The aim of the study was to evaluate the effect of repeated low doses of GnRH agonist buserelin once a day for 5 days on follicle development and ovulation in anovulatory dairy cows with follicles growth only to emergence. The study was conducted on 71 anovulatory Polish Holstein Friesian cows. Anovulation with growth of follicles to emergence was defined as small ovaries with follicles of ≤ 5 mm in diameter and without corpus luteum on two examinations in a 7-10 day interval between 50-60 days after parturition. Cows were allocated to one of two group. Cows from group 1 (n = 58) received 0.4 μg of buserelin (Receptal, MSD, Poland) i.m. once a day for 5 days. Control cows from group 2 (n = 13) received saline. Ovarian structures were monitored weekly after the end of treatment by ultrasound for 4 weeks. The diameter of ovarian follicles on the ovaries was measured and recorded. Occurrence of ovulation was determined by the presence of corpus luteum. Overall, ovulation occurred in 46.6% (27/58) of cows treated with repeated doses of GnRH, while no corpus luteum was observed in the control group during the study period. There were significantly (p<0.05) more follicles 6-9 mm in diameter and 10-20 mm in diameter in cows treated with GnRH than in control cows. In conclusion, repeated low doses of GnRH analogue buserelin once a day for 5 days stimulate the development of ovarian follicles in anovulatory dairy cows with small ovarian follicles and led to ovulation in 46.6% of cows during 4 weeks after the end of the treatment.

The aim of the study was to find out whether the single nucleotide polymorphism (SNP) within arylsulfatase D (ARSD) gene is associated with kinematic parameters of sperm motility in Holstein- Friesian bulls. 367 Holstein-Friesian bulls kept in one AI center were included in the study. Point mutation C/T at position 139037255 on chromosome X (rs42207167) was identified by PCR-RFLP method (Pflm I). Significant associations were found between ARSD genotypes and CASA-derived sperm motility parameters: average TM (Total Motility), average VSL (Straight Velocity), average VCL (Curvilinear Velocity) and for fraction of sperms showing progressive motility (a) of sperms (VSLa, VCLa and BCFa -Beat Cross Frequency). Most significant differences were observed between alternative homozygotes (CC vs TT). Our results suggest new role of arylsulfatase D gene as being involved in sperm motility.

Selected qualitative and biochemical parameters were determined in cryopreserved semen used for artificial insemination, sampled from 120 bulls reared at the Animal Breeding and Insemination Center in Bydgoszcz. The total average motility of the analyzed sperm samples was determined at 62.51%. The percentage of motile spermatozoa displaying progressive forward motility was 21.65%. Analyzed samples were characterized by a high percentage of sperm cells with a intact plasma membrane (71.21%) and active mitochondria (71.32%). High efficiency of the enzymatic antioxidant system of the evaluated sperm cells was demonstrated by high activity of CAT, GPx and SOD (494.37, 2847.83 and 5.31U/1x109 spermatozoa, respectively) values and low values of the DNA Fragmentation Index (9.32). The results of the study, obtained with the involvement of advanced analytical methods, indicate a high fertilizing capability of the analyzed sperm samples.

WC1+ cells in cattle exhibit both regulatory and effector activities. However, it has not been elucidated whether they are so plastic that both activities co-exist in one cell or there are separate subpopulations of effector and regulatory cells. Since the production of IFN-γ and IL-10 seems to be related to WC1+ cells’ effector and regulatory function, respectively, the main aim of this study was to determine whether those cytokines are produced by separate subpopulations of WC1+, or are co-produced by the same cells. Due to increasingly frequent emphasised role of consumption of IL-2 in the mechanism of suppressor action of mouse CD25+CD4+ T regulatory cells, expression of the receptor’s α chain for interleukin 2 (CD25) on WC1+ lymphocytes has been evaluated. An average of 5.21% of WC1+ cells obtained from PBMCs of 12-month-old heifers show constitutive expression of the CD25 molecule, with CD25highWC1+ and CD25lowWC1+ cells accounting for 1.05% and 4.10% of WC1+ lymphocytes, respectively. For detection of intracellular cytokine production, PBMCs were stimulated with concanavalin A. Both IFN-γ- and IL-10-producing cells within the CD25-WC1+ and CD25+WC1+ subpopulations were mainly separate subpopulations. The average percentage of IFN-γ+IL-10-, IFN-γ-IL-10+ and IFN-γ+IL-10+ cells among CD25-WC1+ lymphocytes was 4.03%, 2.67% and 0.51%, respectively. A positive correlation was observed between the presence of the CD25 molecule on WC1+ lymphocytes and production of IL-10 and TGF-β, because the average percentage of IFN-γ-IL-10+ and IFN-γ+IL-10+ among CD25+WC1+ lymphocytes was 3 and 4.5 times higher as compared to the corresponding cells in the CD25-WC1+ subpopulation, whereas the percentage of IFN-γ+IL-10- cells in both the subpopulations was not significantly different. The percentage of TGF-β+ cells within the CD25+WC1+ subpopulation was 2.72 times as high as that of CD25-WC1+ lymphocytes. Therefore, with respect to the production of IFN-γ, IL-10 and TGF-β, CD25+WC1+ lymphocytes turn out to have a more suppressor profile than CD25-WC1+.

Information on the prevalence of subclinical endometritis and its mechanism in repeat breeding cows is very limited. The aims of this study were: a) to evaluate the incidence of this disorder with cytobrush cytology b) to analyze mRNA expression of tumor necrosis factor α (TNFα) and inducible nitric oxide synthase (iNOS) in endometrial biopsy samples collected from repeat breeding cows with and without subclinical endometritis. Two experiments were carried out. In experiment 1, 112 (12.4%) repeat breeding cows (inseminated at least 3 times and not pregnant) were selected out of 902 cows from 8 dairy herds. Cytobrush cytology was performed on these cows, using the threshold of 10% PMNs in uterine smears. The results showed that 45 out of the 112 cows (40.2%) were diagnosed as having subclinical endometritis. In experiment 2, uterine biopsy samples were taken from repeat breeding cows with subclinical endometritis (n = 10) and without this disorder (n = 10). Using reverse transcription-PCR, the mRNA expression of TNFα and iNOS was determined. A statistically significant increase in expression of both substances was measured in the group of cows with subclinical endometritis (p < 0.05). These results provide evidence for a high prevalence of subclinical endometritis in repeat breeding cows as well as the involvement of TNFα and iNOS pathways in the regulation of this pathological condition.

The aim of the in vitro study was to determine the effect of corn dried distillers grains with solubles (corn DDGS), used as a replacement for the concentrate ingredients of sheep diet, on rumen fermentation. The material for the study was the ruminal fluid of Polish Merino sheep which was incubated during 4-, 8- or 24-hour periods. Five groups of samples were prepared for in vitro fermentation: C – control, incubated with the substrate consisting of the concentrate ingredients; D1, D2 and D3, where DDGS was used as a substrate added in proportions of 10, 20 and 30% of dry matter of the concentrate; and D4, where 100% DDGS was used as a substrate. After fermentation, the gas and short chain fatty acids (SCFAs) analyses were performed using gas chromatography. The ammonia concentration and pH were also determined, and the SCFA utilization index (NGR), the fermentation efficiency (FE) and the index of cell yield of ruminal microorganisms (CY) were calculated. This research showed no effect of DDGS on the methane emission. The positive correlations between the amount of methane and ammonia concentrations in the 8- and 24-hour fermentation periods were found. DDGS addition increased propionate proportion, but decreased production of acetate (p<0.01). Additionally, D1, D2, D3 and D4 substrates lowered isobutyrate (p<0.05) and isovalerate (p<0.01) production. Based on the results obtained, it can be stated that partial substitution of the concentrate ingredients with DDGS did not have deleterious effect on sheep rumen fermentation processes.

In this study, we assessed the effect of a humic-fatty acid preparation (HFA) used in rabbit feed on certain haematological and biochemical serum parameters (the lipid profile and the Ca, P and Fe contents). A higher RBC, HGB and HCT values were observed in the groups that were given HFA. An increase in Fe concentrations were also noted. The total and LDL cholesterol were lower in the groups which received HFA than in the control group.

The aim of the study was the evaluation of cadmium effects on the activity of antioxidant enzymes in rat hepatocytes. The studies were conducted with isolated rat hepatocytes incubated for 1 or 2 hours in a modified (deprived of carbonates with phosphates) Williams’ E medium (MWE) in the presence of cadmium chloride (25, 50 and 200 μM). Hepatocytes incubated in the MWE medium without cadmium chloride were used as a control. The application of the modified Williams’ E medium allowed for the appearance of cadmium compounds in a soluble form that is indispensable for suitable estimation of its toxic action. There were evaluated markers of the oxidative stress such as: concentration of thiobarbiturate reactive substances (TBARS) – proportional to the level of lipid peroxidation, concentration of reduced glutathione (GSH), and the activity of antioxidant enzymes, including superoxide dismutase (SOD1 and SOD2), catalase (CAT), total glutathione peroxidase (GSHPx), selenium – dependent glutathione peroxidase (SeGSHPx), glutathione transferase (GST) and glutathione reductase (GSHR). Alterations of antioxidant enzymes activity, the level of TBARS and GSH in isolated rat hepatocytes caused by cadmium in vitro, were shown to depend on the concentration and time of exposure of cells to this metal. The increased level of TBARS and GSH was observed as well as changes in the activity of antioxidant enzymes. The activity of SOD isoenzymes and CAT was increased, whereas GSHPx and GST were decreased. These results indicate that cadmium induces oxidative stress followed by alterations in the cellular antioxidant enzyme system in isolated rat hepatocytes.

In the present study influence of doxazosin on the porcine urinary bladder contractility has been examined. Immature pigs were treated for 30 days with: a) doxazosin (n = 5) per os at a dose of 0.1 mg/kg b.w. or b) placebo (n = 5; control group). Thereafter, animals were sacrificed and urinary bladder strips from the trigone were suspended in organ baths. The tension of the smooth musce was measured before and after exposition to acetylocholine (ACh; 10⁻⁵ - 10⁻³ M), norepinephrine (NE; 10⁻⁹ - 10⁻⁷ M) and 5-hydroxytryptamine (5-HT; 10⁻⁷ - 10⁻⁵ M). Both the ACh and 5-HT at the highest doses significantly increased the contractility in each group, but this response was weaker in doxazosin-treated animals. NE caused relaxation in both groups, but the effect was weaker in doxazosine-treated group. The results of our study have shown that long-term administration of doxazosin caused a desensitization of the detrusor smooth muscle for in vitro applied mediators of the autonomic nervous systems.
