There are many methods for detection of Cryptosporidium oocysts. Most of them (more than 20) enable the microscopic detection of Cryptosporidium oocysts in faecal smears. Such a great variability of diagnostic methods may lead to confusion as far as the choice of an appropriate technique by a given laboratory is concerned. This study evaluated the diagnostic usefulness of Cryptosporidium oocysts and coproantigen detection methods in the diagnosis of cryptosporidiosis in human (266 stool specimen) and animals (205 from cattle, 160 from sheep, 30 from horses, 80 from cats, 227 from dogs and 11 from wild animals). The total number of human and animal stool specimens processed was 266 and 713, respectively. In this study the usefulness of several diagnostic methods was compared. The following techniques were taken into account: wet mounts, hematoxylin staining, four different specific methods (modified Zeihl-Neelsen, Kinyoun's, safraninmethylene blue, as well as carbol-methyl violet and tartrazyne) and commercially available kit based on enzyme-linked immunoassay (ProspecT(r) Cryptosporidium Microplate Assay). The final number of positive specimens was 123. Out of them 77 were positive in all specific methods. The oocysts found in stool specimens were measured. Humans were infected with C. parvum and animals with C. parvum, C. andersoni or C. felis. The statistical analysis has shown that EIA test was a better than microscopy method for identification of Cryptosporidium in faecal samples in human and wild animal. Sensitivity and specificity are important factors for the choice of a proper diagnostic method for Cryptosporidium detection, however other factors such as cost, simplicity and ease of interpretation of results are also important considerations.

Opis:
The eosin colour test estimating the viability of cysts was compared with the method of in vitro excystation on agar substrate (NNE). Single cysts were studied, isolated from populations stored under sterile conditions in water at 4°C for a period ranging from a few days to 16 years. It has been found that the inoculations of single cysts (805 cysts) on the NNE substrate allow a precise determination of the percentage of living cysts. All the cysts stained with eosin, incubated on such a substrate proved to be dead, while the non-staining cysts ex-cistated with varying percentage. Thus, among the non-staining cysts of Acanthamoeba after 2-4 years of storage at 4°C 89.2% excystated while the remaining ones were dead. After 16 years of storage only 29.8% nostaining cysts excystated. Very similar values (89.6 and 28.5% respectively) were obtained when determining the percentage of living cysts with the colour test. The eosin test has proved to be a good method of the estimation of the viability of cysts of Acanthamoeba.

Tetrazolium salts are widely used as indicators of metabolic activity for both eukaryotic and prokaryotic cells. Live cells reduce the tetrazole ring in MTT or XTT salts and then a colored formazane formed can be assessed spectrophotometrically. Despite widespread use of MTT/XTT reduction tests biochemical mechanisms of the reaction are still unknown, and each test application case requires standardization of experimental conditions. In the present study we tested in vitro the utility of both MTT and XTT salts to determine the influence of selected extracellular agents for T. gondii tachyzoites and their host cells (i.e. mouse L929 fibroblasts). The results showed that MTT is reduced more intensively than XTT by host and parasite cells. The attenuation of T. gondii tachyzoites resulted in a decrease of reduction level of both tetrazolium salts, particularly of XTT. Using MTT we found also that T. gondii is not susceptible to extremely toxic substance, sodium azide. Our results confirmed a high usefulness of MTT reduction tests in numerous studies on eukaryotic cells.

Opis:
The studies were carried out in a pig farm and in a dairy cattle farm in South Poland. Thuridan was applied as 0.3% suspension in 3 replications in 7 days interval. The efficacy of Thuridan was established using fly -catchers which were hung on the same places – once before and three times after Thuridan was used. In the animal houses 86% less flies were found.
