Opis:
Bacterial adhesion is an important step in tissue colonization and depends extensively on the surface properties of a bacterial cell. For many microorganisms the prerequisite for host body occupancy is a break in tissue continuity. The next step is ongoing tissue destruction by products of bacterial metabolism: microbial enzymes and toxins. This happens, for example, in the initial phase of periodontitis. The mechanisms of adhesion are related to the specific structures present on the bacterial cell surface. This article summarizes recent data about bacterial attachment to host cells.

Opis:
Porcine circovirus type 2 (PCV2) is an economically important swine pathogen and, although small, it has the highest evolution rate among DNA viruses. Commercial PCV2 commercial vaccines are inactivated PCV2 isolates or a subunit vaccine based on the Cap protein of PCV2. Currently, PCV2 VLPs of individual subtype vaccines are available. Although the main prevalent genotype worldwide is PCV2b, the emerging subtype PCV2d subtype is also increasingly associated with PCV disease. The aim of the study was to evaluate the protective efficacy of VLP based on the PCV2b and 2d subtypes against the mixed challenge of two hypotype PCV2 in mice. Thirty-six female SPV Kunming mice were immunized twice with PCV2b and 2d VLPs, then challenged with PCV2b and PCV2d, to assess the immunogenicity and effectiveness of the VLPs. Vaccination of the mice with PCV2b and 2d VLPs elicited a robust antibody response specific for the PCV2. The virus load detected in the 2b and 2d spleen vaccine group was the lowest compared to other groups. Furthermore, there was no pathological damage in the HE stained sections of the 2b and 2d spleen vaccine, and no virus was detected in the immunohistochemical sections. Our data suggest that the mixed PCV2b and 2d VLP vaccine could protect mice from challenge with the mixed infection of PCV2b and PCV2d.

Opis:
The purpose of the present review was a comparison of the abortions caused by EAV and EHV-1 viruses over the 34 years. A total of 452 tissues samples from aborted fetuses (347) or foals (105) stillborn or newborn that died within 72 hours were investigated. The material for the examinations came from different farms located throughout Poland. The tissue homogenates were examined by using virus isolation test in RK-13 and Vero cell lines and the cytopathic agent was confirmed as EHV-1 by the direct fluorescent antibody test or as EAV by the indirect fluorescent antibody test. The study indicated that EAV was isolated (104 cases, 23%) almost as equally often as EHV-1 (116 cases, 25.6%). Both, equid herpesvirus-associated abortion and the abortion induced by EAV were characterized by cyclicity. The percentage of EAV and EHV-1 isolation alternately reduced and increased, but the increase of isolation of one virus was accompanied by the decrease of the other. The domination of one virus over the other occurred in cycles of a few years.

Opis:
Canine mammary tumors are the most common neoplasms in intact female dogs. The surgery cannot always solve the problem, chemotherapy are recommend to these patients. However, chemotherapy could always fail because of multidrug resistance (MDR). Through stepwise increasing 5-Fluorouracil (5-FU) concentration in the culture medium, a 5-FU-resistant canine mammary tumor cell line CMT7364/5-FU was established to disclose the molecular mechanism of the drug resistance. Cell morphology, cell sensitivity to drugs, growth curves, expression of proteins, and chemo-sensitivity in vivo were compared between the parental cell line and resistant cell line. As compared it to its parental cell line (CMT7364), CMT7364/5-FU showed different morphology, cross-resistant to other chemo-drugs and a prolonged population doubling time (PDT). The drug efflux pump proteins (ABCB1 and ABCG2) in CMT7364/5-FU were up-regulated. In vivo, the similar result revealed that CMT7364/5-FU cell line was more resistant to 5-FU. In conclusion, a 5-FU-resistant canine mammary tumor cell line (CMT7364/5-FU) was successfully established, it can serve as a good model for researching the mechanism of MDR and screening effective agents to reverse drug resistance.

African Swine Fever Virus (ASFV)

Opis:
African swine fever (ASF) is an acute, hemorrhagic, and devastating viral infectious disease that causes important economic losses to the swine industry. Currently, there are no effective vaccines or drugs available. Epigenetic mechanisms, especially cytosine methylation of cytosine-phosphate-guanine (CpG) islands, have a significant impact on the life cycle of several viruses. Hence, drugs targeting DNA methylation may potentially be used for the treatment of ASF. Here, we selected the inner core, core shell, inner membrane, capsid, and external envelope membrane, to analyze the characteristics of CpG islands in the ASF virus (ASFV) genomes. Furthermore, we analyzed the promoters and CpG islands in the upstream regions of these genes. Results showed that the CpG islands of seven genes were conserved in the genomes of two genotype of ASFV strains, whereas the CpG islands of other genes were relatively conserved (ASFV strains differed mainly in the quantity of CpG islands). The different distribution of CpG islands in the genomes of different ASFV strains may affect their methylation status, which may in turn affect the regulation of viral gene expression, leading to different clinical outcomes. In addition, the predicted promoter regions based on the upstream sequences of most genes overlapped with CpG island positions. Methylation of the binding sites of the promoter regions inhibits the binding of the transcription factors to the promoters, thus inhibiting the activation of the promoters and limiting the synthesis of viral proteins. The results of this study provide a basis for exploring new antiviral therapeutic strategies from an epigenetic perspective.

Opis:
There is no special trait for differentiation of Avian Pathogenic Escherichia coli from Avian Fecal Escherichia coli. This investigation is aimed, as a case control study, to evaluate and compare the frequency of iss and irp2 in 43 AFEC strains and also 40 and 56 E. coli strains isolated from the liver and kidney of chickens with colibacillosis, respectively, farmed in Zabol, as a border region of Iran, by PCR. 86.9% and 37.2% of isolates collected from chickens with colibacillosis and feces samples obtained from healthy chickens were positive for iss gene, respectively (P<0.05). On average, 59.3% of E. coli strains isolated from colibacillosis have irp2 gene while 27.9% of isolates from the feces of healthy birds were positive (P<0.05). 52.15% of isolates from colibacillosis and 19.62% of isolates from healthy chicken feces were positive for both genes, with statistical significant difference (p<0.05). This marked difference in the distribution of iss and irp2 genes makes these two genes good markers to differentiate AFEC and APEC strains especially in Sistan region to improve colibacillosis control measurements.

Buffalopox (BPX) is a highly contagious disease that causes high morbidity and production losses in buffaloes. During this study, seroprevalence, effect of various associated risk factors, and pathological studies of BPX were recorded in the Punjab province. A total of 97 blood samples and 63 scabs were collected from clinically pox suspected buffaloes. Serum was harvested to perform single radial hemolysis to assess the seroprevalence, and scabs were subjected to PCR for BPX virus confirmation. Results revealed that, animal demographics and environmental associated factors showed significant effect (p<0.05,1<R2>0) on BPX occurrence. The overall BPX seroprevalence was recorded 4.18% in the Punjab province. The BPX was recorded 5.48% in Nili Ravi breed during winter (7.42%), aged 5-7 years (7.46%) under loose housing (5.51%) in the Faisalabad region (8.03%). Further, BPX was 5.37% in pregnant, 6.86% pregnant milking buffaloes during the 3rd lactation period (7.28%) in dairy herds (5.20%). The BPX was 5.22% in non-vaccinated buffaloes where multiple animals were reared together (4.99%) in the herds having 21-30 total number of animals. A total of 49 scab samples were found positive for the BPX virus via PCR with C18L gene amplification. Grossly, inflammatory lesions with pits in the center and wart-like nodules were seen on teats and udder of buffaloes. Increased leukocytes, especially neutrophils and lymphocytes, were seen in the blood of the infected animals. These results provide a broader window to understand the effect of associated risk factors, strengthen the diagnostic aid, and to contain the current spread of BPX in Pakistan to safeguard large ruminant-based livelihood.

Opis:
The present study was done to evaluate the effect of boldenone undecylenate (BOL) on growth performance, maintenance behaviour, reproductive hormones and carcass traits of male rabbits. Sixty apparently healthy New Zealand White male rabbits, 5 weeks of age, were allotted to 3 equal groups. Each group was subdivided into 5 replicates, where the first group is control. The second group (B1) comprised rabbits that received 2 intramuscular injections of BOL (5 mg/kg) with 3 week intervals (9 and 12 weeks of age), while the third group (B2) included rabbits that received 3 intramuscular injections of BOL (5 mg/kg) with 2 week intervals (8, 10 and 12 weeks of age). The end of the trial was after 4 weeks from the last injection (16 weeks of age). The results revealed that the treated groups had a significant increase in total body weight, daily gain and feed efficiency, with a significant decrease in feed conversion ratio (FCR). Ingestive, locomotion and grooming behaviors were significantly higher in treated groups. Lateral pasture and exploratory behaviors were significantly higher in the control group. Administration of BOL resulted in a significant increase in dressing % and a significant decrease in testes %. Groups treated with BOL had a significantly (P<0.05) decreased serum testosterone level, simultaneously with a significantly increased estradiol level. The results indicate that BOL improves performance and carcass traits. Furthermore, there are hormonal-behavioral correlations through enhancement of ingestive and locomotion behaviors of treated animals.
