The association between physiologically dependent pharmacokinetic parameters (CLB, T1/2β, Vdss) of marbofloxacin and body weight was studied in eight animal species based on allometric equation Y = aWb, where ‘Y’ is the pharmacokinetic parameter, ‘W’ is body weight, ‘a’ is allometric coefficient (intercept) and ‘b’ is the exponent that describes relation between pharmacokinetic parameter and body weight. The body clearance of marbofloxacin has shown significant (P<0.0001) relation with size (Bwt) in various animal species. However, half-life and volume of distribution were not in association with body weight. Although half-life and volume of distribution were not in a good correlation with body weight, statistically significant association between the body clearance and body weight suggests validity of allometric scaling for predicting pharmacokinetic parameters of marbofloxacin in animal species that have not been studied yet. However further study considering large sample size and other parameters influencing pharmacokinetics of marbofloxacin is recommended.

This study analysed the relaxant properties of salbutamol (β₂₋adrenoceptors agonist) and BRL 37344 (β₃₋adrenoceptors agonist) regarding the contractility of porcine myometrium on days 10-14 of the oestrous cycle (cyclic group; n = 10) and on days 3-5 of pregnancy (early pregnant group; n = 6). The activity of myometrial strips (tension, frequency and amplitude) was recorded under isometric conditions using force transducers. The contractility was assessed further following the administration of increasing concentrations of the agonists (10⁻⁹-10⁻⁴ M), both with and without β-adrenoceptor antagonists (butaxamine – a selective β₂₋ adrenoceptor antagonist, propranolol- a non-selective β₁₋and β₂₋adrenoceptor antagonist and bupranolol – a non-selective β₁₋, β₂₋ and β₃₋adrenoceptor antagonist) at a concentration of 10⁻⁴ M. Although neither salbutamol nor BRL 37344 caused changes in the tension, at the highest concentrations they decreased the frequency and amplitude of contractions. These changes were more evident after salbutamol treatment and in the early pregnant group. Antagonists given alone did not cause changes in the parameters examined but changed some activity of the agonists. Butoxamine reduced the decrease in frequency and amplitude induced by salbutamol and produced a decrease in the tension after BRL 37344 treatment in the early pregnant group. Propranolol reduced the decrease in frequency and amplitude induced by salbutamol in both examined groups and did not cause significant changes in BRL 37344 activity. The administration of bupranolol before salbutamol treatment caused an increase in the tension and reduced the decrease in the frequency in the cyclic group. Moreover, bupranolol eliminated a decrease in frequency and induced an increase in amplitude caused by BRL 37344 in both groups and these changes were more evident in the early pregnant group. The data indicates that both β₂₋ and β₃₋adenoreceptors are involved in the regulation of the contractility in both groups, but the changes after agonists and antagonists treatment are more evident in the early pregnant myometrium.

RT-PCR assays for detection of BRSV, based on four different sets of primers were optimized and evaluated for their sensitivity and specificity. Primers used in this study were specific for genes encoding three BRSV proteins, nucleoprotein N and glycoproteins F and G. Our results indicated that RT-PCR with primers B7:B8 for G protein was the most efficient in detecting BRSV. Starters B7:B8 reacted specifically only with BRSV strains, no cross-reaction with other closely related viruses to BRSV was observed. RT-PCR sensitivity was also high and amounted to 101.66 TCID50. Starters for F and N genes of BRSV were not sufficiently specific and cross-reacted with RNA of HRSV. RT-PCR with primers for the genes F and N of BRSV was characterized by a lower sensitivity than RT-PCR with primers B7:B8. In conclusion, RT-PCR specific to a sequence of glycoprotein G gene, seemed to be the most useful for BRSV detection.

Three different rapid strip tests: TRU RSV, BinaxNOW RSV and RSV Respi-strip were compared with RT-PCR and ELISA BRSV Ag for the ability to detect bovine respiratory syncytial virus (BRSV) in nasal swabs collected from calves experimentally vaccinated with live vaccine Rispoval RS-PI3. The reference strains of BRSV (375 and A51908) were detected by ELISA BRSV Ag whereas the strains of human respiratory syncytial virus (HRSV) and bovine parainfluenza virus type 3 (BPIV-3) were not. All rapid strip tests as well as RT-PCR reacted positively both to HRSV and BRSV reference strains and negatively to BPIV-3. The detection limit for RT-PCR was 39.1 TCID50 (strain 375 of BRSV), whereas for each of the rapid tests it was approximately 156 TCID50 and 312 TCID50 for antigen ELISA. Diagnostic sensitivity in detecting BRSV in nasal swabs for TRU RSV and RSV Respi-strip tests was 33% and 50% for BinaxNOW RSV. Diagnostic specificity of TRU RSV was 100%, whereas for both BinaxNOW and Respi-strip it was 87%. We concluded that TRU RSV could be used as a supportive rapid test for BRSV screening in nasal swabs taken directly on a farm. However, due to the small group of animals used in the experiment, the results should be regarded as preliminary and the study should be repeated on a larger number of animals.

Histological and histochemical investigations revealed that the pterygopalatine ganglion (PPG) in the chinchilla is a structure closely connected with the maxillary nerve. Macro-morphological observations disclosed two different forms of the ganglion: an elongated stripe representing single agglomeration of nerve cells, and a ganglionated plexus comprising smaller aggregations of neurocytes connected with nerve fibres. Immunohistochemistry revealed that nearly 80% of neuronal cell bodies in PPG stained for acetylcholine transferase (CHAT) but only about 50% contained immunoreactivity to vesicular acetylcholine transporter (VACHT). Many neurons (40%) were vasoactive intestinal polypeptide (VIP)-positive. Double-staining demonstrated that approximately 20% of the VIP-immunoreactive neurons were VACHT-negative. Some neurons (10%) in PPG were simultaneously VACHT/nitric oxide synthase (NOS)- or Met-enkephaline (Met-ENK)/CHAT-positive, respectively. A small number of the perikarya stained for somatostatin (SOM) and solitary nerve cell bodies expressed Leu-ENK- and galanin-immunoreactivity. Interestingly about 5-8% of PPG neurons exhibited immunoreactivity to tyrosine hydroxylase (TH). Intraganglionic nerve fibres containing immunoreactivity to VACHT-, VIP- and Met-ENK- were numerous, those stained for calcitonin gene related peptide (CGRP)- and substance P (SP)- were scarce, and single nerve terminals were TH-, GAL-, VIP- and NOS-positive.

The genetic stability of ORF1a encoding non-structural proteins nsp1, nsp2, nsp3 and nsp4 of equine arteritis virus (EAV) has been analysed for nearly seven years in a persistently infected stallion of the Malopolska breed. Between November 2004 and June 2011, 11 semen samples were collected. Viral RNA extracted from semen of this carrier stallion was amplified, sequenced and compared with the sequences of the other known strains of EAV. Sequence analysis of ORF1a showed 84 synonymous and 16 non-synonymous mutations. The most variable part of ORF1a was the region encoding nsp2 protein with 13 non-synonymous substitutions. The degree of amino acid identity between isolates ranged from 98.91 to 100%. Only single non-synonymous mutations were detected in nsp1 (one substitution) and nsp4 (two substitutions). The most stable was nsp3 in which no amino acid substitutions were observed during the whole period of observation.

The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

The experimental materials comprised 44 hybrid [ (♀Polish Large White x Polish Landrace) x ♂Duroc] growing-finishing pigs. The animals were randomly divided into two groups: 24 pigs were slaughtered immediately after transport and 20 pigs were slaughtered after a 24-hour rest period in the lairage. The meat content of pork carcasses, carcass dressing percentage, the proximate chemical composition, physicochemical and sensory properties of meat and shear force values were determined. Serum creatine kinase activity and cortisol levels were determined in blood samples collected before transport and during carcass bleeding. Pigs slaughtered immediately after transport, compared with those slaughtered after a 24-hour rest period, were characterized by a higher meat content of the carcass and a higher carcass dressing percentage. Pre-slaughter handling had no effect on pork quality. The incidence of normal-quality meat, partially PSE (pale, soft, exudative) meat and PSE meat was similar in both groups. Chemical analysis showed that the content of dry matter, total protein, fat and minerals in meat was comparable in both groups. As regards the functional properties of the pork, samples from the carcasses of pigs that had rested before slaughter had a higher contribution of the red color component. Meat from pigs slaughtered immediately after transport had more desirable sensory properties. Pre-slaughter resting had a significant effect on those analyzed physiological parameters which were found to be good indicators of pre-slaughter stress. Serum creatine kinase activity and cortisol levels were higher in blood samples collected after transport (during carcass bleeding) than in samples collected before transport, pointing to a strong stress response of animals to pre-slaughter treatment. The decrease in serum cortisol levels in blood samples collected during bleeding from the carcasses of pigs slaughtered after a 24-hour rest period, compared with samples collected from animals slaughtered immediately after transport, suggests that rest before slaughter alleviated stress induced by pre-slaughter handling operations.

Health status of Polish goat population in regard to the viral diseases remained mostly unknown. In order to determine serological status of Polish goats for selected emerging ruminant viruses, 365 serum samples collected between 2017 and 2019 in 36 districts within 10 of Polish provinces, were tested. No antibodies specific to Peste de Petite Ruminants Virus (PPRSV) and capripoxviruses (CaPV) were found in any of the tested animals. Only single individual (0.27%) was seropositive to Blutongue Virus (BTV). Antibodies directed to Schmallenberg Virus (SBV) were detected in 46 goats which represented 12.6% of the tested population. No association between seropositivity to SBV and year of sampling, province of origin, gender and age was found. In conclusion, among studied viral pathogens, currently only SBV seemed to be important for epidemiological status of Polish goats.

The study was carried out on three 4-month old female pigs. All the animals were deeply anesthetized and transcardially perfused with 4% buffered paraformaldehyde (pH 7.4). Vestibular ganglia (VG) were collected and processed for double-labelling immunofluorescence method. The preparations were examined under the Zeiss LSM 710 confocal microscope equipped with adequate filter blocks. Neurons forming VG were round or oval in shape with a round nucleus in the center. The majority of them (58%) were medium (M) (31-50 μm in diameter) while 28 % and 14% were small (S) (up to 30 μm in diameter) or large (L) (above 50 μm in diameter) in size, respectively. Double-labeling immunofluorescence revealed that VG neurons stained for CGRP (approx. 81%; among them 70.5%, 26.2% and 3.3% were M, S and L in size, respectively), VACHT (57%; 63% M, 24% S, 13% L), Met-Enk (25%; 60% M, 12% S, 28% L), VIP (20%; 88% M, 6% S, L), NPY (15%; 67% M, 20% S, 13% L), GAL (15%; 74% M, 21% S, 5% L), SP (12%; 69% M, 25% S, 6% L) and NOS-positive (12%; 50% S, 50% M). The most abundant populations of intraganglionic nerve fibers were those which stained for CGRP or Met-Enk, whereas only single SP- or NOS-positive nerve terminals were observed.

We investigated the measurement of paraoxonase 1 (PON1), as a potential marker of redox state changes in dairy cows, its involvement in lipid metabolism and compared it with superoxide dismutase (SOD) activity changes. We also evaluated lipid metabolism parameters associated with dairy production. PON1 paraoxonase and arylesterase acitvities, SOD activity, beta-hydroxybutyrate (BHB), uric acid (UA), high density lipoprotein (HDL), low density lipoprotein (LDL), cholesterol and triglyceride concentrations were measured in Holstein-Friesian, Polish Red and Norwegian breeds serum in two production cycles. Our data showed a significant postpartum depletion in PON1 activity and lipoprotein and lipid products concentrations, with elevated BHB values. However, there were no significant changes in SOD activity and uric acid concentrations in Holstein-Friesian and Polish Red breeds after calving. At lactation peak there was a significant SOD activity decrease correlated with standardized PON1 activity depletion in all examined breeds. The results suggest that PON1 might be a better parameter for minimal redox state changes in serum, shortly after labour in the examined breeds.

Changes in the level of cellular proteins in cells inoculated with equine influenza virus H7N7 and H3N8 were studied with microarray technique. H3N8 induced pro-apoptotic proteins while H7N7 induced both pro- as well as anti-apoptotic factors. The higher level of some cytoskeleton components and proteins involved in the protein quality control was recorded. Relatively high number of proteins involved in the regulation of transcription was down-regulated. The pattern of changes observed for H7N7 and H3N8 may reflect differences in the biological properties of both serotypes.

Disorders of sex development (DSD) are rare in cats. They can be caused by chromosomal aberrations, gene mutations or other undefined factors. The aim of the present study was to compare the histological structure and immunohistochemical reactivity of testes in cats with DSD and in healthy cats. The research material consisted of the gonads of four cats – phenotypic males with an incorrect structure of the reproductive system. The control group consisted of the testes of four healthy cats – routinely castrated phenotypical males. The material was fixed with formalin and embedded in paraffin; the sections were stained with hematoxylin and eosin. The immunohistochemical investigation were performed using monoclonal and polyclonal antibodies directed against desmin, vimentin, actin of smooth muscles, S100 protein and MCM3 protein. The results obtained allow concluding that the testes of cats with DSD differed in certain respects, mainly in the number of blood vessels, from the normal testes. Moreover, the results of immunohistochemical examination indicate that in the testes of cats with DSD the number of supporting cells is lower, the amount of interstitial cells is comparable and spermatogenesis is correct es compared to those determined in the control gonads. The number of blood vessels in cats with DSD is reduced by about 30%.
