In the present study we investigated the effect of bovine conglutinin on the phagocytic activity of leukocytes. We measured both the chemotactic activity of conglutinin and its effect on the internalization of zymosan particles and E. coli by granulocytes. We also assessed the binding of conglutinin to various microorganisms isolated from clinical cases in cattle. We showed that conglutinin binds strongly to the surface of yeast cells and to mannan-rich zymosan particles, while weak binding was observed in the case of the bacterial strains tested, including those whose O antigen is composed of mannan. Conglutinin (1-10 μg/ml) neither acts as a chemotactic factor for peripheral blood leukocytes nor affects ingestion of E. coli by granulocytes. However, as flow cytometry based assay showed, conglutinin (0.1-1 μg/ml) increased ingestion of zymosan expressed as mean fluorescence intensity (MFI) of positive cells.

Opis:
Lactoferrin (LF) is a glycoprotein found in milk, neutrophil granules, secretions and selected organs of mammals. Lactoferrin exhibits antibacterial, antiviral, fungicidal, immunoregulatory and other functions. Although fish are devoid of this protein and its cell receptors, LF effect on the immune mechanisms of fish has been demonstrated. The objective of this study was to investigate the effect of bovine lactoferrin, applied in vitro, on the activity of head kidney and spleen leukocytes in three freshwater fish species: rainbow trout (Oncorhynchus mykiss), European eel (Anguilla anguilla) and weis catfish (Silurus glanis). The obtained results validate LF beneficial effect on the respiratory burst of phagocytes in rainbow trout and weis catfish despite the fact that the potential killing activity against Aeromonas hydrophila was not stimulated in any of the studied species. Bovine lactoferrin enhanced the proliferation of T-lymphocytes in rainbow trout and European eel, as well as of B-lymphocytes in rainbow trout.

Cyanobacteria (Cyanophyta, Cyanoprocaryota, Cyanobacteria) (blue-green algae) are procaryotic phototrophic microorganisms playing an important ecological role in the freshwater and marine environment as primary producers. However, as a consequence of water eutrophication observed in many reservoirs in different parts of the world, these microorganisms form massive scums, known as water blooms, releasing cyanotoxins hazardous to fish and other aquatic organisms. Cyanotoxins are cyanobacterial secondary metabolites of various chemical structures harmful to humans, terrestial and aquatic animals such as fish. The most abundant cyanotoxins are microcystins and hepatotoxins inducing toxic changes in fish liver, kidney, gills, digestive tract and immune system. Very little is known on the effects of alkaloid neurotoxic anatoxin-a on fish and their immunity. The aim of this study was to assess the in vitro influence of anatoxin-a on immune cells isolated from the common carp (Cyprinus carpio L.). The leukocyte intracellular level of ATP was reduced only at the highest concentration of anatoxin-a. Apoptotic and necrotic leukocytes were observed at the lower and the highest concentrations of anatoxin-a, respectively. Elevated activity of caspases 3/7 after 2 hours and a concentration-dependent decrease in the proliferative ability of T and B lymphocytes was also observed. The results suggest that anatoxin-a could be a possible immunotoxic agent in the aquatic environment and may increase the susceptibility of fish to infectious and neoplastic diseases. Therefore, constant monitoring of anatoxin-a and its producers in lakes and fish ponds should be performed.
