Five different strains of Saccharomyces cerevisiae were tested for their abilities to accumulate uranium from waste water containing competitive ions. Samples of water passing out from a previous uranium mill were used. The strains tested possess different abilities to accumulate uranium. The kinetics of bioaccumulation, the leaching degree, the influence of cell density and their origin were investigated. Under the applied experimental conditions, more than a half of the total activity (uranium and the decay products) could be accumulated after 60 min contact time of 1 mL S. cerevisiae suspension and 5 mL of water. The other cations present in solution effectively competed for the uranium accumulation. 226Ra and its decay products were completely retained using all tested strains.

The possibility of bioaccumulation of uranium species in beer yeast was investigated. The behaviour of the Saccharomyces cerevisiae–UO2 2+ system was studied vs. contact time, pH and anion nature with no ionic competition. Analysis of the data revealed the following optimal working conditions: contact time = 1 h, pH = 6.5 and 10-1 M UO2(CH3COO)2 solution as uranyl source; as a result, the maximum degree of bioaccumulation attends a value nearly 8.75 mmol UO2 2+/g yeast. Both, a scanning electron microscope (SEM) and amino acid determinations lead to the conclusion that the uranyl nitrate solution may devastate the yeast cells provoking membrane damage and the release of the cell constituents (including the bioaccumulated uranium species). The results suggest the possible use of Saccharomyces cerevisiae as a biological decontaminant of uranium containing wastewaters.

Opis:
Obtaining specific information on contaminant biokinetics in marine biota is often necessary for properly interpreting monitoring data on trace contaminant levels in bioindicator species living under varying environmental conditions. Radiotracers have been employed in laboratory experiments to assess the uptake, distribution and retention of selected heavy metals and PCB congeners in three potential marine bioindicators occupying different ecological niches in the coastal zone. Pelagic and benthic jellyfish readily accumulated Co, Ag, Zn, Cd, 137Cs and 241Am from both water and food and retained them with biological half-lives (Tb1/2) ranging from a few days to several weeks. Zinc and silver were accumulated to the greatest degree (CF ~ 4 ´ 102), with benthic jellyfish having a greater affinity for metals than the pelagic species. Results from light–dark experiments indicate that the enhanced metal uptake in the benthic jellyfish is due to the presence of endosymbiotic photosynthetic zooxanthellae situated in the arms of organisms. Shore crabs ingesting Ag, a sewage-related contaminant, readily accumulated the metal with male crabs assimilating some 71% and female crabs 51% of the Ag from their food. Moreover, the assimilated fraction of Ag remained virtually immobile in their tissues as evidenced by an extremely long Tb1/2 for depuration of 7.3 years. Sea stars exposed to 14C-labelled PCB congener #153 in sea water accumulated the congener mainly in the body wall and podia reaching lipid weight CFs that ranged between approximately 2 x 10 5 to 4 x 10 5. In contrast, following exposure in radio-labelled sediments, the corresponding PCB transfer factors in the same tissues were much lower, viz., 3 x 10 2 to 5 x 10 2. Nevertheless, regardless of the exposure mode, CFs of PCB in the other tissues (digestive system, gonads, pyloric and rectal caeca) were consistently one to two orders of magnitude lower, an observation which suggests that sea star body wall and podia could serve as target tissues in biomonitoring studies assessing these toxic compounds.

Caesium bioaccumulation experiments were carried out at 4 to 60°C using natural samples of the lichen Hypogymnia physodes. Thalli were incubated in 2.5 mi mol.l 1 CsCl solutions labelled with 137CsCl for up to 24 h at pH values from 2 to 10. Bioaccumulation of Cs+ ions in the first phase of the lichen-CsCl solution interaction is rapid, neither pH, nor temperature dependent within the range 4 to 60°C and observed also with the lichen biomass thermally inactivated at 60°C or chemically by formaldehyde. The second phase of 137Cs bioaccumulation is time, temperature and pH dependent and is inhibited by formaldehyde and thermal inactivation. The process at the initial concentration C0 = 2.5 ěmol.l 1 CsCl and 20°C reached equilibrium within 12 hours. It can be described by the first order reaction kinetics equation: log [Ct] = 1.89 - 0.00153 t, R = 0.950. Maximal values of Cs-bioaccumulation were observed at 20°C with minimum at 4°C and 40°C and at pH 4 5 with minimum at pH 2 and pH 6. Low caesium efflux values from lichen thalli by water and 0.1 mol.l 1 neutral salts at 20°C and 24 h equilibrium were observed. Efflux characterized by distribution coefficients D = [Cs]solution/[Cs]biomass at biomass/solution ratio 1:25 (w/v, wet wt.), decreases in the order: Li+- 78 × 10 3 >NH4 + = K+ 15 × 10 3 > Cs+ = Na+ 11 × 10 3. Low extractability of caesium from lichen by water and salt solutions can explain long persistent times of radiocaesium contamination sorbed by lichens, observed by many authors in caesiumcontaminated forest and mountain regions. Hypothesis of the role of the lichen secondary metabolites as caesium binders is discussed.

In principle, there are two sources of environmental radionuclides, namely natural and man-made. In comparison with other environmental pollutants as, e.g., heavy metals, some scientific and technical disciplines profit by the natural radionuclides, using them for age determination of the samples, e.g., hydrology, geology and archaeology. Nevertheless, the environmental radionuclides represent mostly risk for a human health. Therefore, in this review, their sources are shortly described and original recent analyses of some bioaccumulation methods are presented, using either selected plant species or microorganisms, respectively. The different uptakes in all systems were measured to evaluate the possibility of application of the tested biological materials as markers of environmental contamination. It was also demonstrated that the detection methods developed for analyses of bioaccumulation of environmental radionuclides could be applied in radiotracer methodology for a study of bioaccumulation of heavy metals. While gamma spectrometry for soil samples and liquid scintillation spectrometry for water samples were used, autoradiography was applied to analyze the distribution of radionuclides during and/or after bioaccumulation.

A model species of saprophytic fungus, king oyster mushroom (Pleurotus eryngii), was cultivated on barley substrate supplied with [Pt(NH3)4](NO3)2, under well defined conditions. The samples of the collected fruiting bodies were digested and analyzed for total platinum content by means of ICP-MS. The results proved that platinum is not accumulated in the fruitbodies of Pleurotus eryngii for a wide range of Pt concentrations in the culture substrate (100 1000 ppb Pt in 50 ml of water solution added to ca. 450 g of hydrated barley seeds per container). Observable levels of Pt were only found in the fruitbodies obtained from the medium contaminated with 10000 ppb (10 ppm) platinum solution. This demonstrates significant difference in the effectiveness of platinum extraction in fungi and plants, which are capable to accumulate platinum even when supplied at lower concentration (<500 ppb). It also shows different physiological pathways of platinum and other elements which are easily accumulated in the fruitbodies of the same species.

The ability of hydroponically cultivated Indian mustard plants (Sinapis alba L.) to accumulate platinum was investigated. The Pt-bioaccumulation in leaves, stem and shoots of plants growing for 2 and 4 weeks at Pt-concentration of 50 and 500 ěg/L was compared. The relation between dry and fresh weight was also estimated. Adsorptive stripping voltammetry (AdSV) and mass spectrometry with inductively coupled plasma (ICP-MS) were applied for determination of Pt. Increasing Pt-concentration from 50 to 500 ěg/L in the medium causes: (1) reduction of the root tissue hydration level at unchanged modification in aboveground parts of the plants and (2) decrease of the Pt transfer factor (TF) for roots and increase for leaves and stem. Duration of the culture influenced on Pt-accumulation in roots and in aboveground organs of mustard plants. Transfer factor for Pt between 560 and 1600 makes Indian mustard plants one at Pt-hyperaccumulators. Distribution of Pt-bioaccumulation in the plant organs may be useful for biomonitoring of platinum in the environment.

Activities of caesium in the mushrooms collected at different localities in Poland and Slovakia have been compared. Discrimination factor, defined as [(Bq.kg 1 137Cs in caps)/(Bq.kg 1 40K in caps)]/[(Bq.kg 1 137Cs in stipes)/ (Bq.kg 1 40K in stipes)], was used to explain mechanisms of uptake and transport of radiocaesium in fungi. The collected specimens were divided into caps and stipes. Activities of 137Cs and 40K were measured using a multichannel gamma spectrophotometer with HPGe(Li) detector. The highest accumulation of 137Cs was found in the samples of Xerocomus badius, Suillus luteus and Tricholoma equestre (2.7, 1.9 and 1.2 kBq.kg 1, respectively). T. equestre and S. luteus proved to hyperaccumulate caesium since 137Cs levels in the caps were two orders of magnitude higher than in the soil while only one order higher in the case of X. badius. Transport of 137Cs from stipe to cap in fruitbody is directly related to K concentration with lack of similar dependence in the case of transport from soil to cap. There is no dependence between activity of 137Cs in the analyzed fruitbodies and its activity in the soil, which makes mushrooms controversial bioindicators

The 60Co and 137Cs bioaccumulation by Helianthus annuus L. was measured during 9 day cultivation at 20 š 2°C in hydroponic Hoagland medium. Previous starvation for K+ and for NH4 + 2.2 and 2.7 times, respectively, enhanced 137Cs uptake rate. Previous cultivation in surplus of K+ ions 50 mmol.l 1 has no effect on 137Cs bioaccumulation rate. Both 137Cs and 60Co bioaccumulation significantly increase with dilution of basic Hoagland medium up to 1:7 for caesium and up to 1:3 for cobalt followed by mild decrease at higher dilutions. Root to shoot specific 137Cs radioactivity ratio (Bq.g 1/Bq.g 1, fresh wt.) increased with dilution from 1.46 to 9.6-9.8. The values root to shoot specific radioactivity ratio for 60Co were less dependent on the nutrient concentrations and were within the range 5.7 to 8.5. 137Cs was localized mainly in young leaves (30%) and roots (39%) and 60Co mainly in roots (67%) and leaves (20%). Obtained data showed less sensitivity of 60Co uptake by sunflower on nutrient concentration in hydroponic media.

In order to explain influence of common cations (K+, Na+ and Ca2+) on uptake and transport of caesium in macromycetes, a culture of a model mushroom species, king oyster mushroom (Pleurotus eryngii) was set up. Fructification in a growing chamber with stabilised temperature (18°C) and humidity (80%) was preceded by mycelial colonization of the sterilized barley seed medium packed into autoclavable plastic containers. Aliquots of test solutions, containing 0.1 mM caesium chloride carrier traced with 137CsCl and the selected ions, were dosed into the interphase between the container wall and the spawn block. This allowed to study influence of the added ions on the uptake of caesium in a way unaffected by the used growing medium, e.g. soil, as it was in the previous studies. The experiments demonstrated that the major amount of radiocaesium was biologically bound and accumulated in the fruitbodies to a higher extent (56 69%) than in the mycelium. Addition of 10 mM Na+ decreased the transfer factor for caesium (cap/soil) while addition of Ca2+ caused an increase of this value. The effect of potassium addition depended on its concentration in the solution. Also the Cs/K ratio in caps was significantly influenced by addition of 10 and 100 mM Na+. However, the Cs/K ratio in stipes was affected by Ca2+. Discrimination factors, calculated from specific activities (137Cs/40K cap d.w.)/(137Cs/40K stipe d.w.), were also changed after addition of the studied cations. Since the activities of caesium measured in the fruitbodies of single fungal species strongly depend on the content of co-supplied ions, further proofs should be achieved before using mushrooms as bioindicators of the soil caesium contamination.

A simple model of Pleurotus eryngii mushroom culture, grown under stringent laboratory conditions, was developed to watch ecophysiological pathways of xenobiotics in saprophytic fungi. The investigated substances may be added in different stages of biological cycle of the fungus. It is emphasized that to obtain the fruitbodies, all the physiological needs of the species have to be fulfilled, i.e.: nutritional requirements, optimal temperature (according to the biological cycle), humidity, aeration (oxygen and CO2), absence or presence of the light in each reproduction phase, as well as the control of infections and plagues through all the production stages. The described model serves for investigation of radionuclide and heavy metal uptake and transfer in fungi. Double or some multiple fructification from the same substrate is possible giving a possibility to investigate bioremediation by mycoextraction.

The constitution of the German Environmental Specimen Bank (ESB) has started in 1985, subsequent to a successful pilot study concerning the feasibility. Since that time, a multitude of technological and methodical standards have been developed, which allow for a high quality of the storage-samples and of the specimen characterization. While the storage-samples are kept for retrospective analysis, by now, already comprehensive data on the material-developing in the environment are available due to a real time monitoring of selected environmental chemicals over a period of up to twenty years. Thus, spatial and temporal trends can be described. Since the state of knowledge on critical tissue concentrations in the sublethal range is extremely low at present, it is however not possible to accomplish a direct assessment of relevancy of the substance concentrations. Hence, within the scope of the German ESB Program, the following strategies on assessment of relevancy are observed: use of biomarkers, histopathological examinations, biometric specimen characterization, use of ecological indicator groups, and development of a reference system with analytical and biometric data. Thus, for example endocrine effects in male breams in the river Saar, which correlate directly to operational discharges from municipal sewage plants, could be detected. By histopathological examinations, fibrotic and necrotic tissue adaptations on the gonads had been ascertained cumulatively, which unambiguously imply a restricted fertility of the male breams. In the river Rhine, an improved growth along the timeline could be described on the basis of biometric characterization of breams, which is regarded as rate for the reaction to all structural and material changes in the water body.
