The aim of this paper was to determine effects of selected genetic and environmental factors on the concentration of acetone in blood of cows in the first trimester of lactation. The study included 124 Polish Holstein-Friesian primiparous cows born, reared and managed on the same farm. The samples of blood were collected on day 2, 5, 30, 60 and 90 after calving. The highest serum acetone concentration in the animals examined was observed on day 5 after calving. The level of this compound was inversely related to the contribution of the original Holstein-Friesian breed in gene pool. Significantly higher serum acetone level was observed in cows calved in January-March period when compared to other months of the year. Besides, the level of acetone determined on day 5 after calving was associated positively with body weight determined at the same day and negatively with body weight changes between days 5 and 60 after calving.

The aim of this study was to determine the degree of Anguillicoloides crassus infection in European eel inhabiting Polish waters based on selected parasitic descriptors and on anatomical pathology of the swimbladder using macroscopic methods. In all, 154 European eel specimens were sampled from eleven sites in Poland and A. crassus was present in the swimbladder of 114 fish. The intensity of A. crassus infection in all the eel specimens ranged from 1 to 62 parasites at a mean value of 7.5. High values of mean infection intensity were noted in samples from Pomeranian lakes Bukowo, Łebsko, and Jamno. The health of the swimbladder was evaluated using the swimbladder degenerative index (SDI). The mean value of the SDI for all of the eel examined was 3.3, and extensively degenerated swimbladders were observed mainly in samples in the Szczecin Lagoon and from lakes. According to the individual SDI ratings, 9.1% of the eel specimens did not exhibit pathological symptoms of the swimbladder (SDI-0) and an extremely damaged (SDI-6) swimbladder was noted in 11.7% of the fish examined. In the case of eel infected with A. crassus, higher SDI values were reflected in initially increasing shares in subsequent categories. In fish that were not infected with the nematode, only 20% (8 individuals) of the swimbladders showed no symptoms of pathology (SDI-0).

The synthetic unmethylated oligodeoxynucleotides with CpG motifs (CpG ODN) were shown to activate Toll-like receptor 21 (TLR21) and stimulate the innate and adaptive immune system. In this study we tested the expression of TLR21, interferon (IFN)-γ and interleukin (IL)-1β mRNA in the blood after subcutaneous and intraocular application of the class B CpG ODN in chicken. The relative expression of mRNA of TLR21, IFN-γ and IL-1β were quantified at 3, 6, 12, 24 and 72 h post-stimulation. The study revealed that IFN-γ mRNA expression was significantly upregulated 12 h after subcutaneous stimulation with a high and low dose of ODN CpG, whereas the IL-1β mRNA expression levels were significantly upregulated 3 and 72 h after subcutaneous administration. After intraocular administration, the IL-1β mRNA levels were the highest at 24 h post-application, albeit not specifically. This data indicates that class B CpG ODN has the ability to induce TLR21 response in blood when administered parenterally in chicken. In contrast, intraocular administration of CpG ODN was not able to produce a significant increase in cytokine mRNA expression in blood. The data suggest that additional stimulus, e.g. the antigen, may be needed on the site of mucosal administration to activate systemic immune response.

Retroviruses comprise a large group of enveloped RNA viruses which have been found in a wide range of vertebrate species including fish. To date a number of fish retrovirus genomes have been partially or completely sequenced. Phylogenetic analysis and genome organization indicate a high diversity of fish retroviruses as well as some unique structural features that have not been found in any other retroviruses. Piscine retroviruses comprise both exogenous and endogenous viruses; most of them are associated with proliferative diseases. Because several of these proliferative diseases have a seasonal trend, they provide an excellent model for studying tumor development and regression. The aim of this work was to review the best-described fish retroviruses.

Metallic nanoparticles, mainly silver ones, have been widely used as antibacterial agents, and some studies shown they also exert direct antiviral activity against both enveloped and non-enveloped viruses. The objective of this study has been to evaluate the virucidal activity of commercial silver, gold, copper and platinum nanocolloids, recommended by the manufacturer as antimicrobials, against the ECBO virus, according to Polish Standard PN-EN 14675:2006. The highest experimentally observed decrease in the viral load was 0.875 log, which - when contrasted with the reduction in virus titre of at least 4 log expected from disinfectants - indicates that none of the analyzed nanocolloids had a disinfectant power towards the ECBO virus under the conditions defined by the standard.

The growing popularity of nanotechnology in the past decade has increased nanomaterial concentrations in the environment and the risk of their toxicity for aquatic organisms. Metal nanoparticles, which are easily absorbed and accumulated by fish, are probably able to interact directly with their immunocompetent cells. The objective of this study was to evaluate the in vitro effect of commercially available silver, gold and copper nanocolloids on the rainbow trout leukocyte and splenocyte activity. At high concentrations, all of the nanocolloids studied had adverse effects on the proliferative response of trout lymphocytes, and the most toxic of them, silver, decreased also the respiratory burst activity of splenocytes. Low concentrations of silver nanocolloid, however, had a stimulating effect on the lymhocyte proliferation.

The aim of the study was to present the results of comparative evaluation of the usefulness of PCR and microscopic methods for the detection of Processed Animal Protein (PAP) in feedingstuffs. In the validation study, the limit of the detection for PCR was determined on 0.05% for beef, 0.1% for pork and 0.2% for poultry meat and bone meal (MBM). Among 62 doubtful samples of feedingstuffs examined by microscopic method 41 (66.13%) were found as positive. Based on the results obtained with the use of the microscopic and PCR methods it is possible to state that the molecular biology methods can, at present, be used as a supplementary method in PAP detection.

Opis:
Vaccination is a most cost-effective way of controlling infectious diseases in fish. However, some vaccination techniques when applied to hatchery conditions are not as effective as we expect them to be. Modern molecular biology techniques offer a number of opportunities for improving existing bacterial or viral vaccines or creating new ones. One of the most promising trends in vaccinology is development of DNA vaccination. DNA vaccines are based on the gene encoding specific antigen, which is expressed in vaccinated organism and induces the host immune system. DNA vaccines, compared to conventional vaccines, have many advantages including ability to trigger wider immune response, bigger stability and possibility of large-scale production. To date, there are several reports indicating effectiveness of DNA vaccines used against fish pathogens.

The aim of the study was to monitor Y. enterocolitica strains from free-living animals captured during 2011-2014 hunting seasons in Poland using warm (ITC) and cold (PSB) enrichment and molecular examination. Over 1600 samples have been cultured. After ITC/PSB enrichment 237 strains presenting features characteristic for Y. enterocolitica were isolated. Molecular examination using multiplex PCR revealed 140 isolates from PSB and 78 from ITC. The concentration of pathogenic Yersinia in asymptomatic carriers is low and the PCR detection should be preceded by bacteriological examination.

The aim of the study was to assess the effect of different amounts of Yarrowia lipolytica yeast on the redox response and content of macro- and microelements in the blood plasma of turkey hens. The experiment was carried out on 240 turkey hens aged from 1 to 16 weeks. The birds were randomly assigned to 3 experimental groups of 80 birds each. Group I served as a control (K) and did not receive any experimental compounds. The turkey hens from the experimental groups (YL3 and YL6) were administered dried Yarrowia lipolytica yeast in their feed mixtures in the amount of 3% (YL3) or 6% (YL6). Yarrowia lipolytica yeast in the feed mixtures for the turkey hens did not induce oxidation reactions in the organism of the birds. However, an increase in catalase activity and a reduction in the level of LOOH , MDA and vitamin C were observed in the blood plasma of the turkey hens whose diet was supplemented with YL yeast. In the case of other indices, such as superoxide dismutase activity and total antioxidant potential (FRAP), the additive caused no significant changes. Administering Yarrowia lipolytica yeast to turkey hens may stimulate the enzymatic response of the antioxidant system (e.g. increasing catalase activity), mainly by increasing the concentration of iron in the plasma.

TLR stimulation in chickens has been shown to play a role in the initiation and regulation of innate and adaptive immune responses. The aim of this study was to use flow cytometry to establish the percentage of T and B subset in blood and lymphoid organs in chicks after CpG oligodeoxynucleotide (ODN) stimulation. It was demonstrated that the percentages of CD3⁺, CD4⁺, TCRγ δ⁺ cells and Bu-1⁺MHC class II⁺ cells in blood 24 h post-injection were significantly higher than in the control groups. It was also shown that the percentages of CD3⁺ and CD4⁺ cells in the spleen at 48 h post-injection were significantly higher than in control groups. The percentage of Bu-1⁺ cells in the bursa of Fabricius after CpG ODN stimulation (98.38 ± 0.84) was significantly higher than that found in the non-CpG ODN control group (94.54 ± 2.51) (p ≤ 0.05). The results indicate that class B CpG ODN increases the percentage of both T (especially CD4⁺ cells) and B cells.

The aim of this study was to investigate the levels of circulating immune complexes (CICs) and concentration of haptoglobin, fibrinogen and C-reactive protein in the serum of horses with recurrent airway obstruction and healthy controls. The study was conducted on a group of 14 adult Polish Konik horses, kept in uniform environmental and living conditions. Horses were divided into two groups: 7 horses were not affected by any respiratory problem (control group) and 7 horses had a history of recurrent airway obstruction (RAO) (study group). A clinical and laboratory evaluation, endoscopic examination and bronchoalveolar lavage (BAL) were performed in all horses. Levels of circulating immune complexes were significantly (p=0.0057) increased in heaves-affected horses compared to healthy controls (median [25th - 75th percentiles]) (3.96 [3.96 - 4.43] vs. 7.46 [5.13 - 11.9]). No significant difference was observed in the levels of the examined acute phase proteins between the groups. Moreover, all results were within the reference range established for horses. The results of this study indicate that heaves in horses is associated with the formation and high level of CICs. Haptoglobin, fibrinogen and C-reactive protein failed as markers of early stage systemic inflammation in the course of RAO.
