Comet assay under neutral conditions allows the detection of DNA double-strand breaks, considered to be the biologically relevant radiation-induced lesion. In this report we describe modifications of the neutral comet method, which simplify and facilitate its use for estimation of DNA double strand breaks in human lymphocytes irradiated with doses of 60Co gamma-rays (from 10 to 100 Gy). The analysis carried out according to this protocol takes less time than those published so far. Also, the use of lysis at 50°C is avoided; this is important in view of the presence of heat-labile sites in the DNA of irradiated cells, recently reported by Rydberg [12]. The comets have well defined, sharp limits, are suitable for computer image analysis and chromatin of the control cells remains condensed.

Plating experiments have been performed in order to obtain fission recoil barriers on low-enriched uranium (LEU) metal foils used as targets suitable for the production of 99Mo. The results of Ni deposition obtained in electroplating and electroless plating baths are presented. The best method of pretreatment uranium prior to plating is suggested. It was found that etching in solutions containing NiCl2 gave better results on uranium rods, whereas electroplating produces coatings with the desired level of adherence and coverage on uranium foils. The thickness of the nickel plates, evaluated both microscopically and gravimetrically, is sufficient to act as a fission recoil barrier.

The application of internal electrolysis for the deposition of ruthenium-106 and iodine-125 on silver was examined. These investigations were aimed at the preparation of sealed radiation sources for brachytherapy. The current and voltage of a cell built of silver cathode and aluminium anode, immersed in electrolytes containing different concentrations of ruthenium were determined. The same parameters were measured for the cell consisting of silver anode and platinum cathode as a function of the concentrations of NaI and NaOH in the electrolyte. The changes of current and voltage of the cells as a function of duration of electrolysis were investigated. The influence of white light observed during the deposition of iodine indicates that photolysis of silver iodide formed on the anode affects the cell parameters. From the results obtained it follows that electrolysis performed without application of external electric tension is a simple and convenient method for fixing considerable amounts of both ruthenium-106 and iodine-125 on silver.

The presentation answers the frequent question about the practical usefulness, advantages and disadvantages of radon risk maps and detailed in-situ classification of radon risk. Czech Radon Programme derives the benefit from radon maps on various scales – 1:500 000, 1:200 000 and 1:50 000, as well as from the uniform method for direct detailed classification of radon risk. The reliability assessment of the practical usefulness is based on the direct comparison between the results obtained from detailed in-situ classification of radon risk of building sites and the corresponding reading from the radon risk map. Altogether almost one thousand of detailed radon risk assessments, i.e. tens of thousands of soil-gas radon concentration measurements, were compared with the expected radon risk categories in five radon risk map sheets on the scale 1:50 000. The new results more specify and correspond to the previous results from comparisons performed in 1992, 1995 and 2002. We can prepare quite consistent maps, which can be successfully used to direct the search of existing houses with higher indoor radon values. On the other hand, the risk of underestimation or overestimation in the case of deriving the radon risk classification of a specific building site from the map seems to be too high to use the maps for direct assessment of specific sites. For new buildings, it is recommended to use detailed in-situ measurements and classification.

Opis:
Adaptive response (AR), an enhanced resistance to a high dose of ionising radiation acquired after pretreatment with a very low dose, was estimated in normal human lymphocytes. The question posed was whether the extent of radioadaptation, assessed by micronucleus test, would be related to the rate of DNA double-strand break (DSB) rejoining. Phytohemagglutinin-stimulated G1-lymphocytes from 5 healthy male volunteers were pre-treated (or not) with an adaptive (5 cGy) dose of X-rays, followed by a higher (5 or 10 Gy) challenge dose after 20-22 h. DSB rejoining after the challenge dose was monitored with the use of two methods: neutral comet assay, modified to reduce the contribution of single-strand breaks (SSBs) and thermolabile sites, and pulse-field gel electrophoresis (PFGE), specific for DSBs. At the level of micronuclei, an AR was observed in lymphocytes of 3 of 5 donors. Up to 60 min, comet assay showed no statistically significant differences in DNA break rejoining between adapted and non-adapted lymphocytes, independently of AR appearance. PFGE gave similar results, although in three donors it revealed secondary increases in DSBs levels at 30 min and/or 60 min post-irradiation in the adapted vs. the non-adapted samples. Failure to demonstrate changes in DSBs rejoining rate in the adapted lymphocytes could be due to diversity of AR intensity/timing at the level of DNA repair in not fully homogenous cell populations. Also, “rare” DNA cuts characteristic of early apoptosis/necrosis could overlap the process of DNA break rejoining.

We compared the effects of bleomycin (BLM) and ionizing radiation on two sublines of murine lymphoma L5178Y (LY): LY-R, radiation resistant and LY-S, radiation sensitive. This radiosensitivity difference is related to the ability to rejoin DNA double strand breaks. LY-S cells were about two times more sensitive to BLM than LY-R, similarly as in the case of sensitivity to X rays. Since there was no difference in the P-glycoprotein-related drug transport system between the sublines, it could be expected that the enhanced sensitivity of LY-S cells to BLM was caused by the DNA repair defect. Growth disturbances in BLM treated cell populations were proportional to the lethal effect and their duration was observed until elimination of dead cells (3-6 days after 50 ěM BLM, 1 h at 37oC). There was no slow growth phase accompanied by normal viability, as previously described for X-irradiated LY-S cells. Initial DNA damage, estimated with the single cell gel electrophoresis method was linearly related to BLM dose in LY-S cells; in LY-R cells - in the low dose range (up to 10 ěM) - there was more damage than in LY-S cells, however, at higher doses the dose - effect curves became identical. The doseeffect relationship for ă rays was linear and identical in both cell sublines. DNA damage distribution in BLM treated cells was much less uniform as compared to that in irradiated cells and indicated the presence of cells with severely damaged DNA, a feature typical for BLM action in vitro.

Radiation sterilization has been considered a mass decontamination technique for biodegradable cultural heritage (CH) since its widespread application in the medical field. Initial experiments have revealed advantages, for example, efficiency and effectiveness, but also disadvantages, namely “side effects” concerning CH materials. More than 50 years later, the adequacy of ionizing radiation for some CH artefacts is still the subject of discussion. The main reason why is that science and industry are not yet able to provide a more efficient technique for treating mass decontamination. For wooden items, there is general agreement that the irradiation dose required for insect eradication is not damaging, even in the case of polychromed wood. For cellulose pulp (paper), there is a reduction in polymerization degree (DP) at the high doses necessary to stop the attack of fungi, but this should be considered taking into account the purpose of the treatment. Emergency or rescue treatments are necessary to mitigate the consequences of accidents or improper storage conditions. In some cases (archives), the value of written information is greater than the historical value of the paper support. For other materials, namely textiles, leather and parchment, less research has been published on the effect of ionizing radiation treatment. As a general rule, irradiation is not necessary when only a few CH elements are present that are affected by biological contamination since restorers can solve the problem by classical means. The need for radiation treatment arises when large collections (hundreds, thousands or even more elements) are heavily affected by the biological attack. In Romania, the IRASM gamma irradiator of IFIN-HH is receiving an increasing number of requests for CH treatment, mainly due to an intensive research programme concerning this topic and close liaison with CH owners or administrators. Besides reviewing the scientific results obtained in Romania and abroad, this paper presents some examples from experiences in Romania.

The difficulty in determining the electron paramagnetic resonance (EPR) line parameters of ferromagnetic semiconductors has been addressed. For these materials, the resonance line is very broad and lies at low resonance fi eld, so that only a part of the line can be detected experimentally. Moreover, the line is of asymmetric (Dysonian) shape as described by the line shape parameter α. We have compared values of line parameters derived by computer fi tting of the whole experimental EPR line to the Dyson function (or modified Dyson function) with the values obtained by applying this procedure to the left and the right half of the line.

This paper addresses the hot channel factors for the thermalhydraulic analysis of Material Testing Reactors (MTR). The criteria for choosing the hot channel factors and their combination methods are presented and discussed. A method for the evaluation of the hot channel factors for the thermalhydraulic analysis is proposed. In the proposed method, the values of the hot channel factor are calculated in terms of their fraction of variation and the degree of dependency of the thermalhydraulic safety parameter of interest on these factors. The equations for the calculation of the thermalhydraulic safety parameters in the hot channel are presented along with discussions. The application of the proposed method has been illustrated by an example.

In this paper the changes of membrane fluidity of liposome with additions of humic substances (humic and fulvic acids) were examined. Liposome were done by the sonication of lecithin EYL. Concentrations of humic substances in attitude to EYL varied between 0–10% of weight. The technique of electron spin resonance (ESR) were used for the examination followed by three spin probes with a variety placement of the membrane located. TEMPO probe melted in the hydrophobic membrane and in the aquatic solution which allowed to determine the spectroscopic partition parameter (F), indicating the changes that occur in water-lipid interphase. Probe 5-DOXYL placed directly under the heads of polar lipids and the order parameter measuring by the TII showed the changing of membrane fluidity at surface area. 16-DOXYL probe penetrated the middle of the lipid bilayer membrane and allowed to determine the rotational correlation time τ parameter, which gives us information about changing of the liquidity lipid bilayer. Studies showed that the tested humic substances significantly changed the membrane fluidity of liposome. The dynamics of this process depends on both: the fraction of humic substances and its quality and quantity as well as the placement area of the membrane.

Magnetic and Mossbauer measurements were performed for Mn2.xFexP0.5As0.5, 0.5 less-than or equal to x less-than or equal to 1.0. It was shown that with decreasing Fe content magnetic interaction significantly weakens. Low temperature magnetic structure could be explained as a superposition of ferromagnetic and antiferromagnetic contributions to the whole magnetic moment of the alloy.

The deposition of iodine-125 on silver bars to be sealed in titanium capsules as seed-type sources for brachytherapy was studied. For this purpose, a method of internal electrolysis was applied. The cell consisted of a silver anode contained in a Pt crucible serving as cathode. The plating solution contained sodium iodide-125 and sodium hydroxide. The yield of iodine-125 deposition for carrier-containing and carrier-free electrolyte solutions was determined. The deposition on uncutted Ag wires and on those cutted into 3 mm long bars was investigated. The influence of visible light on the coating process was examined. The leachability of iodine-125 from the silver wires as well as the activity distribution on the wires was measured. The obtained results indicate that the silver wire should be first coated with iodine-125 and then cutted into 3 mm long pieces. Based on this observation, a device for a batch-type procedure of manufacturing I-125 plated silver bars was designed and made. A design for simultaneous cutting and positioning of the activity-carrying bars into the titanium capsules is presented. This arrangement has been built and tested in trials. The tests confirmed that contamination of the source capsules can be minimized. Laser weldings of the titanium capsules were performed and the cross section of the welds examined by the use of an optical microscope.

Magnetic properties of a Ni50Mn35.5In14.5 Heusler ribbon were studied by ferromagnetic resonance (FMR) in the temperature range of 335–100 K. In the temperature region of 265–170 K, the FMR signal disappeared, in spite of the fact that this region comprised the main crystal transformation temperatures: Ms, Mf, As, Af. In the austenite crystal state, a weak antiferromagnetic interaction was observed, whereas ferromagnetism was detected in the low temperature martensitic state.

The structural and microscopic magnetic properties of the pseudobinary Sc(Fe1 xNix)2 system were studied by XRD and Mössbauer measurements in the range of 0.10 less-than or equal to - x less-than or equal to - 0.60. All investigated samples have the cubic C15-type structure. Both the magnetic hyperfine field on 57Fe and the Curie temperature decrease with increasing Ni content and the system becomes paramagnetic for x H 0.60 at room temperature. The form and temperature dependences of the Mössbauer spectra for 0.40 less-than or equal to - x less-than or equal to - 0.50 indicate the coexistence of paramagnetic and ferromagnetic regions in the samples and occurrence of magnetic clusters with a wide distribution of the Curie temperatures. As follows from the near to zero values of quadrupole shifts in Mössbauer spectra it results that a <100> direction is a local easy magnetization axis for x e 0.10.

The treatment of archives with gamma irradiation is an efficient and environmental friendly alternative for biological decontamination of large volume of archives. It substitutes the use of chemicals for conservation and contributes to safer workplaces. This work is targeting documents from recent archives where the value of information is not obsolete and may become an important historical and cultural testimony. For a successful treatment, an optimal absorbed dose has to be established. An excessive dose may damage papers and an insufficient one will not reduce bioburden to the desired level. An interdisciplinary team was performing various physical and chemical tests in order to evaluate deterioration of paper at high doses. In the case of natural disaster, it is not excluded the “emergency” treatment for documents in immediate danger of total destruction.
