The aim of the study was the determination of IgA, IgM and IgG concentrations in porcine serum and colostrum, in order to evaluate their variations in the perinatal period, as well as to clarify whether there is a correlation between colostrum intake, initial level of immunoglobulins (Ig) in piglet serum and development of their own immunity. The mean IgA, IgM and IgG concentrations in sow serum 10 days before parturition were 1.58, 6.12 and 39.56 mg/ml, respectively. Seven days later only the IgG level was insignificantly lower (34.94 mg/ml, p=0.55), while concentrations of IgA and IgM increased to 2.25 and 7.25 mg/ml, respectively (p=0.23 and 0.62, respectively). The mean initial IgG concentration in colostrum at farrowing was 118.5 mg/ml and differed between sows. The average value of IgA in colostrum at birth was 23.8 mg/ml and decreased to 7.85 mg/ml at 6 hours (h) and to 4.59 mg/ml at 24 h after the onset of farrowing. IgM concentration at birth was 12.1 mg/ml and decreased to 4.23 mg/ml at 24 h postpartum. Positive relationships were found between concentrations of IgM and IgA in serum of piglets at 14 and 56 days of life (r = 0.41 and 0.80, respectively, p≤0.05) as well as for IgG concentration in the piglets serum at 7 days and 56 days of age (r = 0.48, p≤0.05). The above observations suggest that there is a correlation between the level of Ig in piglet serum in the first days of life and improvement of their own immunity.

The research was conducted on clinically healthy mares (n=40) and foals (n=78) duringY. pseudotuberculosis associated enzootics. The animals were divided into groups: I to IV – mares, IA to IVA – their offsprings, IB to IVB – foals which mothers were not treated with any medicaments. The animals in group I, IA and IB were injected with PBS; in group II, IIA and IIB – with Y. pseudotuberculosis strain-based vaccine, in group III, IIIA and IIIB – with P. acnes strain-based immunostimulator; in group IV, IVA and IVB – with P. acnes strain-based immunostimulator and (5 days after the immunostimulator injection)Y. pseudotuberculosis strain-based vaccine. The presence of antibodies was determined by means of ELISA. The study revealed anti-Yersinia pseudotuberculosis IgG only in 19 mares before, and in 25 mares and 26 foals 3 weeks after vaccination. The mean extinction 3 weeks after vaccination amounted to: II-0.489, IV-2.578, IIA-0.572, IVA-0.974, IIB-0.312, IVB-0.418. The cut-off extinction value was 0.154. The presence of anti-Yersinia pseudotuberculosis IgG before vaccination in the sera of clinically healthy mares may suggest that Y. pseudotuberculosis infection occurs definitely more often than is expected. Vaccination preceded by immunostimulation appeared to be the most efficient method of treatment against yersiniosis.

The present study was designed to investigate the influence of Se deficiency on serum histamine concentration and the expression of histamine receptor in the jejunum of chickens. Forty neonatal chickens were randomly divided into two groups. Experimental chickens were fed a low-Se diet (0.034 mg/kg), whereas chickens in the control group were fed a diet with a Se level of 0.229 mg/kg. Ten chickens were sacrificed on days 30, 45, 60 and 75. Blood and jejunum samples were collected. Histamine concentration in the jejunum was measured by ELISA, the jejunal mast cell (MC) ultrastructure was studied by transmission electron microscopy, and the expression level of histamine H2 receptor (H2R) mRNA in the jejunum was examined using real-time PCR. Results: The jejunal histamine concentration in chickens fed the low-Se diet was significantly higher than that in the control group (P<0.01). Se deficiency induced degranulation of MC in the jejunum of chickens in the low-Se diet group; their cytoplasm was filled with fused granules and vacuoles. The expression level of jejunal H2R mRNA in chickens fed the low-Se diet was also significantly higher than that in the control group (P<0.01). The results obtained suggest that Se deficiency stimulates MC degranulation and release of histamine, binding H2R promotes both regulation of digestion and cell proliferation while protects the jejunum from injury induced by Se deficiency.

Sialic acids are important constituents of animal tissue glycoconjugates and are also present in the antigens of some bacterial strains. Capsular polysaccharides with sialic acid (NeuAc) have been extensively studied with regard to sensitivity to the bactericidal action of serum, whereas little is known in this regard about lipopolysaccharides (LPS) which contain NeuAc. Strains of Salmonella 048, able to infect animals and containing the same structures of LPS with NeuAc, were examined for their susceptibility to the bactericidal action of normal bovine serum (NBS). The strains showed varied sensitivity to the bactericidal action of NBS, which indicates that the expression of LPS containing NeuAc residues is not critical for the strains' resistance to the serum's activity. In this study the mechanisms of complement activation responsible for killing serum-sensitive Salmonella 048 rods by NBS were also established. Three such mechanisms were distinguished: activation of the classi- cal/lectin pathways, important (decisive) in the bactericidal mechanism of complement activation, parallel activation of the classical/lectin and alternative pathways, and independent activation of the classical and lectin or the alternative pathway.

Opis:
Serology plays an important role in laboratory diagnosis of leptospirosis. Apart from the most often used microscopic agglutination test (MAT), enzyme-linked immunosorbent assay (ELISA) seems to be useful especially in screenings of animal herds. The ELISA used for detection of antibodies against selected Leptospira serogroups in swine serum samples was investigated during the study. An essential element of this test is heat-stable antigenic preparation from cultures of Leptospira interrogans serovars Icterohaemorrhagiae, Pomona and L. borgpetersenii serovar Sejroe. The aim of the present study was to identify and analyze ELISA heat-stable antigen fractions playing a role in the reaction with leptospiral antibodies indicated in swine serum. Reactivity of the three-component antigenic preparation was compared in immunoblotting with reactivity of six heat-stable antigenic preparations made from the following single serovars: L. interrogans serovars Icterohaemorrhagiae, Pomona, Canicola, L. borgpetersenii serovars Sejroe, Tarassovi and L. kirshneri serovar Grippotyphosa. All antigenic preparations were submitted to SDS-PAGE and transferred to a nitrocellulose membrane using a semidry system. After the transfer, the membrane was incubated with diluted swine serum containing antibodies specific for one of the six above mentioned Leptospira serovars. For the three-component antigenic preparation and antigens prepared from single serovars the immunoblot revealed reaction of sera with fractions of the 20-26 kDa region and around the 14.5 kDa region. The investigated heat-stable Leptospira antigenic preparation contains fractions demonstrating serogroup- and species-specificity. Fraction 20-26 kDa showed serogroup-specific activity, whereas the fraction around 14.5 kDa showed species-specific activity.

The experimental materials comprised 44 hybrid [ (♀Polish Large White x Polish Landrace) x ♂Duroc] growing-finishing pigs. The animals were randomly divided into two groups: 24 pigs were slaughtered immediately after transport and 20 pigs were slaughtered after a 24-hour rest period in the lairage. The meat content of pork carcasses, carcass dressing percentage, the proximate chemical composition, physicochemical and sensory properties of meat and shear force values were determined. Serum creatine kinase activity and cortisol levels were determined in blood samples collected before transport and during carcass bleeding. Pigs slaughtered immediately after transport, compared with those slaughtered after a 24-hour rest period, were characterized by a higher meat content of the carcass and a higher carcass dressing percentage. Pre-slaughter handling had no effect on pork quality. The incidence of normal-quality meat, partially PSE (pale, soft, exudative) meat and PSE meat was similar in both groups. Chemical analysis showed that the content of dry matter, total protein, fat and minerals in meat was comparable in both groups. As regards the functional properties of the pork, samples from the carcasses of pigs that had rested before slaughter had a higher contribution of the red color component. Meat from pigs slaughtered immediately after transport had more desirable sensory properties. Pre-slaughter resting had a significant effect on those analyzed physiological parameters which were found to be good indicators of pre-slaughter stress. Serum creatine kinase activity and cortisol levels were higher in blood samples collected after transport (during carcass bleeding) than in samples collected before transport, pointing to a strong stress response of animals to pre-slaughter treatment. The decrease in serum cortisol levels in blood samples collected during bleeding from the carcasses of pigs slaughtered after a 24-hour rest period, compared with samples collected from animals slaughtered immediately after transport, suggests that rest before slaughter alleviated stress induced by pre-slaughter handling operations.

The increasing interest in enzymatic growth promoters prompted this investigation of the effect of an enzymatic stimulating complex on selected hematological and serum biochemical parameters in boars. The enzymatic complex [five proteases (proteinases-endopeptidases) and two peptidases (exopeptidases) obtained by fermentation from Streptomyces fradiae] was added to diets for three months, at initial doses of 90 (group El) and 120 (group E2) g/ton feed for the first seven days followed by 40 and 60 g/ton, respectively, in the subsequent weeks. The evaluation was based on selected hematological (RBC, WBC, HCT (Ht), HGB (Hb), MCV, MCH, and MCHC) and biochemical (AST, ALT, AP, Ca, Mg, urea, cholesterol, creatinine, inorganic phosphorus, and oxyglucose) parameters determined before and after the experimental period. The reported values were within physiological norms. Statistical differences were found between experimental groups with respect to WBC, HCT (Ht), MCV, MCH, AP, Ca, Mg, creatinine, and inorganic phosphorus. The study results indicate that the complex of proteolytic enzymes administered to boars caused no negative changes in their metabolic profiles. The statistically different hematological and serum biochemical parameters, while within normal limits, point to an early stage of microcytic anemia and heightened agitation that could be the result of intensified nitrogen metabolism.

In cattle, the prevention of diseases might be possible using dietary methods, with nutritional optimization of feed rations. For years significant influence has been associated with the addition of biotin, methionine and zinc (each given individually, or as chelate – Zn-methionine). The aim of this study was to evaluate the influence of biotin or biotin + Zn-methionine additives on the health and performance of cows, in the first and second trimester of lactation. This study was carried out in 3 groups of cows, 30 animals in each group. In the first group cows were fed the usual fodder used on the farm. In the second group biotin at 10 mg/day/cow was added. In the third group both biotin at 10 mg/day/cow and Zn-methionine at 5 g/day/cow were added. The administration of biotin increased the milk yield and reduced the occurrence of retained placenta, as well as endometritis puerperalis. However, the high doses of biotin suppressed ovulation and oestrus symptoms in the investigated cows (low progesterone levels on days 15, 21 and 45 after calving). Zn-methionine in comparison to biotin had a lower effect on the milk yield in the cows. Moreover, its supplementation improved the milk content, as well as some of the blood parameters; it also decreased the number of somatic cells in milk. The concurrent administration of biotin and zinc-methionine seemed to be a good method for the prophylaxis of subacute mastitis and for the improvement of the high yielding dairy cows’ productivity.
