Opis:
Species composition of yeast-like fungi was investigated in 2001-2003 years. Four species from genus Candida were found: Candida albicans, Candida aquatica, Candida krusei and Candida tropicalis. The occurrence of yeast was investigated, using bait and the Sabouraud methods in the fresh waters of the six springs: Antoniuk, Cypisek, Sobolewo, Smolniki, Pólko and Rybnik of Podlasie Province. The fungi were found to be the most common at eutrophized sites of the spring Sobolewo. The spring Sobolewo, as compared to remaining springs, had a considerably higher concentrations of ammonium nitrogen, phosphates, and suspended substances.

The study comprises an analysis of the growth rate of C. albicans and C. stellatoidea strains isolated from the respiratory system and incubated on Nickerson's and Sabouraud's agar in the dark and by light. The property to form hyphae and pseudohyphae by both species was used to evaluate the growth rate. The increase in their growth allowed to isolate the stages characteristic for growth and development of most fungi. Disturbances appeared with the fungi exposed to light which is the factor definitely setting back the growth. Faster growth and development was observed with C. stellatoidea.

Opis:
Mycological investigations were carried out in 20 patients due to the delayed wound healing, following orthopaedic surgery at the Department of Orthopaedics, Medical University of Łódź. The material samples for fungal assessment were taken from the wound, surrounding skin and oral cavity. In all patients, the fungi were isolated at least from one ontocenosis; from the wound in 40.0% of subjects studied, from the wound-surrounding skin in 55.0% and from the oral cavity in 85.0%. It should be emphasized that the occurrence of fungi was more frequently observed simultaneously in two or three ontocenoses (70.0%) than in one (30.0%). Unifocal infection was found only in the oral cavity, while the presence of fungi in the wound was accompanied by their appearance in the region of wound-surrounding skin and/or in the oral cavity.

Candida glabrata is very important pathogen which is resistant to some imidazoles (fluconazole). The aim of study was to compare a susceptibility of Candida albicans and Candida glabrata - strains isolated from patietnts treated in Centre for Treatment of Parasitic Diseases and Mycoses - to miconazole using MIC. 96.5% of strains were wrażliwe to miconazole; values of MIC for C. albicans were lower than for C. glabrata.

112 strains of Candida albicans were isolated from oral cavity and ontocenoses of the upper digestive tract (endoscopy) of children (age: 5-17) with gastrointestinal disorders. Axenic strains were differentiated with API 20C AUX and API ZYM tests (bioMerieux). Then enzymograms and biotypes were determined for all the strains based on the activity of 19 hydrolases. The highest activity was noted for: e₂ - phosphatase alcaline, e₆ – leucine arylamidase, e₁₁ - phosphatase acid, e₅ - lipase (c₁₄); e₇ - valine arylamidase, e₁₂ - naphtol-AS-BI-phosphohydrolase, e₁₆ - α-glucosidase and e₁₈ - N-acetyl-ß-glucosamidase, and the latter four were used for biotyping procedures. Our own system was based on the mathematical binominal distribution formula (1: 4: 6: 4: 1): all „+”; one „ -” three „+”; two „-”, two ,,+”; tbree „-”, one „+”; all „-”. We bave found the following biotypes: A (16.1 ± 3.5%), B₁ (2.7 ± 2.53%), B₃ (8.0 ± 25%), B₄ (22.3 ± 3.9%), C₂ (1.8 ± 1.3%), C₃ (7.1 ± 2.4%), C₆ (30.4 ± 4.3%), D₃ (11.6 ± 3.0%).

The yeast-like fungi are typical opportunistic microorganisms which may be a part of normal commensally ontocenosis. Many species of Candida genus may be also a cause of candidosis in human and animals. Clinical symptoms occurred customly in patient with cellular immunity deficiencies. Horizontal (person-to-person) form of transmission is also possible, including family infection or group infection in animals, mostly in birds. According to actual data, zoonotic status of candidosis is still not fully documented. In animals, candidosis more often occurs as endogenic infection. The separate problem is infection of animals with significance suppressed immune system, e.g. with tumor diseases. In necroscopy investigation very often in these animals disseminated candidosis has been observed. On different way the infections of Candida spp. are manifested in birds, when Candida albicans occurs as marginal population of normal intestinal ontocenosis. During stress the intestinal fungal population is growing up for critical value and typical crop candidosis is developing. In our routine diagnostic work we have isolated Candida strains from nutritional tract in laboratory mice. This yeast were isolated also from the bottom of the boxes and from mice faeces.

Increasing use of elctromagnetic fields (EF) in the treatment of various diseases may have potential impacts on fungi - possible aetiological factors of concomitant mycoses. The aim of this study was to investigate the effects of EF on miconazole susceptibility among fungi of the genus Candida showing confirmed pathogenicity in humans. Fifteen Candida strains obtained from patients were used and their susceptibility to miconazole was determined by diffusion in agar gel. Antifungal activity of miconazole was assessed as minimal inhibitory concentration (MIC) which was calculated for Candida strains not exposed to EF (control) and EF-exposed (3 experiments). In the majority of cases the susceptibility of Candida species to miconazole decreased (higher MICs) after the first week of EF exposure, regardless of the EF's parameters. This was followed by an increase in susceptibility (lower MICs) after the second week of exposure to EF of 2 mT intensity and frequency 3 Hz (experiment I) and an EF of 9 mT intensity and 12.5 Hz frequency (experiment II) relative to control. An increase in susceptibility (lower MICs) was observed in the second week of exposure, whatever the parameters of the EF. The application of low intensity, low frequency EF for a period of at least two weeks may be beneficial in the treatment of mycoses caused by pathogenic fungi of Candida genus.

The aim of the work was to compare adhesive properties of Candida albicans strains isolated from the oral cavity in HIV+ vs. HIV- persons. The materials were Candida albicans strains and buccal epithelial cells isolated from both HIV+ and HIV- persons. We applied the in vitro adherence test, primarily described by Kimura and Pearsall and modified by Macura. The strongest adherence was found between both fungi and epithelial cells isolated from a HIV+ person. The adherence of C. albicans isolated from HIV+ patients was significantly stronger to epithelium collected from HIV+ than HIV- persons.

The purpose of this study was to determine the in vitro susceptibility to 6 antifungal agents of 143 strains of Candida species isolated from 545 patients with suspicion of fungal infection. Test ATB Fungus (bioMerieux) was used. Among all Candida isolates 91.6% were susceptible to tested polyenes and 32.9% to all azoles. Less susceptible to polyenes isolates were noted in low percentage (0.7 to 4.2%) and to azoles more frequcntly (32.9 to 41.2%). Candida isolates resistant to antifungal agents tested were dependent from the kind of clinical samples and species of fungi. Monitoring of the susceptibility to antifungal agents by ATB commercial test seems to be uscful tool for therapeutic purposes.

Opis:
Early detection and correct diagnosis of fungal infection is important for further therapy and prophylaxis. Currently, there are a number of proposals related to rapid diagnostic tests. To those we can include chromogenic media, such as CHROMagar® Candida (GRASO). This medium has been developed to differentiate four species of genus Candida. Its use in laboratory has opened up possibilities for growing not only the genus Candida, but also other fungi. The study suggests that medium CHROMagar® Candida can provide an excellent pre-test, or be used as complementary to biochemical tests. Establishing a full template of colours would enable more efficient and fuller use of the medium properties, especially in the discrimination of the teleomorphic form from anamorphic.

We have studied the capability of dissociation of 22 L-form aminoacids by 27 strains of fungi belonging to species: Candida, Cryptococcus, Geotrichum, Rhodotorula, Saccharomyces, Trichosporon. Fungal strains were cultured on agar medium with phenyl red and studied aminoacid. Disamination of an aminoacid by a fungus resulted in alkalisation of the medium and change of its colour from yellow to pink. Strains from genus Trichosporon and Geotrichum decomposed the most aminoacids (15-16), while fungi from genus Candida decomposed less aminoacid (1-8). Strains form species Candida famata, C. kefyr, C. lambica. C. rugosa, C. neoformans, R. mucilaginosa and S. cerevisiae did not show any ability to desaminate any of the 22 aminoacids used in the study. Aminoacids most frequently disaminated by the studied strains of fungi included acidic aliphatic aminoacids and their amides: L-aspartic acid, L-asparagine, L-glutaminic acid, L-glutamine and neutral aliphatic aminoacids: L-glycin, L-alanine, L-serine. Aromatic and sulphur aminoacids (L-cysteine, L-cystine) were not decomposed by the studied fungal strains.

The research was done in the dermatological laboratory of Bytom Hospital No 2 in the years 1979-1983. 31 842 women were examined at the age 16 to 82. Samples were taken from urethra and neck of the uterus with sterylized oese. Both direct slides in 0,9 % NaCl solution and permanent slides were made. The latter were dyed by means of Gram's method in order to establish identity of C. albicans and by means of Giemsa's method to identity T. vaginalis. During 5 years the cases of trichomonadosis and candidosis recorded in the years of 1982 and 1983 were the most numerous. Seasonal fluctuation of trichomonadosis in 1980 and 1983 years and of candidosis in 1970, 1980, 1983 are also presented.

The objective of the study was the analysis of occurrence and number of Candida in the alimentary tract of 209 patients with intestinal amoebiasis considering their state after the treatment. Amoebiasis was diagnosed on the grounds of Entamoeba histolytica present in the faeces, what has just recently been recognised equal to the presence of E. histolytica complex. In all patients serological investigations were performed using indirect haemagglutination (IHA) test. The cases with relapses or with the intestinal ailments after elimination of the parasite from the body at the simultaneous isolation of Candida in the faeces were focused on. In the compass of several years a wide number of qualitative and quantitative mycological tests were performed comprising 39 patients with relapses after symptomatic amoebiasis and 42 with ailments which had appeared after the treatment form asymptomatic amoebiasis. Observation of the dynamics of number of Candida cells revealed most often their increment in subsequent examinations. It occurred in 69% people examined after the treatment of asymptomatic antoebiasis and in 40,9% after the treatment of symptomatic amoebiasis. The differences obtained were statistically significant. The number of yeast remained at the same high level in 23,8% patients after asymptomatic amoebiasis and in 36% after symptomatic amoebiasis. Drop, of Candida cells number was noted in 7,1% people after asymptomatic amoebiasis and in 11,4% after symptomatic amoebiasis. In the light of the results obtained it turns possible that suffering from amoebiasis could even in the long run affect Candida tendency to proliferate or the tendency to remain at the same high level. It may be presumed that the growing number of yeast was an important reason for the intestinal ailments after the elimination of E. histolytica. The results of mycoserological investigations also were taken into consideration.

The study included 52 children. During endoscopy the contents of oesophagus, stomach and duodenum were collected with catheters, and additionally washings from the oral cavity and anal swabs or samples of faeces. Materials were inoculated on Sabouraud's media. The axenic strains of fungi were identified using own methods and others tests (API 20C, API 20C AUX). The activity of ketoconazole (K) was measured against 100 strains of Candida: C. albicans (91 strains), C. kefyr (4), C. famata (2). C. tropicalis (1), C. guilliermondii (1), C. glabrata (1). The strains susceptibility to K (RO4 1400 Janssen) was estimated with the own system based on the dose - response curves; the minimal inhibitory concentration (MIC) values were canculated. The low values of MIC indicate high susceptibility to K of examined Candida strains.
