The aim of this study was to explain the correlations between selenium deficiency, hemostatic and biochemical disorders, and the progression of pathological changes in calves diagnosed with nutritional muscular dystrophy (NMD). The study was performed on 20 calves with supplementation of 8 ml selenium and vitamin E preparation and 20 calves with symptoms of NMD. Blood was sampled from calves aged 5, 12 and 19 days. On day 19, samples of the biceps femoris muscle were collected from 6 animals in each group for histopathological analysis. The following blood parameters were determined: PLT, PT, TT, APTT, fibrinogen and D-dimer concentrations, antithrombin III activity, glucose, selenium and vitamin E concentrations, activity of CK, LDH and GSH-Px. Muscle sections were stained with H&E and HBFP. Platelet counts were significantly lower in calves with symptoms of NMD. No significant differences in coagulation parameters were observed between the groups. Sick calves were diagnosed with hyperglycemia and elevation of CK and LDH activity. Selenium and vitamin E concentrations in the blood serum were significantly lower in the experimental group together with significant drop in GSH-Px activity. Changes characteristic of Zenker’s necrosis were observed in a muscle of the sick animals. To our best knowledge this is the first study in which the attempt was made to explain the relationship between selenium deficiency and changes in the coagulation system in ruminants.

The experimental materials comprised 44 hybrid [ (♀Polish Large White x Polish Landrace) x ♂Duroc] growing-finishing pigs. The animals were randomly divided into two groups: 24 pigs were slaughtered immediately after transport and 20 pigs were slaughtered after a 24-hour rest period in the lairage. The meat content of pork carcasses, carcass dressing percentage, the proximate chemical composition, physicochemical and sensory properties of meat and shear force values were determined. Serum creatine kinase activity and cortisol levels were determined in blood samples collected before transport and during carcass bleeding. Pigs slaughtered immediately after transport, compared with those slaughtered after a 24-hour rest period, were characterized by a higher meat content of the carcass and a higher carcass dressing percentage. Pre-slaughter handling had no effect on pork quality. The incidence of normal-quality meat, partially PSE (pale, soft, exudative) meat and PSE meat was similar in both groups. Chemical analysis showed that the content of dry matter, total protein, fat and minerals in meat was comparable in both groups. As regards the functional properties of the pork, samples from the carcasses of pigs that had rested before slaughter had a higher contribution of the red color component. Meat from pigs slaughtered immediately after transport had more desirable sensory properties. Pre-slaughter resting had a significant effect on those analyzed physiological parameters which were found to be good indicators of pre-slaughter stress. Serum creatine kinase activity and cortisol levels were higher in blood samples collected after transport (during carcass bleeding) than in samples collected before transport, pointing to a strong stress response of animals to pre-slaughter treatment. The decrease in serum cortisol levels in blood samples collected during bleeding from the carcasses of pigs slaughtered after a 24-hour rest period, compared with samples collected from animals slaughtered immediately after transport, suggests that rest before slaughter alleviated stress induced by pre-slaughter handling operations.

The aim of this study was to evaluate the influence of season on selected qualitative semen characteristics and biochemical markers of canine seminal plasma. Whole ejaculates were collected from 5 crossbred dogs aged 2-8 years. The study covered a period of one year divided into four seasons: spring (March, April, May), summer (June, July, August), autumn (September, October, November) and winter (December, January, February). Semen samples were subjected to macroscopic and microscopic analyses to determine semen volume, total sperm counts and sperm morphology parameters. The study also involved the determination of sperm motility parameters (CASA system), sperm plasma membrane integrity (SPMI, fluorescent staining SYBR-14/PI), sperm mitochondrial membrane potential (MMP, fluorescent staining JC-1/PI) and the ATP content of sperm cells. Total protein content (TPC) and the activity of alkaline phosphatase (AP) and acid phosphatase (AcP) were determined in biochemical analyses of seminal plasma. No significant differences in ejaculate volume, SMPI or ATP content of sperm cells were observed between seasons. The highest total sperm counts were reported in ejaculates acquired in summer and autumn. The lowest MMP values were determined in summer ejaculates. No significant differences in sperm motility (MOT) were observed throughout the experiment, but ejaculates collected in autumn and winter were characterized by the highest progressive motility (PMOT). AP activity and TPC were not significantly affected by season. However, AcP activity levels were significantly lower in autumn than in the remaining seasons. Seasonal variations in the analyzed macroscopic and microscopic parameters of ejaculates and biochemical markers of seminal plasma did not exert a clear negative effect on the quality of canine semen.

The aim of the study was an evaluation of fluorescence polarisation assay (FPA) as a potential tool improving specificity of serological diagnosis of brucellosis in pigs. The evaluation was done by comparing the results of FPA with the results of rose bengal test (RBT), serum agglutination test (SAT), complement fixation test (CFT) and ELISA when false positive sera were tested. One hundred ninety porcine samples, reacting positively in at least one classical serological assay were used. We observed that among 198 sera, 104 were also positive in FPA. The studies confirmed that porcine FPA adds little as far as specificity in comparison to other methods is concerned.

Forty four pigs with typical characteristics for false positive serological reactions (FPSR) were examined for the presence of Yersinia enterocolitica O:9. The positive reactions were observed in rose bengal test (RBT, N=23 sera), serum agglutination test (SAT, N=16), complement fixation test (CFT, N=9), indirect ELISA (i-ELISA, N=11) in first, and in RBT (N=14), SAT (N=8), CFT (N=7) and i-ELISA (N=18) in second examination, respectively. In bacteriological examination Y. enterocolitica was confirmed in 12 cases. Six of these isolates were identified with PCR as Y. enterocolitica O:9.

The aim of this study was to evaluate the influence of training on body surface temperature over the joints in racehorses, measured by infrared thermography. The study involved monitoring of 14 Thoroughbred racehorses in 6 imaging sessions over a period of 3 months. Temperature measurements of the forelimb and hindlimb joints were made before and just after training. Joint temperature of limbs increased significantly after training. Environmental temperature had a statistically significant influence on surface temperature over the joints. The lowest surface temperatures were recorded over the metacarpophalangeal and metatarsophalangeal joint and the highest temperatures in the shoulder, elbow, hip and stifle joint. The metacarpophalangeal and metatarsophalangeal joints warmed the least during training, but were influenced the most by differences in environmental temperature. The surface temperature difference before and after training is an important indicator of the thermoregulatory response to exercise in racing horses. Understanding surface temperature changes in response to regular training is necessary for future studies on diagnosing injuries of joints.

This is the first study to identify 23 protein spots corresponding to 13 proteins in mitochondria isolated from rabbit epididymal spermatozoa. In the group of protein spots identified in stress-induced samples, the abundance of 20 protein spots increased, whereas the abundance of three protein spots (GSTM3, CUNH9orf172, ODF1) decreased relative to the control. The results of this study provide valuable inputs for future research into the molecular mechanisms implicated in pathological processes during oxidative stress (OS).

The aim of this study was to examine the potential interactions of IGF-I with TNF-α and IFN-γ with regard to regulation of the myogenesis and proliferative potential of mouse C2C12 myoblasts. The stimulation of myogenesis by IGF-I (30 nmol/l) was manifested by an enhanced myoblast fusion and expression of myosin heavy chain (MHC) during the first 3 days of differentiation. IGF-I-dependent fusion and MHC expression was reduced by TNF-α and IFN-γ. Both cytokines prevented the stimulatory effect of IGF-I on MyoD expression with minor modification of the myogenin level. Both TNF-α and IFN-γ activated the expression of cyclin A in myoblasts restimulated to proliferation; however, when used in combination with IGF-I these cytokines prevented the rise in cyclin A induced by growth factor. In conclusion: i) TNF-α and IFN-γ reduce IGF-I-dependent myogenesis which was manifested by the reduction of myoblast fusion and MHC cellular levels, ii) Molecular mechanisms of inhibitory action of TNF-α and IFN-γ on IGF-I-mediated differentiation involve a decrease in MyoD whereas myogenin level plays a minor role, iii) TNF-α and IFN-γ increase the proliferative potential of myoblasts; however, they reduced the mitogenic effect of IGF-I, manifested by a decrease of IGF-I-stimulated cyclin A expression in myoblasts reinduced to proliferation. Interactions among IGF-I and proinflammatory cytokines are therefore important to establish a number of myoblasts and the onset of myogenesis during muscle regeneration.

A total of 42 Brucella strains were isolated from animals in Poland in years 2003-2012. Most of them (N=37) originated from wild animals, 3 from cattle, 1 from pig and 1 from sheep. The strains were characterised using both bacteriological and molecular (Bruce-ladder and MLVA) methods. The examinations revealed that all strains from wild boars, hares, cattle and pigs (N=41) had the same phenotypic characteristics and were classified as B. suis biovar 2. The remaining strain, isolated from sheep, was classified as B. ovis. The molecular examination showed that all B. suis biovar 2 strains, except one, had the same molecular profile as reference strain B. suis bv2 Thomsen. Different from the others strain originated from boars imported to Poland and its VNTR profile was typical for Iberian strains.

The aim of the study was to examine potential interactions among IGF-I and proinflammatory cytokines, TNF-α and IFN-γ, in the regulation of local IGF-I bioavailability and cellular proteins mediating myogenic signals. We investigated levels of IGFBP-4, -5, -6, protein kinase Czeta (PKCζ), p38 and extracellular signal-regulated kinase (ERK1/2) in differentiating mouse C2C12 myoblasts. IGF-I significantly stimulated expression of IGFBP-5. TNF-α and IFN-γ attenuated the expression of IGFBP-4 and -6 under basal conditions and in the presence of IGF-I, and inhibited IGF-I-induced IGFBP-5 expression during 5-day myogenesis. TNF-α and IFN-γ markedly attenuated p38 expression in the presence of IGF-I on the 5th day of myogenesis. When combined with IGF-I the cytokines exerted opposite effects on the PKCζ level, i.e. TNF-α caused an increase, whereas IFN-γ reduced the cellular content of this kinase. Exposition of C2C12 myoblasts to IGF-I or cytokines led to the stimulation of ERK1/2 phosphorylation; however, both TNF-α and IFN-γ exerted an inhibitory effect on the activation of ERK1/2 in myoblasts cultured in the presence of IGF-I. We concluded as follows: i) TNF-α and IFN-γ present in the extracellular environment of differentiating C2C12 myoblasts can alter the local bioavailability of IGF-I by inhibiting the expression of IGFBP-4, -5, and -6, ii) the decrease in p38 expression and ERK1/2 phosphorylation in C2C12 myoblasts exposed to cytokines can lead to disturbances in IGF-I-regulated myogenesis.

Effectiveness of long-term anti-BVDV vaccination program in reducing prevalence of persistent BVDV infection in cattle herds was evaluated in seven years observational study (2005-2011). Among three seropositive dairy cattle herds (within herd seroprevalence 100%, confirmed by ELISA Herd Check BVDV Ab, IDEXX, Sweden) vaccination program based on inactivated vaccine (cytopathic strain 5960) was commenced in 2007 in two herds and continued till 2010. In the years 2007-2011 all calves aged 2-12 weeks in all three herds were tested yearly with RT-PCR in order to detect persistently infected individuals. For the entire study period true prevalence of BVDV persistent infection was significantly lower in vaccinated than in non-vaccinated herd. This may imply the role of long-term vaccination program in reducing prevalence of persistent BVDV infection in cattle herds.
