After duplicating the chromosome number by colchicine in sterile F₁ 16-chromosome hybrid T. pratense × T. diffusum some partially fertile plants with 32 chromosomes were found. Male fertility (viability of pollen grain) was from 69.3% to 86.2% (on average 81.8%), whereas female fertility estimated as seed setting after cross- and self-pollination was 21.8% and 6.9%, respectively. Male and female fertility as well as somatic chromosome number were examined in the F₂-F₄ generations. Selection for female fertility resulted in increasing seed setting in the first two generations (F₂ and F₃) and in decreasing it in F₄ generation. An average seed setting in the F₂-F₄ generations after cross-pollination amounted to 22.2%, 43.6% and 12.9%, respectively; after self-pollination it was 25.2%, 27.6% and 1.9%. In the F₂ generation all the plants had 2n=32 chromosomes. In the next generations there appeared aneuploids, among which 30-chromosome individuals were predominant.

Three diploid Trifolium (Fabaceae) species, T. campestre, T.fragiferum and T. montanum, were electrophoretically assayed to elucidate the range and organization of genetic variation in relation to the mating system, reproduction mode and longevity of the species. T. campestre is an annual self-pollinated species. T.fragiferum and T. montanum are cross-pollinated perennials. The former species reproduces both sexually and vegetatively, whereas the latter relies solely on sexual reproduction. Several populations of each species collected throughout Poland were surveyed for 15 enzymes. The measures of variation included: proportion of polymorphic loci per population, average number of alleles per locus, average gene diversity (He) and genetic distance (D). To describe the organization of diversity in each species, total genetic variation per locus (Ht) was calculated and partitioned into intra-(Hs) and inter-(Dst) populational components. Both cross-pollinated species had more polymorphic loci, higher numbers of alleles per locus and higher level of diversity than self-pollinated T. campestre. Most of genetic variation of the latter species was allocated among populations unlike in T.fragiferum and T. montanum.

Opis:
A method for the detection of segregating major genes based on the analysis of estimated marginal posterior major gene variance density was examined. The properties of the method were investigated using data sets simulated for a real population of laying hens consisting of eleven generations. Marginal posterior densities of model parameters were estimated by the Gibbs sampling approach proposed by Janss et al. (1995). With the data of about 4000 observations it was possible to detect a major gene responsible for one third of the genetic variance and one tenth of the phenotypic variance, irrespectively of the degree of dominance at the major locus. The inference based on the posterior marginal major gene variance can be sensitive to skewness of the data. It was shown that skewness of 0.2 can lead to a false detection of a major gene. The method is robust against a non-genetic mixture of normal distributions.

The genetic similarity of three species: Septoria tritici, Stagonospora nodorum and Stagonospora avenae f. sp. triticea - important pathogens in many cereal production areas worldwide was assessed by random amplified polymorphic DNA (RAPD) assay. In preliminary research DNA of 14, 9, and 7 monopyenidios- pore isolates of S. nodorum, S. tritici, and S. a. tritícea, respectively, were amplified by PCR with four primers. Afterwards the research was focused on three mono- pyenidiospore isolates from each species studied. The isolates of each species selected for the study varied in pathogenicity and were diverse geographically. PCR with the set of 14 selected primers resulted in 99 different bands, ranged from 180 to 2500 base pairs in length. Most primers in PCR (especially RAD11, RAD31, RAD32, RAD33) revealed uniform bands for isolates of S. a. tritícea, that allow to identify this species among the others. The cluster analysis using Unweighed Pair-Group Method with Averaging (UPGMA) revealed interspecies disagreement among the isolates ranging from 32 to 53%. The intraspecies disagreement ranges were 17-20%, 38-43%, 42-44% for S. avenae f. sp. triticea, S. nodorum and S. tritici, respectively. Cluster analysis classified isolates into three homogeneous clusters. Each cluster grouped isolates of one species according to their current taxonomie ranks based on spore size, colony morphology and host ranges. In addition, two of the clusters represented by isolates of S. nodorum and S. a. tritícea were distinctly separated at a lower linkage distance from the third one comprising isolates of S. tritici. A slight inconsistency found in grouping some isolates indicates that such groupings should be done with caution. The present study indicates that the PCR- RAPD assay is of a potential use in taxonomy of Stagonospora spp. and Septoria tritici as well as in molecular identification of casual disease agents.

Opis:
Gametoclonal variation among anther culture-derived plants of three barley genotypes was estimated on the basis of cytological analysis (DH1, DH2 generation), observation of morphological variants (DH2, DH3) and chlorophyll mutation test (DH2, DH3). Individual head rows were grown in the field to detect possible chimeric structure of regenerants and to assess the number of variants and mutations in each line. Spontaneously doubled plants were the most frequent class (70%) among regenerants and almost 90% of them were completely fertile. There was a difference in proportion of haploids produced by different genotypes, but the highest frequency observed did not exceed 21%. The remaining regenerants were tetraploid, and contained chromosomal mutations or chimeras. In total, there were about 15% of polyploids and plants carrying chromosomal aberrations (translocations, inversions) among DH1 individuals. The changes in chromosome number and structure were the main source of observed variation. The level of gene mutation induced in vitro was relatively low. No more than 1% of microspore-derived plants expressed visible morphological changes in DH2 progeny. Only two morphological variants derived from the Bruce cultivar proved to be homozygous mutants (dwarf type) stable up the to third generation. The frequency of DH plants carrying chlorophyll mutation was 5.8%, but most of them (82%) were chimeric and had only a small mutation sector. The level of gametoclonal variation depended on the donor plant genotype. The highest proportion of variants and mutations was observed among DH plants derived from the Bruce cultivar, while the lowest was recorded among plants regenerated from anther culture of the doubled haploid line H930-36. Mechanisms leading to the observed variation and implications resulting from the presented experiments concerning implementation of anther culture in barley breeding were discussed. It was concluded that this method resulted in a high frequency of spontaneous doubling, a low frequency of genetic changes, and being less time and effort-consuming than the ’Bulbosum’ technique, can be applied to most barley breeding programs.
