Opis:
The aim of this study was to perform molecular analysis of canine adenovirus 2 (CAV-2) E1B 19K gene fragment isolated from 20 dogs of various breeds (12 males and 8 females aged 1-9 years), with clinical symptoms of upper respiratory tract infections, from the Lubelszczyzna region. Nasal swabs were taken from dogs. DNA of CAV-2 was detected using the PCR method in 16 swabs. All PCR products were sequenced, and the obtained sequences were compared with each other and with the sequence of the E1B 19K gene of the CAV-2 strain from an online database of NCBI GenBank: AC 000003. Based on analysis of the obtained sequences, three polymorphic variants of CAV-2 (No.1-3) with homology of 78 - 100% were distinguished. The nucleotide and amino acid sequences of the most frequently represented polymorphic variant, No. 1, differed from the sequences of polymorphic variant No. 2 with one substitution. The nucleotide and amino acid sequence of the E1B 19K gene of CAV-2 AC 000003 differed from the analogous sequences of representatives of variant No. 1 with 44 nucleotide and 19 amino acid substitutions. The small number of nucleotide differences in the E1B 19K CAV-2 gene among the examined own isolates, compared with AC 000003, suggest that the infections in dogs were caused by a relatively genetically stable virus which occurs in eastern Poland.

The objective of this study was to estimate a herd-level seroprevalence of bovine herpesvirus type 1 (BHV-1) in herds with clinical symptoms of the respiratory tract. Eighty-three herds with suspected BHV-1 infection were selected and divided into two categories with respect to their size: small (n=27) and large herds (n=56). Samples were collected from calves, heifers and cows older than 24 months. Seroprevalence was determined using the gB ELISA test. The herd level seroprevalence was estimated as 53% (44/83) in the tested herds, 11.1% (3/27) in the small herds and 73.2% (41/56) in the large herds. Our study suggests that the current biosecurity measures still warrant improvement.

In this study, a pigeon paramyxovirus type 1 (PPMV-1) isolated from a flock of ornamental pigeons in Poland in 2010 is described. The PPMV-1/Poland/H2/10 isolate showed the amino acid sequence at the cleavage site of F2/F1 ¹¹²KRQKRF¹¹⁷ i.e. typical of virulent strains. Despite having the monoclonal antibody binding pattern typical of pigeon variants PPMV-1 (antigenic group “P”), the Polish isolate clustered into genetic sublineage 4a, which is usually associated with PMV-1 isolated from poultry.

A highly sensitive and specific real-time PCR assay was used for detection and quantitation of equine herpesvirus type 1 (EHV-1) in the different internal organs of aborted fetuses. Tissue samples from 23 aborted fetuses submitted to the Department of Virology of the National Veterinary Research Institute in Pulawy between 2012 and 2013 were used for testing. Total DNA was extracted using a phenol-chloroform-isoamyl alcohol standard protocol. A real-time PCR with forward and reverse primers encompassing a highly conserved region encoding viral glycoprotein B was adapted for diagnosis of EHV-1 infection. The detection limit of the assay was shown to be 6.0x100 of viral DNA copies and the obtained standard curve exhibited a linear range from 100 to 107 molecules. Sixteen out of twenty three aborted fetuses (69.5%) were positive for EHV-1 in real- time PCR. The highest EHV-1 DNA load was obtained for liver (mean Ct value: 15.7) and lung (18.2) samples, while the lowest was in the thymus (29.6) and placenta (28.4).

GALT induces tolerance to foreign food antigens and plays an important role in the development of food allergies and the inflammatory bowel disease. The immune function of GALT is significantly influenced by an equilibrium between Th1 and Th2 subpopulations and the cytokines they produce. Th1 cytokines participate in the induction of a cell-mediated immune response, whereas Th2 cytokines induce powerful antibody-mediated responses. Changes in Th1/Th2 cell polarization of an immune response are associated with susceptibility to autoimmune and infectious diseases. This experiment investigated changes in cytokine levels produced by Th1 and Th2 cells in ileal Payer’s patches in gilts exposed to ZEN doses below the NOEL (approximately 8 μg kg⁻¹ BW) for 14, 28 and 42 days. A significant linear increase in IL-4 (40.32 ± 1.55 ng mg⁻¹ - 137.60 ± 29.96 ng mg⁻¹), and IL-10 (5.99 ± 0.15 ng mg⁻¹ - 16.39 ± 1.11 ng mg⁻¹) concentrations was observed. An increase in Th1 (IL-2 and IFN-γ) cytokine levels was also noted in the experimental group, but it was not statistically significant. An HPLC analysis of Peyer’s patches in group E animals revealed a linear increase in ZEN concentrations (3.65 ± 0.91 ng g⁻¹ - 4.72 ± 1.85 ng g⁻¹) and an absence of α-ZEL. IL-4 stimulates monocytes and macrophages, it induces the production of proinflammatory cytokines and it may directly and indirectly contribute to the development of inflammatory foci. Higher IL-4 levels could shift polarization toward Th2 cells, stimulate B cells to undergo class switching to produce IgE and contribute to the development of allergies.

The presence and distribution of vasoactive intestinal polypeptide (VIP) receptor VPAC1 was studied in the ovary, oviduct and uterus (uterine horn and cervix) of the domestic pig using methods of molecular biology (RT-PCR and immunoblot) and immunohistochemistry. The expression of VPAC1 receptor at mRNA level was confirmed with RT-PCR in all the studied parts of the porcine female reproductive system by the presence of 525 bp PCR product and at the level of proteins by the detection of 46 kDa protein band in immunoblot. Immunohistochemical stainings revealed the cellular distribution of VPAC1 receptor protein. In the ovary it was present in the wall of arterial blood vessels, as well as in the ovarian follicles of different stages. In the tubular organs the VPAC1 receptor immunohistochemical stainings were observed in the wall of the arterial blood vessels, in the muscular membrane, as well as in the mucosal epithelium. The study confirmed the presence of VPAC1 receptor in the tissues of the porcine female reproductive tract what clearly shows the possibility of influence of VIP on the porcine ovary, oviduct and uterus.

The purpose of the study was to examine the mechanisms important for early myogenesis in mouse C2C12 myogenic cells exposed to interleukin-1β. Cyclin A and cyclin B1 were increased by interleukin-1β (1 ng/ml), but the level of cyclin D1 and total DNA content was unaffected. Fusion index and the rate of protein synthesis was increased in the presence of IL-1β, but these effects were limited to 3-day-treatment. IL-1β increased the level of MyoD, myogenin and MHC on the 3rd day of differentiation, without altering the content of the active form of myostatin, as well as it augmented the level of fibronectin, integrin β1 and full length 100 kDa form of ADAM12. IL-1β caused a decrease in IGFBP-4 and IGFBP-6 levels and a marked increase in IGFBP-5. The phosphorylation of PKB and ERK1/2 and the cellular content of p38 were elevated by IL-1β. We conclude that the myogenic effect of IL-1β was limited to the onset of myoblast fusion and was associated with: i) increase in the level of myogenic transcription factors i.e. MyoD and myogenin expression, ii) modification of extracellular matrix assembly and signaling, manifested by an increase in fibronectin, integrin-β1 and ADAM12 content, iii) drop in IGFBP-4 and IGFBP-6, and an increase in IGFBP-5, that could alter the local IGF-1 bioavailability, and iv) increase in phosphorylation of PKB and ERK1/2, and the expression of p38 kinase, leading to activation of intracellular pathways essential for myogenic differentiation.

High concentration of glutamate (Glu) is excitotoxic for nervous system structures. This may lead to glial reactivity ie. increased expression of glial fibrillary acidic protein (GFAP) and S100β protein, and also to hypertrophy and proliferation of cells which are determined by the presence of Ki-67 antigen. The aim of the study was to analyse the immunoreactivity of the GFAP, S100β and Ki-67 proteins in astrocytes of hippocampal CA1 region in young rats after administration of monosodium glutamate (MSG) at two doses: 2 g/kg b.w. (I group) and 4 g/kg b.w. (II group). In rats from I and II group morphologically altered astrocytes with the GFAP expression were observed in the SLM of the hippocampal CA1 region. The cells had eccentrically located nuclei and on the opposite site of the nuclei there were single or double, long and weakly branched processes. Moreover, in the SLM the increase of the number of GFAP and S100β immunopositive astrocytes and nuclei with Ki-67 expression, in contrary to control individuals, was observed. These results suggest the increased expression of the proteins in early reactions or hyperplasia which, together with cell hypertrophy, indicate late reactivity of astroglia in response to glutamate noxious effect.

From economical point of view, Dicrocoelium Dendriticum (D. dendriticum) causes a lot of damages to the livestock industry annually. So, the rapid diagnosis of infection is very important. The diagnosis is based on egg per count of feces (EPG) test because detection according to clinical symptoms is difficult. Since EPG is not accurate and sensitive, the serological methods become important for the diagnosis of this parasite as they are more accurate in comparison to EPG test and they are able to diagnose infection in a short time. In this study, somatic and Excretory-secretory antigens (EsAg) were isolated. The ELISA test was set up according to positive and negative sera and the results which were obtained compared to those obtained by the EPG test. The prevalence of infection in 550 samples by ELISA and EPG methods were 56% and 7% respectively, which shows the significant difference between these methods in examining the rate of infection. Based on the results, the specificity and sensitivity in ELISA test were 95% and 94%, respectively. The results showed that the ELISA is a more reliable test in comparison to EPG test for the rapid diagnosis of D. dendriticum infection.

The synthetic unmethylated oligodeoxynucleotides with CpG motifs (CpG ODN) were shown to activate Toll-like receptor 21 (TLR21) and stimulate the innate and adaptive immune system. In this study we tested the expression of TLR21, interferon (IFN)-γ and interleukin (IL)-1β mRNA in the blood after subcutaneous and intraocular application of the class B CpG ODN in chicken. The relative expression of mRNA of TLR21, IFN-γ and IL-1β were quantified at 3, 6, 12, 24 and 72 h post-stimulation. The study revealed that IFN-γ mRNA expression was significantly upregulated 12 h after subcutaneous stimulation with a high and low dose of ODN CpG, whereas the IL-1β mRNA expression levels were significantly upregulated 3 and 72 h after subcutaneous administration. After intraocular administration, the IL-1β mRNA levels were the highest at 24 h post-application, albeit not specifically. This data indicates that class B CpG ODN has the ability to induce TLR21 response in blood when administered parenterally in chicken. In contrast, intraocular administration of CpG ODN was not able to produce a significant increase in cytokine mRNA expression in blood. The data suggest that additional stimulus, e.g. the antigen, may be needed on the site of mucosal administration to activate systemic immune response.

The objective of the study was to evaluate the effect of energy contents in a dry period diets in “7+1” feeding strategy of dry cow including 7 weeks of the dry period (far-off) and 1 week of a close-up period, on colostrum quality and the immune status of calves. Forty Holstein multiparous cows were dried at 56d before the expected date of calving and were assigned to the higher energy diet group (HE; 0.69 UFL/kg DM, NDF 52% DM), or the lower energy diet group (LE; 0.61 UFL/1kg DM, NDF 56% DM). From -7d to the expected calving date up to 21 d of lactation, all cows were fed the same fresh, lactation diet. Samples of colostrum were collected within 2h after parturition and its density, dry matter content and concentrations of immunoglobulins, fat, protein, lactose, urea and somatic cell counts were measured. Calves were weighed 2 h after calving and on the 21d of life. On 3d and 21d of calves’ life blood samples were collected and the concentration of IgG, IgA, IgM, insulin-like growth factor 1 (IGF-1) as well as total protein and albumin concentrations were determined. Treatments had no significant effect on composition of colostrum and serum immunoglobulins and IGF-1 concentration. In both groups the weight of calves at birth was similar, still daily body weight gain during 21 day of rearing period in HE group was higher than in the LE group (P=0.05).

Most research studies investigating the estrogenic effects of zearalenone (ZEN) focus on the mycotoxin’s effect on the reproductive system. Since estrogen receptors are present on various types of immunocompetent cells, ZEN can also modify diverse immune functions. This study analyzed immunocompetent cells isolated from Peyer’s patches in the ileum of pigs administered ZEN in the estimated daily dose of 8 μg kg⁻¹ BW (equivalent of 100 μg kg⁻¹ feed per day⁻¹). The objective of the study was to determine whether long-term exposure to low ZEN doses below the NOEL threshold leads to changes in the percentages of lymphocyte subpopulations and cytokine secretion by Th1 (IL-2, IFN-γ) and Th2 (IL-4 and IL-10) lymphocytes in Peyer’s patches of the ileum after in vitro stimulation with lipopolysaccharides (LPS). Immunocompetent cells isolated from Payer’s patches on experimental days 0, 14, 28 and 42 were cultured in vitro and stimulated with LPS. The presence of IL-2, IFN-γ, IL-4 and IL-10 in culture media was determined by the ELISA method. The results of the study indicate that ZEN inhibits IL-2 and IFN-γ secretion and stimulates IL-4 and IL-10 production by Th1 and Th2 lymphocytes by shifting the Th1/Th2 balance towards the humoral immune response. The above can promote allergic responses, as demonstrated by the increase in the size of B1 cell populations producing more autoantibodies. ZEN can also lower resistance to viruses and tumors by inhibiting the proliferation of NK cells and IFN-γ secretion.
