Cucumber mosaic virus (CMV; family Bromoviridae, genus Cucumovirus) is the most cosmopolitan plant virus occurring worldwide. In the present study, leaf samples showing deformations, mosaics, and chlorotic spots symptoms were collected from naturally infected Basella alba, Telfairia occidentalis and Talinum fruticosum in a home yard garden in Ibadan, Nigeria. Total nucleic acid was extracted from leaves and used as template for cDNA synthesis. RT-PCR was carried out using CMV-specific primers targeting RNA-1 segment. Samples were also tested by RT-PCR using Potyvirus and Begomovirus genusspecific primers. DNA fragments with the expected sizes of ~500 bp were amplified by using CMV-specific primers; however, the expected amplicons were not produced using specific primers used for the detection of potyviruses and begomoviruses. The nucleotide and deduced amino acid sequences obtained for the isolates studied contained 503–511 nt and 144 aa, respectively. The isolates shared 81.9–85.3% nucleotide and 74.3–77.8% amino acid sequence identities with each other. The results of BLASTN analyses showed the highest identities of the isolates (80–93%) with CMV strains from Japan, USA and South Korea. Alignment of deduced partial protein revealed multiple amino acid substitutions within the three isolates and high identities with CMV subgroup I. Phylogenetic analyses putatively categorized the isolates in close association with subgroup IB isolates. The three isolates clustered together into a separate subclade, indicating possible new CMV strains. The results provide the first molecular evidence for CMV infections of T. fruticosum and B. alba in Nigeria and seem to show the possible presence of new strain(s). These findings also add three new hosts to the list of natural host range of the virus in Nigeria.

Serine inhibitors have been described in many plant species and are universal throughout the plant kingdom. Trypsin inhibitors are the most common type. In the present study, trypsin and chymotrypsin inhibitory activity was detected in the seed flour extracts of four Egyptian varieties of soybean (Glycine max). The soybean variety, Giza 22, was found to have higher trypsin and chymotrypsin inhibitory potential compared to other tested soybean varieties. For this reason, Giza 22 was selected for further purification studies which used ammonium sulphate fractionation and DEAE-Sephadex A-25 column. Soybean purified proteins showed a single band on SDS-PAGE corresponding to a molecular mass of 17.9 kDa. The purified inhibitor was stable at temperatures below 60°C and was active at a wide range of pH, from 2 to 12 pH. The kinetic analysis revealed a non-competitive type of inhibition against trypsin and chymotrypsin enzymes. The inhibitor constant (Ki) values suggested that the inhibitor has higher affinity toward a trypsin enzyme than to a chymotrypsin enzyme. Purified inhibitor was found to have deep and negative effects on the mean larval weight, larval mortality, pupation, and mean pupal weight of Spodoptera littoralis. It may be concluded, that soybean protease inhibitor gene(s) could be potential targets for those future studies which are concerned with developing insect resistant transgenic plants.

The rosaceous branch borer, Ospheranteria coerulescens, is an important pest of rosaceous trees. This insect feeds on the twigs and branches of living trees and causes their death. The characterization of the insect phenoloxidase (PO) is of interest when doing comparative investigations, and so that we may be able to understand its biochemical properties. When designing new methods of insect control such as the use of PO inhibitors, an understanding of the biochemical properties is fundamental. In this study, PO from hemolymph of the rosaceous branch borer was purified using ammonium sulfate precipitation, gel-filtration, and ion-exchange chromatography. The biochemical properties were characterized using l-dihydroxyphenylalanine (L-DOPA) as the specific substrate. The apparent molecular weights of the three isoforms of PO were determined by SDS-PAGE to be 85.23, 79.45, and 66.06 kDa. Optimal pH for PO activity was pH 8, and the optimal temperature was 45°C. Phenoloxidase lost less than 50% of its relative activity after a 60 min incubation at the optimal temperature. The effects of ions and chemical materials such as K+, Ba2+, Zn2+ and EDTA on PO showed that PO activity was strongly inhibited by Zn2+. The Michaelis constant (Km) and maximum velocity (Vmax) were 88.61 mM and 0.14 μmol/min, respectively. The inhibitory effects of kojic acid, 4-hexylresorsinol, and quercetin on PO were determined, and the IC50s (inhibitory concentration) were estimated as 23.31 for kojic acid, 35.75 for 4-hexylresorcinol, and 60.8 μM for quercetin. The inhibitory potency of kojic acid was 1.54 times higher than that of 4-hexylresorcinol and 2.58 times higher than that of quercetin. Phenoloxidase was effectively inhibited by 4-hexylresorcinol, and the inhibition type was competitive. The inhibition types of PO by kojic acid and quercetin were found to be mixed.

The role of cytochrome P450 monooxygenase in pyrethroid resistance was studied in different strains of the cotton bollworm, Helicoverpa armigera, from India. Filed collected strains of Nagpur and Delhi were compared to the laboratory reared population. The results showed a high resistance to deltamethrin, α-cypermethrin, and β-cyfluthrin. The results also showed that this resistance could be reduced by using piperonyl butoxide (PBO). The Nagpur and Delhi strains were found to have a 2.40 and 1.79 fold higher monooxygenase activity compared to a susceptible strain. A strong, positive correlation between monooxygenase activity and pyrethroid resistance was observed (r = 0.86 - 0.98). The relative expression of the housekeeping gene, EF-1α, and three P450 genes, was studied in the 5th instar larval midgut of the three strains. Out of the three P450 genes examined, expression of CYP6B7 mRNA was not detected in the midgut of the susceptible strain though it was highly expressed in the resistant strains. The midgut of the Nagpur strain had a 2.60 fold overexpression of CYP6B7 mRNA compared to the moderately resistant, Delhi strain. The mRNA of CYP4G8 and CYP6B2 were not overexpressed in either the Nagpur or Delhi strain. The results indicated that the elevated cytochrome P450 monooxygenase activity is associated with pyrethroid resistance in Indian strains of H. armigera, and CYP6B7 could be the P450 form responsible for pyrethroid resistance.

The future of food security in Africa is being severely threatened due to an exponential increase in population, which is almost three times the increase of food production. Maize production is constrained by stem borers which cause significant yield losses. Yield losses can be further compounded by higher temperatures due to climate changes, which are expected to increase the population of maize stem borers. While several methods have been employed in stem borer management, there is still significant damage caused by maize stem borers. This necessitates better control methods including the adoption of recent biotechnological advancement in RNA interference (RNAi) technology. This review highlights evidence of an increase in the stem borer population as well as the foreseen decline in maize production worldwide due to the effects of climatic changes. Furthermore, we have drawn attention to improved methods that have been used to control stem borers in maize production as well as a reluctant acceptance of traditional biotechnology in Africa. Finally, we suggest the application of alternative RNA interference techniques to breed maize for efficient pest control in order to achieve food security, improve nutrition and promote sustainable maize production.
