Opis:
The paper is focused on application of sensitivity methods to analysis of signaling pathway models. Two basic methods are compared: local, based on standard sensitivity functions, and global, based on Sobol indices. Firstly, a general outline of modeling of signaling pathways by means of ordinary differential equations is briefly described. Afterwards, the methods of sensitivity analysis, known from literature, are introduced and illustrated with a simple example of a dynamical system of the second order. Subsequently, the analysis of the p53/Mdm2 regulatory module, which is a key element of any pathway involving p53 protein, is presented. The results of this analysis suggest that no single method should be chosen for investigation of any signaling pathway model but several of them should be applied to answer important questions about sources of heterogeneity in cells behavior, robustness of signaling pathways and possible molecular drug targets.

Opis:
The paper addresses the problems of the sensitivity analysis and optimal design of multi-span sandwich panels with a soft core and flat thin steel facings. The response functional is formulated in a general form allowing wide practical applications. Sensitivity gradients of this functional with respect to dimensional, material and support parameters are derived using adjoint variable method. These operators account for the jump of the slope of a Timoshenko beam or a Reissner plate at the position of concentrated active load or reaction, thus extending the sensitivity operators known in literature. The jump of slope is the effect of shear deformation of the core. Special attention is focussed on sensitivity and optimisation allowing for variable support position and stiffness, because local phenomena observed in supporting area of sandwich plates often initiate failure mechanisms. Introducing optimally located elastic supports allows to reduce the unfavourable influence of temperature on the state of stress. Several examples illustrate the application of derived sensitivity operators and demonstrate their exactness.

Opis:
Evolutionary solid bodies undergoing changes of mass, of properties, and of shapes are considered in models of growth and adaptation and similarily in structural optimisation. A fundamental separation of different growth phenomena and a subsequent parametrisation using independent design variables for the amount of substance as well as for molar mass and molar volume facilitates an efficient formulation of the design space. Thus, the effects of design variations, i.e. change of amount of substance, on the variations of the structural response, i.e. the deformation in physical space, can be clearly described. Overall, a novel treatment of growth processes based on an evolution of the amount of substance is outlined. The parallelism of variations in physical and design space are highlighted and compared with the multiplicative decomposition of the deformation gradient into a growth and an elastic part incorporating an incompatible intermediate configuration. This drawback is overcome by a compatible manifold based on material points modelling the amount of substance outside of any geometrical space.

Opis:
An elastic structure subjected to thermal and mechanical loading with prescribed external boundary and varying internal interface is considered. The different thermal and mechanical nature of this interface is discussed, since the interface form and its properties affect strongly the structural response. The first-order sensitivities of an arbitrary thermal and mechanical behavioral functional with respect to shape and material properties of the interface are derived using the direct or adjoint approaches. Next the relevant optimality conditions are formulated. Some examples illustrate the applicability of proposed approach to control the structural response due to applied thermal and mechanical loads.

Opis:
Anisotropic rotor configurations influenced by the presence of a large number of geometrical parameters in a permanent magnet assisted synchronous reluctance (PMASR) motor pose design challenges in obtaining a robust geometry satisfying the requirements of reduced torque ripple and high torque density. Therefore, the purpose of this work is to perform detailed geometrical sensitivity analysis of a 36 slot/4 pole permanent magnet assisted synchronous reluctance (PMASR) motor using h-indexing and level sensitivity analysis in order to specify a guideline for designers to prioritize the design variables for optimization. Systematic multi-level design optimization for multiple objectives is implemented by an NSGA-II algorithm aided by the finite element analysis tool, hardware prototyping and experimental validation. The optimized designs also exhibit better structural and thermal characteristics.

Opis:
The rotating machines with overhung rotors form a broad class of devices used in many types of industry. For this kind of rotor machine in the paper, there is investigated an inf luence of dynamic and static unbalance of a rotor, parallel and angular misalignments of shafts, and inner anisotropy of rigid couplings on system dynamic responses. The considerations are performed through a hybrid structural model of the machine rotor-shaft system, consisting of continuous beam finite elements and discrete oscillators. Numerical calculations are carried out for parameters characterizing a heavy blower applied in the mining industry. The main goal of the research is to assess the sensitivity of the imperfections mentioned above on excitation severity of rotor-shaft lateral vibrations and motion stability of the machine in question.

Opis:
The problem discussed in the paper is numerical modeling of thermal processes in the domain of biological tissue secured by a layer of protective clothing being in thermal contact with the environment. The cross-section of the forearm (2D problem) is treated as non-homogeneous domain in which the sub-domains of skin tissue, fat, muscle and bone are distinguished. The air gap between skin tissue and protective clothing is taken into account. The process of external heating is determined by Robin boundary condition and sensitivity analysis with respect to the perturbations of heat transfer coefficient and ambient temperature is also discussed. Both the basic boundary-initial problem and the sensitivity problems are solved by means of control volume method using Voronoi polygons.
