Opis:
Effective antibacterial coatings are in demand in medicine, especially for urological medical devices such as catheters and stents. We propose the production method of an antibacterial hydrogel coating on polydimethylsiloxane (PDMS, silicone), a popular surface for medical materials. The coating process consists of the following steps: PDMS surface activation (introduction of hydroxyl groups), silanisation (introduction of amine groups) and application of chitosan/alginate hydrogel with the addition of lysozyme as an antibacterial agent using the layer-by-layer method. We investigated the effect of polyion concentration on the coating mass, swelling ratio and stability. We analysed the adsorption of Micrococcus luteus, Escherichia coli and Proteus rettgeri on a PDMS surface using confocal laser scanning microscopy. The chitosan/alginate hydrogel coating with immobilised lysozyme protected the PDMS surface against adhesion for all three tested bacterial strains.

Opis:
Fifteen samples of silicone plates (PlateSi, area=12 cm2), with surfaces modified layer-by-layer with chitosan and unfractionated heparin, were obtained. The sample surfaces were pre-treated by cold oxygen plasma in a planar-type plasma chemical reactor with 50 W power before coating with layered polysaccharides. Pre-treatment was carried out in two alternative operation modes of the reactor, namely in the plasma etching mode and in the reactive-ion etching mode. Thromboresistance was assessed in vitro in contact with human blood. The thromboresistant silicon plates, modified layer-by-layer (3, 5, 7, and 9 bilayers) with chitosan, with molecular weights of 65 kDa, increased with the increase in the number of layers, up to 5. An increase in the duration of thromboresistance was observed in layer-by-layer modification of the surface of the plates with chitosan with a molecular weight of 200 kDa or with quaternized chitosan with a molecular weight of 200 kDa. Some samples of highly thromboresistant, modified PlateSi contributed to the adhesion of platelets and the haemolysis of red blood cells to a lesser extent than untreated silicon plates. The three most promising samples of modified PlateSi were selected.

In this study using methods of Atomic force microscopy (AFM) and Quartz crystal microbalance with dissipation monitoring (QCM-D) the special characteristics of formation and architecture of thin films coatings based on natural polysaccharides chitosan and pectin were investigated. A layer-by-layer (LbL) deposition technique for assembling of oppositely charged layers was used. The main factors, which have an influence on the process of thin film formation and molecular structure of these coatings, were investigated.
