Foodborne microorganisms harbor and adheres itself to the food material and surrounding surfaces for a long time and influence the food quality and consumers health. Among these microbes the presence of Enteric indicator bacteria in food premises confers the indication of entero-pathogens, i.e. E. coli, Salmonella, Shigella and Campylobacters that could cause severe systemic infections in consumers. In this study, the hygienic status of confectionery and supplementary food processing facility was evaluated. A total of 10497 examinations were performed on 3499 swab samples collected from food premises and handlers for the analysis of Enteric indicator bacteria. From swabs, 1277 (12.2%) isolates were identified in which Enterobacteriaceae were found with higher frequency 604 (47.3%) followed by Coliforms 293 (30.8%) and Escherichia coli 280 (21.9%) respectively. The mean count (CFU/cm2) was found maximum for plain surfaces (floors, walls and door), while the lowest was for equipment and machinery. Overall isolates percent prevalence was determined where Enterobacteriaceae were 47%, Coliforms 31% and Escherichia coli 22%. Majority of the floor surfaces were highly contaminated, where washing and sanitation practices were observed to be inappropriate. Worker hygiene status was lacking essential food safety and hygiene standards. In general, the Enteric bacteria were found with higher ratio, that could affect the food quality and quantity both to a greater extent with some influences on consumers health.

Urinary tract infections (UTI’s) are a bacterial infection that affects any part of the urinary tract. Urinary tract infection is one of the major diseases that affect people of all age groups and sexes and can be separated into asymptomatic and symptomatic. The normal urinary tract is sterile but gets infected with the normal flora and opportunistic pathogens. Urinary tract infections (UTIs) are a serious health problem affecting millions of people each year. This is the second most common type of infection in the body. In the present study 20 morphologically different bacterial isolates were recovered from 50 different urine samples from UTI’s patient. The prevalence of the isolates was varied with E. coli (40%), followed by Staphylococcus spp., Pseudomonas spp. and Klebsiella spp. each of (20%) respectively. All the isolates of different bacteria were tested for antibiotic sensitivity patterns against common antibiotics viz. Ampicillin, Tetracycline, Chloramphenicol, Vancomycin, Nitrofurontione and Ciprofloxacin with potencies of 30 μg each. The sensitive pattern of the isolates towards antibiotic Nitrofurontoine was (75%) followed by Tetracycline (55%) and Ciprofloxacin (45%). Intermediate activity was reported against Ampicillin (45%), Chloramphenicol and vancomycin (40%). Resistant pattern shown against vancomycin (60%), Chloramphenicol (45%), while both tetracycline and Ciprofloxacin shown (30%) against the UTI’s isolates. Therefore, drugs like Nitrofurontoine, Tetracycline and Ciprofloxacin are the most effective choices against the common UTI’s isolates.

Optimizing codons of gene of interest has offered a value-added way to increase heterologous expression of proteins. Different algorithms have been designed to achieve this purpose. These algorithms make use of parameters such as codon Adaptation index (CAI), codon context (CC), percentage guanine-cytosine (GC) content and RNA instability for predicting optimal codons responsible for increased protein expression. Lipase 3646 is an enzyme with great potential for industrial applications. This enzymes has been described to possess thermostable properties with relative stability in high alkaline pH and at different concentrations of organic solvents and inhibitors. This research therefore used JCat, Codon optimization online (COOL), presyndocon and ExpOptimizer algorithms to predict expression level of lipase 3646 enzyme in silico by optimizing its gene coding sequence. The results showed that there were variations in the CAI generated by the algorithms for the same 3646 DNA sequence which suggests that each algorithm is specific for its own generated CAI. COOL algorithm prediction based on other parameters showed good results for potential expression of the lipase. Thus, we recommend COOL algorithm for codon optimization of the lipase 3646 gene for industrial applications.

Consumption of water contaminated with pathogens and heavy metals is responsible for the demise of millions of people around the globe. To find a low cost, facile and benign route for developing a biosorbent capable of handling the challenges associated with unsafe drinking water, the study report the preparation, activation, adsorption and removal of cadmium and pathogenic organisms from water samples. Modified plantain peel biochar (MPPB) was characterized using Braunauer-Emmett-Teller (BET) surface area and X-Ray diffraction (XRD). Surface area and pore size of the MPPB determined from BET surface area plot are 8.79 cc/g and 16.69 m2/g respectively with XRD crystallite size of 14.56 nm. The maximum adsorption capacity was observed to be 366.300 mmolkg-1. Equilibrium isotherm indicated the coefficient of determination to increase from Jovanovich →Elovic→Temkin→Freundlich→Langmuir model. Surface interaction and mass transfer kinetics shows a mixture of intra-particle diffusion and mass transfer process. The adsorption kinetics and surface interaction models coefficient of determination increased from Bangham pore diffusion →mass transfer model→Liquid film diffusion→pseudo first order→Dunwald-Wagner→Elovic→Pseudo-second order→Weber and Moris model. The adsorption indicated a chemisorption process with rate limitimg step likely to involve electron pair donor-acceptor interactions, hydrogen bonding and dipole dipole interactions. Antimicrobial evaluation of MPPB showed that it is a potential antimicrobial agent against common micro-organisms.

This paper presents the synthesis and antibacterial studies some novel halogenated Platinum (II) complexes of active Schiff’s base ligands of the type [Pt(L1H)2]Cl2, [Pt(L2H)2]Cl2 and[Pt(L)2] derived from 5-Bromo Isatin. Where L1H= hydrazinecarbothiamide of 5-bromoindolinedione and L2H = hydrazinecarboxamide of 5-bromoindolinedione. All complexes reported here had been characterised by 1H NMR and IR spectral studies. Molar conductance studies support the 1:2 natures of these complexes. Pt (II) complexes are diamagnetic in nature and showing square planner geometry. The IR spectral data reveals that both the Schiff’s bases (L1H and L2H) behave as bidentate ligands and are co-ordinated to Pt (II) metal through the sulfur and hydrogenic nitrogen atom.all the new synthesized compounds were screened for antibacterial activity against four of the test organisms like Bacillus pumilus, Micrococcus Lutes, E. coli and Salmonella abony

The survival of potentially infectious enteropathogens in digestate from two-phase mesophilic anaerobic digestion (Two-phase MAD) technique raises concern on handling and application of the resulting digestate as far as hunan health and environment safety is concerned. In this research, temperature validation and hygienization potential of Two-phase MAD operated as a completely stirred-tank reactor with semi-continuous feeding was investigated. High enteropathogens survival up to 6.4 log E. coli and 6.1 log Salmonella Senftenberg was observed in digestate resulted from anaerobic digestion of potato peels artificially spiked with 8.7 log Salmonella Senftenberg and 8.8log E. coli simulating extra high pathogens contamination in biowaste used as feedstock. Even though an average decrease in pH by 2.3 units from the original value of 6.5 observed in hydrolysis reactor created unfavorable environment on the survival of both E. coli and Salmonella Senftenberg causing a 3.6 log and 3.8 log reduction, yet the alkaline condition in the methanation reactor pH 7.7 boosted the surviving rate by 1.3 log in E. coli and 1.2 log in Salmonella Senftenberg confirming the inefficiency of Two-phase MAD set-up at providing a completely hygienized digestate. This renders the biosafety of the resulting final digestate questionable and limit the possibility of farmapplication for nutrients recycling due to the involving environmental threat and health risks in case the digestate is to be used. Temperature-time combination at 65 °C for 30min was confirmed sufficient at completely elimination of E. coli and effecting >1 log cycle die-off equivalent to 90% die-off in Salmonella Senftenberg with z-Value of 11 ºC validating the temperature-time regime of 70 ºC for 60min proposed in European Commissions Regulation (EC) No. 208/2006.

The austenitic cold-rolled AISI 316L stainless steel was used for the studies. Corrosion resistance measurements were performed on the samples after three types of treatments: abrasive finishing (MP), standard electropolishing (EP), and magnetoelectropolishing (MEP). They were carried out in Ringer's solution at a room temperature, indicating a considerable difference in the breaking potential Epit values, dependent on surface treatment. Two groups of samples, those after EP and MEP, were submerged in broth culture wild strain of Escherichia coli from contaminated river water. Images of the steel samples submerged for 2 and 4 hours are displayed in the paper. A statistical approach has been performed. In each case the number of bacteria deposited on the MEP steel samples was higher in comparison with that one noticed on EP samples, increasing much faster with time on the MEP ones.

Bacteria are capable of invading and infecting humans, leading to disease and sometimes death. Different body tissues, organs and systems are vulnerable to different organisms. This study was aimed to isolate and identify the bacteria causing Urinary Tract Infection (UTI) in different age groups and also screened for ESβL production. The mid-stream urine samples showing symptoms of urinary tract infections were collected in a wide container from different hospitals and diagnostic centres of Kalaburagi city. The isolation of uropathogen was done by semi-quantitative method of inoculating the samples on the selective and differential media such as Eosin Methylene Blue (EMB) and MacConkey (MAC) agar media respectively. The isolated pathogen was identified by conventional methods like cultural, morphological and biochemical tests. The antibiotic susceptibility test was carried out by Kirby-Bauer disc diffusion technique and ESβL production by double disk-diffusion test (DDDT) as per CLSI guidelines (2013). Out of 550 samples screened, a total of 288 bacteria were isolated of those half of them (146) were E. coli isolates and all were found to be multidrug resistant and half of them (50%) were of ESβL producers. All ESβL producing E. coli isolates were resistant to ceftazidime and exhibited higher level of resistance to Cephalothin, Erythromycin, Cotrimoxazole and Aztreonam. As ESβL producing organisms limits the available treatment options, so, the timely administration of sensitive antibiotic and avoiding antibiotic abuse will help in curing the disease without going for the costly drugs such as Carbapenems. Our study revealed that nitrofurantoin, ofloxacin and cefoxitin may be considered as drug of choice for the treatment of UTI patients. The ESβL production in uropathogens should be continuously monitored in the clinics and hospitals as to avoid the emergence of more multi drug resistance strains.

The purpose of this research is to determine the best concentration and effect of garlic extract incorporated on tilapia skin gelatin-based edible coating to preserve and extend the shelf life of fish meatball at cold storage. This research was conducted from May 2018 until October 2018 on Fisheries Product Processing Laboratory of The Faculty of Fisheries and Marine Sciences, Universitas Padjadjaran. The garlic extract conducted with maceration method with ethanol 96% was performed at Organic Chemical Laboratory FMIPA Universitas Padjadjaran, and for TPC test, inhibition test of edible coating Tilapia’s fish-skin with garlic extract was conducted in Microbiology and Molecular Biotechnology Laboratory of Faculty of Fisheries and Marine Sciences Universitas Padjadjaran. The method used is experimental method with 5 treatments which is meatballs without edible coating. The addition of garlic extract was done with various concentration at 0%, 1%, 1.5%, 2%. The observations were Total Plate Count (TPC) test, inhibition zone test against Pseudomonas aeruginosa, and pH value. The results showed that the best treatment is 2% garlic extract with TPC value 1.1×105 cfu/g during 14 days of storage and concluded that it could extend the shelf life of meatball until 14 days, pH value was 7.03 and inhibition zone against P. aeruginosa was 7.01 mm. The addition of garlic extract on tilapia skin gelatin-based edible coating could extend the shelf life of fish meatballs until 14 days on cold storage.

Rift Valley fever (RVF) disease is an emerging viral zoonotic disease caused by Rift Valley fever virus (RVFV). RVF disease is seemingly becoming more severe in endemic populations and in areas of new outbreak. RVF disease is listed by WHO as requiring urgent research and development attention. There is no therapeutics or licensed vaccine for human use in the case of an eventual outbreak. There is a need to clone RVFV genes in cloning vectors which may be useful to produce antigens in plant cells or may be tested directly as DNA vaccines. A synthesized Rift Valley fever virus fusion gene was cloned in a bean yellow dwarf virus derived replicon vector; PBYR2e3K2Mc-GFP by removing and replacing the GFP gene. The cloned PBYR2e3K2Mc-RVFV fusion gene was confirmed by PCR, restriction digestion and DNA sequencing. This cloned RVFV fusion gene in plant virus vector can be used for subsequent protein expression in plants cells or used directly tested as DNA vaccines in future studies.
