Opis:
The purpose of examinations was defined role of environment of school in interindyvidual transmisions. The yeast and yeast - like fungi isolated from the oral cavity, troat and nose from 270 healthy children, at the age of 6 to 15 years, from country and municipal schools. The frequency of occurrence of fungi in country was higher than in minicipal schools by the resambling biodifferentiation. Totally 13 species belonging to 5 genera were detected in the examined material: Candida, Rhodotorula, Saccharomyces, Saccharomycopsis i Trichosporon. Increasing part of S. capsularis was noted in ontocenoses of healthy children. The qualitative and quantitative changes testify about changes of environment. Simultaneously, they inform about timely condition of mycoflora of the examined ontocenosis and surroundings.

Mycotic infections are an increasing diagnostic and therapeutic problem in pathology of the alimentary tract. The work shows the results of research on the large intestine candidosis. The following invasive factors were isolated: Candida albicans, C. guilliermondi, C. pseudotropicalis, C. tropicalis. Rectoscopic views were described. In severe C. albicans infections HIV test were performed. Four patients were HIV positive.

Many recent epidemiologic and experimental data showed immunomodulatory action of several helminths. Of particular interest have been the results demonstrating that helminths infections are able to inhibit the development of autoimmune processes and pathology in animal experimental models, and probably in humans. The article presents and discusses selected aspects of this novel therapeutic approach which seems particularly important for combat a dramatic rise in the incidence of allergy and autoimmune diseases in the West.

The objective of the study was the analysis of occurrence and number of Candida in the alimentary tract of 209 patients with intestinal amoebiasis considering their state after the treatment. Amoebiasis was diagnosed on the grounds of Entamoeba histolytica present in the faeces, what has just recently been recognised equal to the presence of E. histolytica complex. In all patients serological investigations were performed using indirect haemagglutination (IHA) test. The cases with relapses or with the intestinal ailments after elimination of the parasite from the body at the simultaneous isolation of Candida in the faeces were focused on. In the compass of several years a wide number of qualitative and quantitative mycological tests were performed comprising 39 patients with relapses after symptomatic amoebiasis and 42 with ailments which had appeared after the treatment form asymptomatic amoebiasis. Observation of the dynamics of number of Candida cells revealed most often their increment in subsequent examinations. It occurred in 69% people examined after the treatment of asymptomatic antoebiasis and in 40,9% after the treatment of symptomatic amoebiasis. The differences obtained were statistically significant. The number of yeast remained at the same high level in 23,8% patients after asymptomatic amoebiasis and in 36% after symptomatic amoebiasis. Drop, of Candida cells number was noted in 7,1% people after asymptomatic amoebiasis and in 11,4% after symptomatic amoebiasis. In the light of the results obtained it turns possible that suffering from amoebiasis could even in the long run affect Candida tendency to proliferate or the tendency to remain at the same high level. It may be presumed that the growing number of yeast was an important reason for the intestinal ailments after the elimination of E. histolytica. The results of mycoserological investigations also were taken into consideration.

In mycoses congenital – nonspecific innate as well as acquired immunity (involving neutrophiles, monocytes, macrophages, dendritic cells and lymphocytes) both play important roles in host defence. Th1 lymphocytes release cytokines (IL-2, IL-12, IFNg) and stimulate cytotoxic cells and neutrophiles to destroy fungal cells. Th2 lymphocytes, on the other hand, suppress cellular immunity by releasing the cytokines IL-4, IL-6 and IL-10 which counter regulate the secretion of IL-2, IL-12, IFNg and depress the activity of macrophages. Cellular mechanisms play essential roles in host responses to fungal infections. Dysfunction of T lymphocytes and a reduction in their number are typically observed in patients with mycotic diseases. There occurs a reduction of both T lymphocyte populations and the T-helper to T-suppressor cell number ratio, and these are of critical importance in explaining the diminished IgA production and enhanced adhesion of fungal cells to the surface of host cells as well as in facilitating the intrusion of fungi throughout the skin and mucous membranes. The specific immunological reaction, associated with the synthesis of antibodies against fungal cell wall or cytoplasmic antigens, is of little significance in protective immunity, but nevertheless has a rather important role to play in diagnosis as well as in supporting phagocytosis by inhibition of fungal cell adherence. In patients with mycoses, typically low blood serum level of the immunoglobulin class G and A and low sIgA in saliva are observed. A detailed understanding the nature and function of the immune system in mycoses is necessary to enable improvements in pharmacotherapy with antifungal antibiotics and chemotherapeutics, as well as to treatments based on immunotherapy and vaccination.

Analyse of funigi prevalence and their species in the digesive tract of adult persons and children. 1081 faecal sample were examined, 723 from adult persons and 358 from children. Fungal strains were recovered in 67.9% samples from adult persons and in 69.8% from children. There were found 8 species of fungi: Candida albicans, C. famata, C. tropicalis, C. krusei, C. glabrata, C. guilliermondii, C. kefyr, Saccharomyces cerevisiae.

Over the years 1996-1997 mycological examination were performed in 367 persons with cutaneous lesions, returning from the tropics. Fungal infection was diagnosed in 141 patients. Twenty nine (42.65%) of the cases were of single-focal, 32 (47.06%) of bifocal and 7 (10.29%) of multifocal nature. One hundred forty one isolates included 50 (35.6%) moulds 23 (16.3%) yeast - like fungi and 68 (48.1%) dermatophytes. Genus classification of the dermatophytes was as follows: Microsporum - 16 (23.5%), Epidermophyton - 15 (22.1%), Trichophyton - 37 (54.4%). Over the years 1996-1997 the following dermatophyte species were isolated: M audouini, M. ferrugineum, M canis, T. rubrum, T. mentagrophytes var. granulosum, T. tonsurans, T. violaceum, E. floccosum. Microsporum genus was isolated from persons returning from East, West and Central Africa, and from South Europe, Trichophyton genus was isolated from persons returning from Asia and South America.

The highly toxic organophosphorus compound (isopropylmethylphosphonofluoridate — IMPF) belonging to the group of the so called toxic of warfare agents can impair immune effector mechanisms determining the development of parasitic infection. Preliminary studies have demonstrated that IMPF delays elimination of adults Trichinella spiralis forms from the intestine of C57B1/6 mice and B6C3F1 hybrids. Besides, the studied compound modifies the proliferative activity of T-cells in the above mentioned mice and reduces the production of anti-SRBC antibodies in C57B1/6 strain. The mechanisms are discussed of the organophosphorus compound effect on neuroimmunological processes connected with antiparasitic immunity, paying attention also to the function of these processes by the parasitic infection itself, in host-parasite relationship. The probability is also considered of increased sensitivity to Leishmania infection in humans poisoned with IMPF and/or other organophosphorus compounds.

The aim of this study was to estimate the development of P. aeruginosa infection in mice infected with T. spiralis or T. pseudospiralis. The investigations were carried out on the following strains of mice: C57B1, C3H and B6C3F1-. 20 days post parasitic infection P. aeruginosa were administrated by inhalation or by intraperitoneal injection. After 4 hrs development of infection in mice was evaluated (the number of bacteria in lung or in the liver tissue were counted by plating). It was found that the bacterial infection developed with the highest rate in C3H mice followed by C57B1/6, in comparison with control mice. In C3H mice infected with T. pseudospiralis, the number of bacteria was higher in lung and liver tissue, in comparison with those infected with T. spiralis. In B6C3F1 hybrid the infection rate was significantly lower after intraperitoneal administration in T. spiralis infected mice. In our experiments, the development of P. aeruginosa infection in mice with trichinellosis was dependent on the strain of mice and the routes of bacteria administration.

Contemporary problems have been presented, related to pathogenesis, clinical pathology and epidemiology of leprosy. It has been stressed that intensity of cell mediated immune response in the patient determines not only resistance or susceptibility to infection with Mycobacterium leprae but also defines traits of clinical pathology in leprosy, thus providing the basis for modern clinical classification. Present aims and investigative methods in lepra epidemiology have been shown. Current data have been provided on spread of leprosy in various parts of the world, mainly those remaining under monitoring by the World Health Organization. The data point to significant decrease in registered leprosy cases in the regions beginning from 1986 and, in particular, in years 1990- 1994. The decrease has resulted from adequate verification of the disease stage in the patients and from applying the recommended multidrug therapy. The needs and perspectives of actions aiming at combatting leprosy have been described, aiming at eradication of leprosy as the international health problem till the year 2000. It has been stressed that at present, leprosy is no longer a hopeless disease and that it can be cured when sufficiently early diagnosed and properly treated.

The present study deals with the ecology of Lyme borreliosis in Northern Poland. The complexity of environmental conditions are: (1) the presence of certain rodent hosts for the Borrelia, (2) the seasonal patterns of abundance and infection rate of /. ricinus in the population of rodents and on vegetation, (3) the prevalence of B. burgdorferi s.1., B. garinii and B. afzelii. The longterm studies were carried out on a monthly basis from April to October (1998-2001) in woodlands at Urwitałt near Mikołajki on the Mazury Lakes. Two methods of spirochate detection were used: indirect immunofluorescence assay (IFA) and polymerase chain reaction (PCR). The dynamics of ticks infestation, differences in infestation rates between years, months, age and sex for two species of rodents (Clethrionomys glareolus, Apodemus flavicollis) were analysed using the software package Statgraphics version 7 and GLIM 4 (statistical system for generalized linear interactive modeling, Royal Statistical Society 1993). We were able to demonstrate that in selected habitat of Mazury Lakes the spirochaetal agent of Lyme borreliosis occurs quite frequently, both in rodents and in vector — /. ricinus. Comparing C. glareolus and A. flavicollis, each rodent species showed different, seasonal and sex dependent, host infection with Borrelia. Due to the courses of prevalence of infection, both for partially engorged ticks and questing ticks, the risk of human infection seems to be highest in spring. The infection rate of engorged larvae was much higher than in questing larvae. We conclude that both species of rodents, highly abounded in the Mazury Lakes district, may represent significant zoonotic reservoir for B. burgdorferii sl., B. garinii and B. afzelii.

The prevalence and abundance of Cryptosporidium parvum were studied over a three year period (1997-1999) in three species of rodents sampled from forest and abandoned fields in the Mazury Lake District, Poland. The overall prevalence was consistently higher in voles compared with Apodemus flavicollis (70.6% in Clethrionomys glareolus, 73.0% in Microtus arvalis and 27.8% in A. flavicollis). The prevalence and abundance of infection also varied across the 3 years of the study with 1998 being the year of higher prevalence and abundance of the parasite. Fewer older animals carried the infection, and their infections were relatively mild. We found no consistent pattern of seasonal changes despite the significance of seasonal differenccs. Host sex did not influence either the prevalence or abundance of infection with C. parvum. A great proportion of recaptured voles developed chronic infections between consecutive trapping sessions and only a small number of animals recovered. However, yellow-necked mice seem to be much more resistant to infection that became self-limiting. Our results firmly establish that the common woodland and grassland wild rodents in the Mazury Lake District constitute a significant and hazardous reservoir of C. parvum for animals and humans.

A clear stimulation of the activity of glutathione reductase (RG, EC 1.6.4.2), one of the major enzymes participating in glutathione metabolism, was observed in the skeletal muscles from Trichinella spiralis infected mice. The maximal, almost twofold increase in RG activity was observed in week 5 post-infection. Administration of albendazole, an anthelmintic widely used in trichinellosis treatment, resulted in an additional, about 40% stimulation of RG activity (small, but statistically significant) on week 2 post-infection. In weeks 6 and 7 postinfection, when no stimulation of RG activity in infected mice caused by drug was observed, the drug modified the structure of that allosteric enzyme, affecting the kinetics of its substrate saturation in this way that the shape of the substrate saturation curve changed from „double sigmoidal”, typical of this enzyme kinetics in the muscles from infected mice, to hyperbolic, typical of this enzyme kinetics in the control, uninfected animals.

Opis:
To differentiate between acute and past Toxocara canis infections, a modified enzyme-linked immunosorbent assay, based on the dissociation of antigen-antibody complex with 6 M urea solution was used to measure levels of avidity of specific immunoglobulin G antibody against Toxocara canis excretory-secretory antigen. IgG avidity index was determined in the sera of 212 patients, divided into two groups with various stages of Toxocara infection. Patients were classified according to time period, from the onset of clinical symptoms or first positive serology result. Low IgG avidity index was found in the sera of 6.6% of patients in the acute phase, and all samples from the chronic stage of infection were characterized by high avidity values above 0.5 (mean 0.74). It was documented, that the avidity test for specific IgG antibodies is a valuable diagnostic method that helps to distinguish the early from the later phase of Toxocara canis infection. Indications for anti-parasitic treatment according to immunological activity of Toxocara infection were discussed.
