The aim of this study was to explain the correlations between selenium deficiency, hemostatic and biochemical disorders, and the progression of pathological changes in calves diagnosed with nutritional muscular dystrophy (NMD). The study was performed on 20 calves with supplementation of 8 ml selenium and vitamin E preparation and 20 calves with symptoms of NMD. Blood was sampled from calves aged 5, 12 and 19 days. On day 19, samples of the biceps femoris muscle were collected from 6 animals in each group for histopathological analysis. The following blood parameters were determined: PLT, PT, TT, APTT, fibrinogen and D-dimer concentrations, antithrombin III activity, glucose, selenium and vitamin E concentrations, activity of CK, LDH and GSH-Px. Muscle sections were stained with H&E and HBFP. Platelet counts were significantly lower in calves with symptoms of NMD. No significant differences in coagulation parameters were observed between the groups. Sick calves were diagnosed with hyperglycemia and elevation of CK and LDH activity. Selenium and vitamin E concentrations in the blood serum were significantly lower in the experimental group together with significant drop in GSH-Px activity. Changes characteristic of Zenker’s necrosis were observed in a muscle of the sick animals. To our best knowledge this is the first study in which the attempt was made to explain the relationship between selenium deficiency and changes in the coagulation system in ruminants.

The aim of this study was to evaluate the suitability of thromboelastometry for the analysis of blood test results in goats after the use of hemostatic dressings to control massive bleeding. The study was carried out on 12 goats, 6 animals in each of two subgroups. In all experimental animals incision of the femoral artery was performed, and bleeding was controlled with QuikClot gauze in the first group and Celox gauze in the second group. Dressings were applied for 60 minutes. Blood samples for thromboelastometry were collected from the jugular vein before the incision and 60 min after the application of a dressing. Clotting time (CT), clot formation time (CFT), maximum clot firmness (MCF) and α angle (°) were measured in three standard ROTEM assays (system with generation of reaction curve, numerical parameters and size of the blood clot): intrinsic coagulation pathway (INTEM), extrinsic coagulation pathway (EXTEM) and functional fibrinogen (FIBTEM). Complete hemostasis of the injured femoral artery was found in all goats. No significant differences between pre- and post-incision thromboelastometric parameters were found in any tests in any of the groups, which indicates that the use of dressings was not associated with blood coagulation disorders. This study is the first to describe the use of thromboelastometry in goats for the assessment of clot formation and hemostatic disorders.

The objective of the study was to evaluate the effect of energy contents in a dry period diets in “7+1” feeding strategy of dry cow including 7 weeks of the dry period (far-off) and 1 week of a close-up period, on colostrum quality and the immune status of calves. Forty Holstein multiparous cows were dried at 56d before the expected date of calving and were assigned to the higher energy diet group (HE; 0.69 UFL/kg DM, NDF 52% DM), or the lower energy diet group (LE; 0.61 UFL/1kg DM, NDF 56% DM). From -7d to the expected calving date up to 21 d of lactation, all cows were fed the same fresh, lactation diet. Samples of colostrum were collected within 2h after parturition and its density, dry matter content and concentrations of immunoglobulins, fat, protein, lactose, urea and somatic cell counts were measured. Calves were weighed 2 h after calving and on the 21d of life. On 3d and 21d of calves’ life blood samples were collected and the concentration of IgG, IgA, IgM, insulin-like growth factor 1 (IGF-1) as well as total protein and albumin concentrations were determined. Treatments had no significant effect on composition of colostrum and serum immunoglobulins and IGF-1 concentration. In both groups the weight of calves at birth was similar, still daily body weight gain during 21 day of rearing period in HE group was higher than in the LE group (P=0.05).

The purpose of this study was to investigate possible alterations in acid-base balance parameters and the coagulation profile in neonatal diarrheic calves. Twenty neonatal diarrheic and 20 clinically healthy neonatal calves aged between 1 week to 10 days were used. All blood samples were taken on the third day from the onset of diarrhea symptom. Venous blood samples were collected from each animal to determine platelet numbers, pH, pCO₂, pO₂, HCO₃⁻, BE, O₂SAT, ctCO₂ and electrolytes (K⁺, Na⁺ and Cl⁻). Plasma samples were collected from each animal for the measurement of prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT), the concentrations of fibrinogen, D-dimer and the activity of antithrombin III (AT III). Blood pH (7.19), BE (-10.6 mmol/l), HCO₃⁻ (25.15 mmol/l), pO₂ (3.33 kPa), O₂SAT (24.12 %) were significantly lower and serum concentration of K⁺ (6.55 mmol/l) was significantly higher in diarrheic calves. These changes indicate the state of uncompensated metabolic acidosis with accompanying hyperkalemia. TT (32.05s) and APTT (39.9s) values were more prolonged in calves with diarrhea than in the control group. D-dimer (587.25 μg/l) concentrations were significantly increased while a visible drop in AT III (103.75%) activity and platelets counts (598 x10⁹/l) were observed in diarrheic group of calves. The results suggest that a consumptive type of disseminated intravascular coagulation (DIC) developed in diarrheic calves.

The aim of this study was to determine the presence of cocaine- and amphetamine-regulated transcript-like immunoreactive (CART-LI) neurons and co-localisation of CART with vesicular acetylcholine transporter (VAChT), neuronal nitric oxide synthase (n-NOS), vasoactive intestinal polypeptide (VIP), substance P (SP) and leu-enkephalin (LENK) in the enteric nervous system of the porcine gastric cardia by using a double-labelling immunofluorescence technique. CART-LI neurons were observed in the myenteric plexus (18.2}2.6%). A dense network of CART-LI nerve fibers was mainly observed in the muscular layer. CART showed co-localization mainly with VAChT, n-NOS, VIP and to a lesser degree with LENK and SP. Distribution of CART and its co- ocalization with other neurotransmitters suggest that this peptide plays an important role in gastric motility in the pig.

Non-invasive methods of skin condition assessment include, among others, the evaluation of transepidermal water loss (TEWL). The aim of the study was to examine whether TEWL values depend on horse breed. The study was conducted on four breeds: “Felin” ponies (FP) (n=16), Polish koniks (PK) (n=15), Polish cold-blooded horses (PcbH) (n=11) and Wielkopolska horses (WH) (n=12). It was found that horse breed influences TEWL values. In the neck region, statistically significant differences were found between PK and FP (p=0.006), and PK and WH (p=0.0005). In the lumbar region, there were statistically significant differences between FP and PK (p=0.0009), FP and PcbH (p=0.0016) as well as between PK and WH (p=0.000037), and PcbH and WH (p=0.0006). In the inguinal region statistically significant differences were found between FP and PK (p=0.0003), FP and PcbH (p=0.0005), PK and WH (p=0.009) and PcbH and WH (p=0.006). In the lip region statistically significant differences were observed between FP and PK (p=0.013) as well as between PK and PcbH (p=0.029) and PK and WH (p=0.009). In the examination of TEWL animal breed should be taken into consideration. The non-significant differences found in three of the examined body regions may suggest that these regions are the most adequate for TEWL assessment.

Echinacea purpurea (EP) and Echinacea angustifolia (EA) are ones of the most important world’s herbs with immunotropic activity. They were traditional medicinal plants used by North American Indians for the treatment of various illnesses. Now they are cultivated in many countries and are used mainly to treat respiratory tract infections. Rhodiola rosea (RR) and Rhodiola quadrifida (RQ) are medicinal plants originated from Asia and used traditionally as adaptogens, antidepressants, and anti-inflammatory remedies. We previously reported, that extracts of underground parts of RR and RQ exhibited immunotropic activity. We have demonstrated in pigs that in vitro RR or RQ supplementation of blood lymphocyte cultures stimulated T cell proliferative response to Con A in lower, and inhibited it in higher Rhodiola extract concentrations. The aim of this work was to evaluate the in vivo effect of these herbal remedies on the in vitro proliferative response of mouse splenic lymphocytes to another T-cell mitogen- Phaseolus vulgaris haemagglutinin (PHA). We have found significant stimulation of proliferative response, in comparison to the controls, in mice fed lower doses of tested remedies, and inhibition, no effect or lower stimulation, in mice fed higher doses of these drugs.

Opis:
Mature males of a wild boar-pig crossbreed, during the long and short day season, were used for the study which demonstrates that the chemical light carrier CO regulates the expression of biological clock genes in the hypothalamus via humoral pathways. Autologous blood with experimentally elevated concentrations of endogenous CO (using lamps with white light-emitting diodes) was infused into the ophthalmic venous sinus via the right dorsal nasal vein. Molecular biology methods: qPCR and Western Blot were used to determine the expression of genes and biological clock proteins. The results showed that elevated endogenous CO levels, through blood irradiation, induces changes in genes expression involved in the functioning of the main biological clock located in suprachiasmatic nuclei. Changes in the expression of the transcription factors Bmal1, Clock and Npas2 have a similar pattern in both structures, where a very large decrease in gene expression was shown after exposure to elevated endogenous CO levels. The changes in the gene expression of PER 1-2, CRY 1-2, and REV-ERB α-β and ROR β are not the same for both POA and DH hypothalamic structures, indicating that both structures respond differently to the humoral signal received. The results indicate that CO is a chemical light molecule whose production in an organism depends on the amount of light. An adequate amount of light is an essential factor for the proper functioning of the main biological clock.

The aim of the study was to investigate the effect of different energy concentrations in the isonitrogenous diet fed during the dry period on postpartum health, fertility and blood variables. Forty Holstein multiparous cows were dried 56 days before the expected day of calving and assigned to group (M) with moderate energy concentrations of 0.69 UFL/kg DM or to the low-energy group (L) with energy density of 0.61 UFL/kg DM. From the 7d before the expected day of calving until the 21d of lactation, all the cows were fed the same fresh transition diet (0.82 UFL/kg DM). From the 22d to the 90d of lactation, all the cows received the same highest energy-density lactation diet (0.90 UFL/kg DM). During the dry period the decline of BCS in groups M and L were 0.07 and 0.12 units respectively. The average decrease of BCS from calving to 56 d of lactation were the same in both experimental groups (0.21 BCS). The first-service conception rate tended to be higher in the M group. Insulin-like growth factor-1, glucose, β-hydroxybutyric acid, non-esterified fatty acid, thyroxine serum concentrations prepartum and 3 and 5d postpartum were not significantly affected by the treatment in the dry period.

The genus Rhodiola (Crassulaceae) consists of many species, growing mainly in Asia and traditionally used as adaptogens and anti-inflammatory drugs. In order to elaborate herbal immunostimulator which could be safely given to pregnant women, we performed a study on immunotropic effects of feeding pregnant and lactating mice Rhodiola kirilowii extracts. This paper presents the results of the first part of our study – spleen content of selected polyphenols, spleen cellularity, splenocytes phenotype and their response to mitogens. Experiments were performed on adult inbred females of Balb/c strain, mated with adult males. Females, since copulatory plug was noted, up to the 28-th day after delivery were fed daily with 20 mg/kg b.m. water (RKW) or hydro- lcoholic (RKW-A) extracts of Rhodiola kirilowii. Results: 1. Significantly lower proportion of pregnant mice in experimental groups than in the control. 2. Cellularity of spleen and flavonol quercetin spleen concentration were significantly lower in experimental groups in comparison to the controls. 3. Flavanols ( (+)-catechin and epicatechin) levels were significantly higher in the spleens of experimental mice than in the controls. 4. Positive correlation between spleen cellularity and quercetin, and negative correlation between spleen cellularity and epicatechin content were observed. 5. Spleen mass and spleen lymphocytes phenotype and proliferation in RKW and RKW-A fed mice did not differ from the control. These results, together with suspicion of some embryo- oxicity, are worrying and eliminate the possibility of use Rhodiola kirilowii extracts for long-term treatment in pregnant females.
