The objective of the study was to evaluate the effect of inulin or dandelion, chicory and Jerusalem artichoke powder on lipid indices and fatty acid profile in fattener tissues. The experiment involved 120 crossbred pigs (PL × PLW) × Duroc with an initial body weight of 25.0 ± 0.5 kg. Animals were assigned into 6 groups. A diet for group I (control) did not comprise an inulin additive, group II and III received 2% inulin (water or water-alcohol extraction of inulin from chicory root, respectively), while the mixtures for group IV, V and VI contained 4% root powder from Jerusalem artichoke (topinambur), chicory or dandelion, respectively. The animals were slaughtered at 115 kg body weight. The samples of blood, liver and the muscle longissimus dorsi tissues as well as whole hearts were collected for analysis. Fatty acid profile, some lipid indices and crude fat and cholesterol content were evaluated. Dietary supplement of 40 g dandelion powder resulted in preferable significant changes in the blood lipid indices and fatty acids composition (increased PUFA share and decreased n-6/n-3 ratio).

The aim of this study was to determine the effect of fish meal and mineral-vitamin premix, supplementing organic diets, on the performance of pigs and some meat and blood characteristics. The experiment was conducted on an organic pig fattening farm. The study involved 120 pigs with an approximate 25 kg body weight. Animals were divided into 3 groups, kept in pens, 10 animals each. Group I (control) animals were fed with plant feedstuffs of organic origin. Diets for group II and III were enriched with fish meal or fish meal and vitamin-mineral premix, respectively. The experiment was carried out till pigs reached a weight of 115 kg. Feed samples were subjected to laboratory analyses. Body weight (3 times) and feed intake were recorded. Blood samples were collected (2 times) to determine hematological and biochemical indices. Some parameters in meat samples were also determined. The fish meal addition improved (P ≤ 0.05) the average daily gains as well as feed conversion ratio during fattening period and mineral-vitamin premix significantly (P ≤ 0.05) fortified fish meal influence. Fish meal supplement improved (P ≤ 0.05) also some carcass characteristics. Supplementation of the diet with premix additionally decreased (P ≤ 0.05) backfat thickness and increased share of meat in carcass. Fish meal improved (P ≤ 0.05) some meat characteristics and elevated content of some polyunsaturated fatty acids. An increase in hemoglobin, red blood cell, white blood cell and cholesterol level in blood of animals from both experimental groups were also found. The results obtained proved the usefulness of fish meal and mineral-vitamin premix in the fatteners nutrition based on organic diets.

The aim of this study was to determine the degree of Anguillicoloides crassus infection in European eel inhabiting Polish waters based on selected parasitic descriptors and on anatomical pathology of the swimbladder using macroscopic methods. In all, 154 European eel specimens were sampled from eleven sites in Poland and A. crassus was present in the swimbladder of 114 fish. The intensity of A. crassus infection in all the eel specimens ranged from 1 to 62 parasites at a mean value of 7.5. High values of mean infection intensity were noted in samples from Pomeranian lakes Bukowo, Łebsko, and Jamno. The health of the swimbladder was evaluated using the swimbladder degenerative index (SDI). The mean value of the SDI for all of the eel examined was 3.3, and extensively degenerated swimbladders were observed mainly in samples in the Szczecin Lagoon and from lakes. According to the individual SDI ratings, 9.1% of the eel specimens did not exhibit pathological symptoms of the swimbladder (SDI-0) and an extremely damaged (SDI-6) swimbladder was noted in 11.7% of the fish examined. In the case of eel infected with A. crassus, higher SDI values were reflected in initially increasing shares in subsequent categories. In fish that were not infected with the nematode, only 20% (8 individuals) of the swimbladders showed no symptoms of pathology (SDI-0).

Calretinin (CR) as a buffer and sensor protein plays an important role in regulatory processes of Ca²⁺ and anty-apoptotic cellular protection. In the present study, immunohistochemical peroxidase- antiperoxidase (PAP) method was used in order to determine the numbers, morphology, morphometry and distribution pattern of CR in neurons of the chinchilla’s claustrum (Cl) and endopiriform nucleus (EN). In Cl and EN the presence of several classes of neurons with different immunoreactivity to CR was found. In Cl, CR-immunoreactive (IR) neurons were predominantly found in close vicinity to insular border while CR-IR neurons were evenly scattered throughout EN. In general, immunoreaction to CR was observed in neuronal cytoplasm, nucleus and in fibres-like nerve extensions. Statistical analysis showed the differences between average large diameter as well as cross-sectional area of CR-IR neurons present in Cl and EN. It is suggested, that CR acting as a calcium binding protein may play a role in neuronal network. Further co-localization studies are necessary to fully elucidate neurophysiology and neuropathology of the chinchilla’s Cl and EN neurons.

Opis:
Haemophilus parasuis belongs to opportunistic microorganisms of undefined virulence. The purpose of the studies was to compare suitability of PCR/RFLP in our modification and ERIC PCR for epidemiological study of domestic strains of H. parasuis. The results were evaluated taking into account two different aspects: suitability of the tests for isolating the highest possible number of clone groups and subjective evaluation of the method judged with respect to the following criteria: difficulty, availability of equipment and reagents as well as time and cost of the study. The results obtained in the present study show that the two methods used for typing of H. parasuis had high discriminatory power. Taking into account this parameter it can be concluded that ERIC PCR is more suitable than PCR/RFLP. This justifies the use of ERIC PCR for routine epidemiological analyses of mentioned pathogen. Taking into account the complexity of method used, ERIC-PCR based on random amplification of DNA, proved to be comparable to PCR/RFLP. The last mentioned technique is relatively less expensive and labour-consuming, especially when diagnostic PCR method is used for the epidemiological studies.

Combination chemiotherapy is the current standard of care for dogs with lymphoma. Multidrug resistance is one of the most important factors contributing to the efficacy of chemiotherapy. The major protein responsible for this phenomenon is P-glycoprotein. Little is known about P-glycoprotein expression in particular subtypes of lymphomas. The aim of the study was evaluation of P-glycoprotein expression in various subtypes of canine lymphomas. Positive reaction with P-glycoprotein was found in 12/25 cases of various morphological subtypes of lymphomas, however, in 3/11 lymphomas the percentage of positively weakly stained cells was <10% and those tumors were also considered negative. Tumors with 10-50% P-glycoprotein positive cells were found in single cases of centroblastic and centroblastic-centrocytic tumors. In 5 lymphomas P-glycoprotein expression exceeded 50% of tumor cells. Those cases were found among centroblastic, centroblastic-centrocytic, lymphoblastic and Burkitt-like subtypes. Positive reaction was observed mainly in the cell cytoplasm, however, in some cases prominent perinuclear dot-like staining pattern was found. In 2 cases focal staining pattern comprised dominant type of immunolabelling. Among all lymphomas containing P-glycoprotein positive cells intensity of imunolabelling was assessed as weak (6/25), moderate (2/25) and strong (3/25). Our results indicate that P-glycoprotein expression is present in nearly one third of newly diagnosed canine lymphomas of different morphological subtypes including those most commonly occurring, such as cenroblastic lymphomas. Hence, determination of P-glycoprotein expression at the time of diagnosis could provide valuable information for the design of treatment protocols. Moreover, our results have shown that P-glycoprotein expression in canine tumors could be located in Golgi-zone.

A separate, cat-specific hospitalization room away from dogs is recommended to reduce stress in cats; however, this can be difficult for some hospitals to provide. In such cases, measures are undertaken to reduce the cat’s stress by providing a place to hide. However, inability to observe the cat’s condition may be an obstacle to providing veterinary care. The use of a one-way mirror to create a sheltered environment while allowing observation of the cats was assessed. Five healthy cats were assessed using the Cat Stress Score (CSS) while in a cage with either a transparent panel or a one-way mirror. No significant differences in the CSS between the transparent panel and one-way mirror were observed. Variations in the CSS scores depended on the cat’s personality, with friendlier and more sociable cats showing a lower CSS with the one-way mirror. A one-way mirror may be useful to reduce stress in hospitalized cats.

Helicobacter pullorum ( H. pullorum) is a bacterium that colonizes the intestines of poultry and causes gastroenteritis. Because these species are known as human and/or animal pathogens, identification of H. pullorum is becoming increasingly necessary. The bacterium has been linked to colitis and hepatitis in humans after being transmitted by infected meat consumption. Misdiagnosis of other enteric zoonotic pathogens such as Campylobacter and other Helicobacter species makes the diagnosis of H. pullorum extremely difficult. This study focused on the molecular detection of H. pullorum from the stomach (proventriculus and gizzard) of different avian species as new target organs for detection and transmission between avian species. Proventriculus and gizzards were obtained from 40 freshly dead chickens and resident wild birds (n=40). Diarrhea was found in the farms that were surveyed. DNA was extracted from all collected samples to conduct PCR amplification. The samples were screened for Helicobacter genus-specific 16s using C97 and C05 primers. To confirm the existence of H. pullorum, the positive samples were sequenced. H. pullorum was recorded in two out of 40 chicken samples. In addition, H. pullorum was recorded in one out of 40 resident wild birds. The 16S rRNA gene sequence for Helicobacter genus-specific in poultry and wild birds showed a 100% homology. In conclusion, broiler chickens and resident wild birds are possible reservoirs for H. pullorum, according to this report, and possibly act as a source of infection for humans via the food supply.

Bovine Rotavirus and Bovine Coronavirus are the most important causes of diarrhea in newborn calves and in some other species such as pigs and sheep. VP8 subunit of rotavirus is the major determinant of the viral infectivity and neutralization. Spike glycoprotein of coronavirus is responsible for induction of neutralizing antibody response. Studies showed that immunoglobulin of egg yolk (IgY) from immunized hens has been identified to be a convenient source for specific antibodies for using in immunotherapy and immunodiagnostic to limit the infections. In this study, chimeric VP8-S2 gene was designed using by computational techniques. The chimeric VP8-S2 gene was cloned and sub-cloned into pGH and pET32a (+) vectors. Then, recombinant pET32a-VP8-S2 vector was transferred into E. coli BL21 CodonPlus (DE3). The expressed protein was purified by Ni-NTA chromatography column. Hens were immunized with the purified VP8-S2 protein three times. IgY was purified from egg yolks using polyethylene glycol precipitation method. Activity and specificity of anti-VP8-S2 IgY were detected by dot-blotting, Western-blotting and indirect ELISA. We obtained anti-VP8-S2 IgY by immunizing hens with the recombinant VP8-S2 protein. The anti-VP8-S2 IgY was showed to bind specifically to the chimeric VP8-S2 protein by dot-blotting, Western-blotting analyses and indirect ELISA. The result of this study indicated that such construction can be useful to investigate as candidates for development of detection methods for simultaneous diagnosis of both infections. Specific IgY against the recombinant VP8-S2 could be recommended as a candidate for passive immunization against bovine rotavirus and bovine coronavirus.

In the present study we investigated the effect of bovine conglutinin on the phagocytic activity of leukocytes. We measured both the chemotactic activity of conglutinin and its effect on the internalization of zymosan particles and E. coli by granulocytes. We also assessed the binding of conglutinin to various microorganisms isolated from clinical cases in cattle. We showed that conglutinin binds strongly to the surface of yeast cells and to mannan-rich zymosan particles, while weak binding was observed in the case of the bacterial strains tested, including those whose O antigen is composed of mannan. Conglutinin (1-10 μg/ml) neither acts as a chemotactic factor for peripheral blood leukocytes nor affects ingestion of E. coli by granulocytes. However, as flow cytometry based assay showed, conglutinin (0.1-1 μg/ml) increased ingestion of zymosan expressed as mean fluorescence intensity (MFI) of positive cells.
