Opis:
This paper presents the physiological activity of the cerebral cortex in sheep in electroencephalographic findings. The study was performed to evaluate and improve understanding of brain monitoring methods in freely moving animals without the use of any anaesthetic methods during the acquisition stage. The aim of the study was to determine the physiological activity of the cerebral cortex in animals in a sheep model (using clinically healthy Polish Merino rams, aged 1 yr.) to determine its clinical EEG protocol. The EEG was registered using an in-lab EEG device as well as ambulatory systems (Holter EEG). The bioelectrical activity of the sheep cerebral cortex was recorded using gold disc and needle electrodes placed on experimentally determined locations on the scalp. The main finding of this study was the rhythmic EEG activity of the sheep brain in wakeful and conscious states (eyes open). The rhythm was bilaterally synchronous and determined a basic pattern in the registration of physiological activity of the cerebral cortex. The acquired rhythm may correspond to the theta rhythm in humans, which is a normal activity controlled by the hippocampus. This background activity comprised intermittent episodic slow waves and synchronous beta waves.

Opis:
Trichomonadidae family is a protozoan occurring in different animal species. It inhabits the gastrointestinal and urinary tracts. P. hominis is rarely found in faecal samples of dogs, and its identification and differentiation from other trichomonads by light microscopy are difficult. Methods of molecular biology are the most effective in this case, because they confirm the presence of the specific species in animal organisms, irrespective of the protozoan form. The aim of this study was to find P. hominis in selected dog kennels in North-Eastern Poland. Forty-one faecal samples of dogs from 7 dog kennels were examined. The occurrence of P. hominis in 5 faecal samples of dogs with no symptoms of diarrhoea was the first one to be confirmed in Poland.

Opis:
The aim of this study was to compare the effect of different packaging systems on some parameters of cryopreserved canine spermatozoa. The experimental material consisted of the sperm-rich fractions of ejaculates collected from four Beagle dogs. Semen samples for cryopreservation were stored in 0.25 ml plastic straws and two aluminum tubes with a total volume of 5.0 ml. Semen was frozen in static nitrogen vapor for 10 minutes (0.25 ml straws) or 15 and 20 minutes (aluminum tubes). Post-thaw assessments involved the determination of sperm motility parameters using a computer assisted sperm analyzer (CASA), sperm plasma membrane integrity (SPMI), mitochondrial membrane potential (MMP) and acrosome integrity (normal apical ridge, NAR). Regardless of the packaging system applied, no significant differences in total sperm motility (TMOT) or selected kinematic parameters were observed after freezing-thawing. However, spermatozoa frozen in 0.25 mL straws were characterized by improved functionality, in particular mitochondrial function, after thawing. The results indicate that large quantities of canine semen can be frozen in aluminum tubes. Further studies are required, however, to evaluate different freezing and thawing rates of aluminum tubes.

Opis:
The aim of this study was to analyze disorders of acid-base balance in calves with chronic diarrhea caused by mixed, viral, bacterial and Cryptosporydium parvum infection. We compared results obtained with the classic model (Henderson-Hasselbalch) and strong ion approach (the Steward model). The study included 36 calves aged between 14 and 21 days. The calves were allocated to three groups: I - (control) non-diarrheic calves, group II - animals with compensated acid-base imbalance and group III calves with compensated acid-base disorders and hypoalbuminemia. Plasma concentrations of Na+, K+, Cl-, Cl2+, Mg2+, P, albumin and lactate were measured. In the classic model, acid-base balance was determined on the basis of blood pH, pCO2, HCO3-, BE and anion gap. In the strong ion model, strong ion difference (SID), effective strong anion difference, total plasma concentration of nonvolatile buffers (ATot) and strong ion gap (SIG) were measured. The control calves and the animals from groups II and III did not differ significantly in terms of their blood pH. The plasma concentration of HCO3-, BE and partial pressure of CO2 in animals from the two groups with chronic diarrhea were significantly higher than those found in the controls. The highest BE (6.03 mmol/l) was documented in calves from group II. The animals from this group presented compensation resulted from activation of metabolic mechanisms. The calves with hypoal-buminemia (group III) showed lower plasma concentrations of albumin (15.37 g/L), Cl- (74.94 mmol/L), Mg2+ (0.53 mmol/L), P (1.41 mmol/L) and higher value of anion gap (39.03 mmol/L). This group III presented significantly higher SID3 (71.89 mmol/L), SID7 (72.92 mmol/L) and SIG (43.53 mmol/L) values than animals from the remaining groups (P<0.01), whereas ATot (6.82 mmol/L) were significantly lower. The main finding of the correlation study was the excellent relationship between the AGcorr and SID3, SID7, SIG. In conclusion, chronic diarrhea leads to numerous water-electrolyte disorders. Characterization of acid-base disturbance in these cases suggests that classic model have some limitations. This model can not be recommended for use whenever serum albumin or phosphate concentrations are markedly abnormal.

Opis:
Malignant lymphoma is one of the most common malignant tumours occurring in dogs. Fine needle aspiration biopsy (FNAB) is an excellent, specific diagnostic procedure used to assess pathological processes in lymph nodes. The aim of the present study was to conduct a cytopathological analysis of lymphoma in dogs and to analyse some epidemic aspects of occurrence of lymphoma in 100 dogs using Giemsa stained slides. Samples were obtained by fine needle aspiration biopsy, fine needle non-aspiration biopsy, lymph node impression smears and by examination of body cavity effusions. The determination of the type and subtype of tumour was made on the basis of the updated Kiel classification adopted for dogs. Based on cytopathological analysis, the lymphoma was diagnosed in 100 dogs: 44 were female and 56 male. The animals’ age ranged from 1.5 year to 15 years (median: 7.5 years), the animals were of different breeds (72% of all dogs belonged to 28 different breeds) and crossbreeds (28%). In 29% of dogs the regional or general lymphadenomegaly was the only clinical sign observed, in remaining cases (71%) at least one abnormality connected to lymphoma was found. Among all diagnosed lymphomas, high-grade lymphomas were more prevalent (86% of all cases) than low-grade lymphomas (14% of all cases). The possibility of boxers having a predisposition to T cell lymphoma development could be also suspected.

Opis:
The study was aimed at determining the age-depended changes in insulin-like growth factors 1 and 2 (IGF-1 and IGF-2) mRNA levels in relation to the gastrointestinal tract (GIT) development in newborn calves. Twenty four male calves (5-day-old) were randomly allocated to 4 groups which were slaughtered at day 5, 12, 19 and 26 of their life, respectively. Tissue samples from the rumen, abomasum, duodenum, jejunum and liver were taken for analysis, and the level of IGF-1 and IGF-2 mRNA was determined using RT semi-quantitative PCR method. Both IGF-1 and IGF-2 mRNA level was the biggest on 5 day of life, significantly decreased up to day 12-19 in most of analyzed GIT parts, and started to increase thereafter. In the rumen the reduction in the amount of IGFs transcripts was associated with the reduction in papillae length and tunica muscularis thickness. The abomasum weight and tunica mucosa thickness increased from the 5 day up to 19 day of life, whereas abomasal IGF-1 mRNA level decreased together with calf's age up to 19 day of life, and the level of IGF-2 mRNA did not change. The reduction in IGFs mRNA level after 5 day of life in the duodenum (IGF-1 and IGF-2) and in the jejunum (IGF-1) was associated with reduction in villi length (duodenum and jejunum), and the increase of crypt depth (duodenum).

Opis:
In this study plasma concentrations of IL-1α and IL-8 in 29 female dogs with malignant mammary gland tumours (19 without metastasis and 10 with metastasis) and in 10 healthy control animals were determined. Concentrations of IL-1α and IL-8 were analysed using a specific canine ELISA assay. Mean plasma concentrations of IL-1α and IL-8 were significantly higher (p<0.05) in female dogs with both non-metastatic and metastatic malignant tumours compared to the healthy animals. The concentrations of both tested cytokines were significantly increased (p<0.05) in the dogs with metastasis. In female dogs with mammary carcinomas, the plasma concentration of IL-1α was significantly higher (p<0.05) in the animals with grade 3 tumours compared to the dogs with grade 1 tumours. The concentration of IL-8 was significantly higher (p<0.05) in the dogs with grade 3 tumours compared to that found in the animals with grade 1 and grade 2 tumours. A moderate correlation (r=0.433) was found between IL-1α and IL-8 concentrations in the female dogs. These findings suggest that increased malignancy and invasiveness of canine mammary tumours is associated with an increased production of IL-1α and IL-8 in the tumour microenvironment, which, in turn, leads to an increase in their circulating levels. This may indicate that circulating levels of the cytokines investigated could be considered as diagnostic and prognostic markers in canine malignant mammary tumours. However, further studies in this fields are needed.

Opis:
In the presented work, we compared the results of determination of rabies antibodies using three in vitro methods: rapid fluorescent focus inhibition test (RFFIT), fluorescent antibody virus neutralisation test (FAVNT) and the immunoenzymatic assay (ELISA). 196 dog sera samples were examined with FAVNT, RFFIT methods and the ELISA test. Sera with low and sufficiently high titre of antibodies had a similar result in determining by all methods. A critical level of rabies antibodies close to the required protection level (0.5 IU/cm3) was seen in sera of 18 dogs (9.18%); these were the sera obtained after primary vaccination of dogs. At this level, even small differences can cause a change in the assessment of the patient's serum seronegativity or seropositivity. Therefore, it is important to choose the appropriate method that has sufficiently strict criteria while having a good reproducibility.
