Opis:
Porcine reproductive and respiratory syndrome (PRRS) has become one of the most economically important diseases for the swine industry worldwide. The objective of the study was to determine selected blood antioxidant enzymes (glutathione peroxidase (GPX), superoxide dismutase (SOD)), biochemical and haematological parameters in PRRS positive and negative pigs of three different categories, mainly to test oxidative stress hypothesis in pigs naturally infected with PRRS virus. Ninety PRRS positive and 90 PRRS negative pigs were included in the study. The presence of PRRS was confirmed by serological detection of antibodies against PRRS virus (PRRSV) and detection of PRRS viral RNA by RT-PCR. Pigs were further divided into three groups of 30: piglets just before weaning (weaners), fatteners and finishers. Blood samples for determining selected blood parameters were collected from the vena cava cranialis. Significantly (P < 0.05) higher activities of SOD in weaners and fatteners and of GPX in weaners were determined in PRRS positive pigs than in corresponding groups of PRRS negative pigs. In contrast, significantly (P < 0.05) lower GPX activity was observed in finishers of PRRS positive pigs than in the corresponding group of PRRS negative pigs. Concentrations of serum total protein in PRRS positive weaners and fatteners were significantly (P < 0.05) higher than those found in PRRS negative pigs. Leukopenia was observed in all three groups of PRRS positive pigs. It has been demonstrated, for the first time, that oxidative stress might be increased in PRRSV naturally infected pigs, especially in weaners.

Opis:
The process of vitrification of the cumulus-oocyte complex (COCs) often results in cold shock. When warming, heat shock occurs which can disrupt the balance of intracellular calcium (Ca2+) intensity. Drastic changes in temperature cause Reactive Oxygen Species (ROS), affecting changes on Ca2+ in COCs. The role of calcium is needed for oocyte activation in the fertilization process. The purpose of this study was to measure the expression of Ca2+ and the intensity of Ca2+ in COCs after vitrification. The study was divided into 2 groups, the control group (C) of fresh COCs, and the treatment group (T) of COCs after vitrification. After vitrification for 24 hours, then thawing, the expression of Ca2+ was examined using the Immunocytochemistry (ICC) method and the intensity of calcium (Ca2+) with a Confocal Laser Scanning Microscope (CLSM). The research data obtained were analyzed statistically by T-Test. The results showed that the expression of Ca2+ in the control group (12.00±0.00) was different from the treatment group (0.35±0.79). The intensity of Ca2+ in the control group (1059.43±489.59) was different from the treatment group (568.21±84.31). The conclusion of this study is that cryopreservation affects calcium in COCs; there were differences in the expression and the intensity of Ca2+ between fresh COCs and COCs after vitrification. Ca2+ intensity of COCs after vitrification was concentrated in the nucleus, while in fresh COCs it was concentrated in the cytoplasm.

Opis:
The transition from late gestation to early lactation is a critical period in a dairy cow’s life so that dairy cows undergo tremendous changes during this period. The aim of this study was to determine blood levels of ghrelin, leptin, glucose, β-ydroxybutyrate (BHB), non-esterified fatty acids (NEFA), triglycerides (TG), triiodothyronine (T3) and thyroxine (T4) in dairy Holstein cows (n = 20) and their correlations during the transition period. Blood samples were collected weekly from 3 wk antepartum to 6 wk postpartum from 20 high-yielding Holstein-Friesian cows. Ghrelin and leptin of plasma and glucose, BHB, NEFA, TG, T3, T4 of serum were then measured. Early lactation cows showed significantly higher (p<0.05) values of ghrelin, BHB and NEFA, and lower levels of leptin, TG, T3 and T4 (p<0.05) compared to late dry cows. Serum concentrations of glucose did not differ significantly at any time (P>0.05). Plasma ghrelin concentrations showed positive correlations with the serum BHB and NEFA (p<0.01), while plasma ghrelin had negative correlations (p<0.01) with leptin, TG, T3 and T4. In addition, no significant correlation (p>0.05) was found between ghrelin and glucose. The results of the study showed that blood ghrelin, leptin, BHB and NEFA levels are sensitive indicators of the energy balance during the peri-partum period in dairy cows and glucose values may not be considered as a precise indicator of negative energy balance in dairy cows.

Opis:
The zebrafish (Danio rerio) has become known as an excellent model organism for studies of vertebrate biology, vertebrate genetics, embryonal development, diseases and drug screening. Nevertheless, there is still lack of detailed reports about usage of the zebrafish as a model in veterinary medicine. Comparing to other vertebrates, they can lay hundreds of eggs at weekly intervals, externally fertilized zebrafish embryos are accessible to observation and manipulation at all stages of their development, which makes possible to simplify the research techniques such as fate mapping, fluorescent tracer time-lapse lineage analysis and single cell transplantation. Although zebrafish are only 2.5 cm long, they are easy to maintain. Intraperitoneal and intracerebroventricular injections, blood sampling and measurement of food intake are possible to be carry out in adult zebrafish. Danio rerio is a useful animal model for neurobiology, developmental biology, drug research, virology, microbiology and genetics. A lot of diseases, for which the zebrafish is a perfect model organism, affect aquatic animals. For a part of them, like those caused by Mycobacterium marinum or Pseudoloma neutrophila, Danio rerio is a natural host, but the zebrafish is also susceptible to the most of fish diseases including Itch, Spring viraemia of carp and Infectious spleen and kidney necrosis. The zebrafish is commonly used in research of bacterial virulence. The zebrafish embryo allows for rapid, non-invasive and real time analysis of bacterial infections in a vertebrate host. Plenty of common pathogens can be examined using zebrafish model: Streptococcus iniae, Vibrio anguillarum or Listeria monocytogenes. The steps are taken to use the zebrafish also in fungal research, especially that dealing with Candida albicans and Cryptococcus neoformans. Although, the zebrafish is used commonly as an animal model to study diseases caused by external agents, it is also useful in studies of metabolic disorders including fatty liver disease and diabetes. The zebrafish is also a valuable tool as a model in behavioral studies connected with feeding, predator evasion, habituation and memory or lateralized control of behavior. The aim of the present article is to familiarize the reader with the possibilities of Danio rerio as an experimental model for veterinary medicine.

Opis:
A significant number of cases of clinical canine epilepsy remain difficult to control in spite of the applied treatment. At the same time, the range of antiepileptic drugs is increasingly wide, which allows efficient treatment. In the present paper we describe the pharmacodynamics and pharmacokinetics of the newer antiepileptic drugs which were licensed after 1990 but are still not widely used in veterinary medicine. The pharmacokinetic profiles of six of these drugs were tested on dogs. The results of experimental studies suggest that second generation antiepileptic drugs may be applied in mono- as well as in poli- treatment of canine epilepsy because of the larger safety margin and more advantageous pharmacokinetic parameters. Knowledge of the drugs’ pharmacokinetics allows its proper clinical appliance, which, in turn, gives the chance to improve the efficiency of pharmacotherapy of canine epilepsy.

Opis:
This study was designed to assess alterations of hormone and cytokine levels associated with growth period during puberty in Honamli goats which were identified as a new goat breed and had one of the highest meat production potential among the other goat breeds in Turkey. Honamli goats are originated from native hair goats, so parallel studies of sampling and analyzing were conducted also in native hair goats which have moderate meat production. Blood serum samples of Honamli (n=90) and native hair goats (n=90) were obtained from the pure herds in Korkuteli and Ka districts of Anatolia. Concentrations of growth hormone (GH), myostatin (MSTN), insulin-like growth factor (IGF), growth hormone releasing hormone (GHRH), growth hormone releasing peptide (GHRP), leptin, transforming growth factor-betal (TGF-ß1) and vascular endothelial cell growth factor (VEGF) levels were measured by ELISA in each breed in the age groups of 4, 8 and 12 months. The present results indicate interesting correlations among the age groups and all the examined hormone and cytokine parameters exhibited significant (P<0.05 and P<0.001) differences. The parameters investigated were usually begun to increase after 4 months of age in the both breeds and sexes. Therefore, this paper supported the view that the beginning of hormonal alterations of goats could occur at 4th month of age. The results reported here emphasize the primary role played by GH, MSTN, IGF-1, leptin, GHRH, GHRP, TGF-ßi and VEGF in the first year growth period of goats.

Opis:
The purpose of the present review was a comparison of the abortions caused by EAV and EHV-1 viruses over the 34 years. A total of 452 tissues samples from aborted fetuses (347) or foals (105) stillborn or newborn that died within 72 hours were investigated. The material for the examinations came from different farms located throughout Poland. The tissue homogenates were examined by using virus isolation test in RK-13 and Vero cell lines and the cytopathic agent was confirmed as EHV-1 by the direct fluorescent antibody test or as EAV by the indirect fluorescent antibody test. The study indicated that EAV was isolated (104 cases, 23%) almost as equally often as EHV-1 (116 cases, 25.6%). Both, equid herpesvirus-associated abortion and the abortion induced by EAV were characterized by cyclicity. The percentage of EAV and EHV-1 isolation alternately reduced and increased, but the increase of isolation of one virus was accompanied by the decrease of the other. The domination of one virus over the other occurred in cycles of a few years.

Opis:
The effect of dietary Echinacea purpurea (EP) on the response of zebrafish (Danio rerio) to a Flavobacterium columnare vaccine was investigated. Two hundred D. rerio with an average weight of 290 ± 40 g were selected and fed different levels of E. purpurea (5 g kg⁻¹ diet - group 1, 10 g kg⁻¹ diet - group 2, 20 g kg⁻¹ diet - group 3, 30 g kg⁻¹ diet - group 4, and 0 g kg⁻¹ diet - group 5). Experimental feeding was begun 3 weeks prior to bath immunization and continued until the end of the experiment. Twenty-eight days after immunization the fish were challenged by bath immersion with F. columnare at a concentration of 1x106 CFU/ml. The relative percent survival of the experimental groups (1, 2, 3, 4 and 5) was 5.0, 6.0, 30.0, 36.0 and 5.0, respectively. In conclusion, diet supplementation with E. purpurea may effectively enhance the response of zebrafish to a F. columnare vaccine.
