advantage of a spherical design is the large solid angle of acceptance which improves the sensitivity and increases signal-to-noise ratio (SNR) of the image. The use of a liquid active medium enabled us to design a spherical detector. LXe, due to the intrinsic physical properties, is an excellent liquid medium for accurate tracking of gamma rays in the relevant energy range. The performance of SBPET was evaluated by Monte Carlo simulation tools (GATE) and compared to ECAT HRRT. The numerical results showed the SBPET has a sensitivity of 1.14% and spatial resolution of ~2.7 mm FWHM which is superior to ECAT HRRT especially at high-count rates.
