European white oak species (Quercus sect. Quercus) are known to share a substantial part of their nuclear and cytoplasmic genomes as a result of extensive interspecific hybridization and introgression. We studied natural populations of three species distributed in Slovakia, namely Q. robur, Q. petraea and Q. pubes- cens, which are largely sympatric but have contrasting ecological requirements, using a combination of leaf morphometry, neutral nuclear microsatellite markers (nSSR) and potentially adaptive amplified fragment length polymorphisms (AFLP). Bayesian analysis of population structure relying on nSSR revealed that the degree of admixture was relatively low in Q. robur but higher in Q. petraea and Q. pubescens. The inferred Q. robur ancestry closely correlated with morphometric scores of the canonical discriminant analysis based on 13 leaf traits, while for the other two species the correlation was much weaker. We identified two AFLP fragments associated with climatic variables, mainly with air vapour pressure and characteristics of temper- ature regime at the sites of origin. These associations indicate that climatic adaptation is the mechanism driving interspecific divergence and maintaining integrity of white oak species.

Masson pine (Pinus massoniana) is an important species for resin tapping in China. Molecular assisted selection (MAS) has been gradually introduced in to the selection and breeding of masson pine. Until now, the lack of connection between genome information and phenotypic data has hindered the breeding and selection of important chemical compounds of xylem resin. In this study, gas chromatography mass spectrometer (GC-MS) was employed for qualitative and quantitative analysis of chemical compounds of resin collected from 69 masson pine germplasm resources. Then, genome-wide association study (GWAS) was conducted for the terpenoids with higher heritability. A total of 15 primary compounds, including six diterpenes and nine monoterpenes/sesquiterpenes, were detected from xylem resin. By using five GWAS methods, 42 associated SNPs were identified from these masson pines. The P. taeda genome database was used for detecting candidate genes related to the associated SNPs. By using transcriptome data, several members of PKc_like super family, Cytochrome P450, AP2-ERF, F-box, and ABC transporter were found to be highly expressed in the masson pines with higher resin-yielding capacity. The terpenoids associated SNPs can be directly used for MAS. These candidate genes, especially the two F-box members, will contribute to the study of terpenoid biosynthesis mechanisms in the future.

Chinese fir (Cunninghamia lanceolata (Lamb.) Hook.) is a major timber conifer species in southern China. In this study, we aimed to capture the rarely advanced phenomenon for selfing in this species and illustrated the underlying molecular mechanism, especially the hub gene-regulated networks and pathways, by global transcriptome analysis assays (RNA-Seq). Self-pollination trials revealed a wide variation of selfing effects among parents. Parent cx569 produced a selfed family with the best growth performance at the seedling stage. The growth-based extremely advanced (AD) (n=3) and depressed (DE) variants (n=3; different types) were then subjected to comparative RNA-Seq. The transcriptome data revealed more than 5000 differentially expressed genes (DEGs) for each comparison group (AD versus DE). Weighted gene co-expression network analysis (WGCNA) further identified more than 80 important DEGs that were significantly associated with growth traits in each comparison group. A subsequent enrichment analysis showed that the identified DEGs belonged to six main types, including xylem metabolism-related, sugar and energy metabolism-related, plant hormone signal transduction-related, stress response-related, cytochrome-related, and transcription factor genes. Ten hub genes represented by the ERF071, MYB-relate 305, WRKY6, WRKY31, PER3, LAC4, CESA8, CESA9, GID1, and PR1 genes were co-identified between AD and DE variants. These genes exhibited rather different expression patterns between AD and DE variants, especially of the transcription factor ERF071 gene that presented a low transcript level in the AD seedlings with only 4.45% activity compared to DE's. While, the plant hormone signal transduction GID1 gene was significantly upregulated in AD by about 20-fold when compared to DE's, and fold change of the lignin biosynthesis-related PER3, CESA9 and LAC4 gene expression parallel reached to 10–15 times in an upregulation pattern in AD seedlings. The set of hub gene-linked interaction networks and pathways revealed in this study may be responsible for the rarely advanced phenomenon for selfing at the seedling stage in Chinese fir.

In addition to other traits, value of stands for breeding can be evaluated according to the number of plus trees and productive trees. According to this property, stands with the highest value for breeding are located in eastern Lithuania. Trees start to differentiate into breeding categories at the juvenile stage: in Scots pine stands on Pinetum myrtillosum sites, at the age of 6to 8 years and in Scots pine stands on Pinetum vacciniosum sites, at age of 10 to 12 years. According to radial increment, the trees start to differentiate into selection categories of productive, medium and minus trees, which often remain in these categories until mature age. The following four types of radial increment of trees were distinguished: (1) fast growth at the juvenile stage, followed by a gradual decrease, (2) fast growth at the juvenile stage, followed by a stable growth from age 20 to 30 years, (3) slow growth at the juvenile stage, followed gradual increase, (4) unstable growth all the time. In Lithuania, abundant seed yield used to occur in three to four year intervals. Populations with abundant flowering may produce approximately 10 kg of seeds (filled and empty) per ha, while the populations with weak flowering - 1 to 2 kg per ha. In mature Scots pine stands of stocking level ca 0.7, 6to 8% of the seeds are produced by plus trees, 24% by productive trees, 55 to 60% by trees of medium productivity and 10% by minus trees. Being suppressed, minus trees produce 2% of the seeds only.

Plant breeders have focused much attention on polyploid trees because of their resistance for forestry. To evaluate the impact of intraspecies genome duplication on NaHCO3 stress, a series of Betula platyphylla autotetraploids and diploids were generated from the same family. The growth, proline content and proline-associated gene expression of these autotetraploid individuals were compared with those diploid trees. Autotetraploids were superior in injury index and relative growth of height and base diameter compared to diploids. The proline content was higher in autotetraploid individuals compared to diploids. Gene expression data revealed autotetraploids were generally higher expression in BpP5CS1, BpP5CS2, Bp- P5CR1, BpP5CR2, BpP5CR3 and BpOAT and were lower expression in BpProDH and BpP5CDH compared to diploid trees. These results shed light on resistance variation in birch autotetraploidization and polyploidy breeding as a new approach for genetic improvement of birch trees.

Modern forest selection programs concern natural and protected populations of trees characterized by a great genetic variability and also production stands frequently deformed by human activity. In the latter, artificial regeneration is prevailing. Forest nurseries working in the organizational structures of State Forests can guarantee the proper provenance (genotype)of seeds and the quality of seedlings obtained from them. The condition, however, is a close uniformity in the conservation of forest gene resources, and selection breeding of forest trees. The success of the program depends also on the development of modern infrastructure for forest nurseries and applied technologies. The latter task was the field of activity of Professor Bolesław Suszka, Ph.D., and his many-year research and organizational and didactic activity in Poland and abroad. The methods in long-term storage and their pretreatment in case of dormant seeds, developed by Professor Suszka, created the theoretical and practical basis for the conservation of forest gene resources not only in Poland.

Present status of a postglacial relict – Salix myrtilloides L. was investigated in north central Poland at 27 localities. Consequently 3 sites were confirmed, 4 were not confirmed, and the remaining 20 were considered to be extinct. In comparison to the historical distribution, ca. 74% of the sites existing 50 years ago do not exist today in the studied region. The confirmed populations were found near Oporówek (ca. 1600 stems), Rakowiec (ca. 100) and Bobrowo (15). At three sites confirmed in this study, the existence of vigorous hybrids between S. myrtilloides and S. aurita L. (S. ×onusta Besser) were observed. The most important threats to Salix myrtilloides L. populations in the studied region were assessed to be hybridization with other commonly occurring willow species, site isolation and habitat destruction. Natural habitat protection with adequate buffer zone and gene pool protection in a form of plantations was suggested as a conservation strategy. As an alternative strategy active protection ‘insitu’ was proposed based on cutting competitive common species of willows.

In the Forest Gene Bank cold stores, both long-term and strategic gene resources are stored to be used during periods of poor seed production. Beech seeds of the following harvests were evaluated: 1995, 1998 and 2000. Quality of seeds was determined with different methods from the date of reception to FGB through preparation for long-term storage, storage in cold stores and the presowing treatment. The usefulness of staining tests for rapid evaluation of beech seed viability is not sufficiently sure. The floating test in water significantly improves the level of seed purity. The current method of seed drying does not remain without influence upon beech seeds and causes a slight decrease of viability. Next to beech seed lots of rapidly declining quality in the FGB are stored seed lots whose viability does not decrease during identical technological processes. Seed lots of low initial viability should be eliminated.

English yew (Taxus baccata L.) is a rare tree species in Austria and gene conservation forests reserves are used to maintain English yew populations by silvicultural treatments. This paper describes the current situation of an English yew population at “Stiwollgraben”in Austria with regard to stand structure, vitality and natural regeneration. The area is one of the most important sites in Austria as it consists of 2236 yews.The vitality condition of the yew is very good, and more than 79% of the yews have been assessed as very vital to vital. The potentiality for natural regeneration (13019 one-year seedlings ha–1) is very high, but not all height classes are represented. This indicates a high survival ability of English yew at this site. Three different treatment strategies are described to maintain the yew population. The future effects of these treatments are discussed in the light of the environmental requirements of English yew.

At present, Norway spruce stands occupy 22% of the total forest area in Lithuania. Most of them are located in the northeastern highland (Žemaitija) and the central part of the country. 25% of the spruce-dominated stands are pure, the rest are mixed. Natural calamities like storms, droughts, and pests cause substantial damage and occur quite frequently. About 40 thousand ha of stands are cut after each storm. The mean wood yield of the spruce stands is 304 m3 ha-1, and the current annual increment is 6.2 m3 ha-1. The climatic conditions of Lithuania are variable enough to cause differentiation of habitats. For Norway spruce, 6 provenance regions have been established. The national gene conservation programme is based on (a) in situ genetic reserves, seed collection stands, and selected genotypes, and (b) ex situ clonal archives, seed orchards, experimental plantations, and gene bank collections. The present-day breeding of Norway spruce comprises family tests of populations for individual and population selection, and plans for inter-population hybrids. A strategy for Norway spruce breeding has been approved for the years 2004-2013. There is a genetically diverse material for future breeding: long-distance provenance tests, and population and family tests. Assessments of two provenance tests (aged 9 and 17 years) in central Lithuania revealed superior performance of central and northeastern Polish provenances: superior height, good stem quality, and late bud-burst in spring. This may be attributed to the favourable effect of transfer: avoidance of spring frosts (late bud-burst and good stem quality), and utilisation of the later part of the growing period for growth (late bud-set and superior height). Norway spruce is a climax species with different domestic and Darwinian fitness. Therefore, we suggest that the domestic fitness of local genotypes may be improved by introducing a few Polish clones in Lithuanian breeding populations.

Picea abies (L.) Karst is one of the most important coniferous species of Europe from both ecological and economical points of view. Traditional methods for the gene pool conservation and biodiversity maintenance in forest ecosystems have been practiced in many countries. For progress in this field using highly polymorphic genetic molecular markers is needed. Our goal was to demonstrate the utility of two polymorphic mitochondrial markers mt15-D02 and nad1 b/c in identification native Norway spruce stands. This molecular markers were tested in 1401 individuals from 59 Polish Norway spruce populations. We detected three alleles, which are called1, 2 and3, for locus mt15-D02 and two alleles , which are called1 and2, for locus nad1 b/c in our material. All five variants of alleles indicate the natural origin of P. abies. Result of this study shows that molecular marker mt15-D02 is easy to use and more informative in compare to marker nad1 b/c.

The paper describes the measures adopted by the Czech State Forests for the conservation and reproduction of the Norway spruce gene pool in the Moravian-Silesian Beskid Mts. In addition to the assurance of natural regeneration in genetically valuable stands and the use of high-quality autochthonous stands as seed sources for artificial regeneration, the measures include selection of plus-trees, establishment of seed orchards, seed stands and clone archives, and identification of gene pools. Seeds of the most valuable regional populations are deposited in a seed bank.

This paper presents the newly suggested long-term breeding strategy for Norway spruce in Lithuania to share with the optimisation approach, which may be useful in synchronising the long-term breeding and gene conservation plans in neighbouring regions. The breeding programme in Lithuania is at a stage where the long-term plans need to be developed. Our strategy is to proceed with a closed-nucleus breeding population which is structured into subpopulations according to adaptation zones (4-5 subpopulations). The size of each subpopulation is 50 unrelated and progeny-tested individuals. The breeding cycle consists of double- pair mating and a balanced within-family selection forward. The candidates will be clone-tested. Advanced genotypes will be selected into an advanced nucleus; an isolated foreign breeding population is planned. Positive assortative mating is used for deployment. An experimental "shuttle-breeding" strategy may be tested: selection with avery high intensity, controlled crosses, nursery tests for vigour and growth rhythm, cloning by hedges into short-rotation clonal plantations.

The work outlines the directions in the past and present research into the variability of Norway spruce in Poland, and presents the results of provenance experiments concerning the genetic and breeding value of provenances tested in the Polish range of spruce distribution. The Istebna race proved to have a good value, however, so far only the progeny of single stands have been tested. It is thus necessary to determine the range of distribution for this race. The altitude of the location of mother stands significantly correlates with the genetic and breeding value of their progeny cultivated at different altitudes. This suggests that the altitude and exposure of plantations to be established in mountain forest belts should be specified for individual provenances (altitudinal zoning). New inventory provenance tests should be run under varied site conditions to assess both the genetic value and plasticity range of the provenances. In view of the biotic and abiotic threats facing spruce, there is a need to work out detailed programmes designed to preserve its genetic resources in gene banks and in vivo archives.
