The article considers the effect of chitosans of different origin, molecular weight, and degree of deacetylation on the dynamics of phenolic substances in the leaves of Fragaria ananassa Duch. These substances potentially act as agents in plant responses to biotic stresses. We showed that the daily variations in individual phenolic compounds are most pronounced in the group of ellagitannins and kaempferol-3-b-D-[6-O-(E)-coumaroyl]-glucopyranoside. The most stable indicators were concentrations of kaempferol glycoside and ellagic acid pentoside. The strongest plant response to leaf treatment with low- and high-molecular-weight chitosans was a significant increase in the concentration of hexahydroxydiphenyl (HHDP)-glucose in leaves, peaking at 12 h after treatment. Treatment of F. ananassa with various forms of chitosans resulted in different concentrations of the basic phenolic substances. In particular, the plant response to a single treatment with 0.4% low-molecular-weight chitosan (LMC) was accompanied by a significant decrease in the ellagic acid concentration, after a slight increase in the first hour after treatment. The opposite effect was observed in plants after treatment with high-molecular-weight chitosan (HMC). We also determined the daily dynamics for the quantitative and qualitative composition of phenolic compounds in the control group of plants. By using the Biplot analysis, we showed a clear difference between phenol concentrations in the evening (0, 24, and 48 h) and in the morning (12 h). The strongest difference was an increase in the concentration of HHDP-glucose, galloylquinic acid, ellagic acid pentoside, kaempferol glycoside, and ellagic acid in leaves in the morning, followed by a decrease in the evening and nighttime, due to the physiological reactions of plants to external abiotic and biotic factors.

Opis:
Eichhornia crassipes (water hyacinth) possesses several biological properties. Periplaneta americana (cockroach) is a household insect pest that can disseminate pathogens. The present study analyses the chemical constituents of water hyacinth leaf methanol extract and its in vitro effects on antioxidant and esterase enzymes in cockroaches. Water hyacinth methanol extract (WHME) was subjected to gas chromatography-flame ionization detection (GC-FID). Crude enzyme preparations were obtained from nymph and adult cockroaches. In vitro effects of different concentrations (10–80 μg/ml) of WHME on superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase (GST), acetylcholinesterase (AChE), and carboxylesterase (CE) in crude enzyme preparations were spectrophotometrically measured using cypermethrin (CYP) and chlorpyrifos (CPF) as reference insecticides. The GC-FID chromatogram of WHME showed the presence of gallic acid, para-hydroxybenzoic acid, eritosin, gentisic acid, catechin, protocatechuric acid, quercetin, and eugenol. The WHME and CPF reduced catalase in adults and SOD in nymphs, while CYP and CPF reduced SOD activity in the adults of P. americana. GST was significantly increased by WHME in both nymph and adult cockroaches. AChE was reduced by WHME, CYP, and CPF in nymphs, while CE was increased by WHME in nymphs and adults as compared to the control. In conclusion, our study indicates a high phenolic content in the water hyacinth leaf methanol extract, which increased the activities of the antioxidant enzymes AChE and CE in P. americana. The results also show that the efficacy and the mode by which the extracts could induce toxicity depend on the growth stage of the cockroach.

Opis:
Green synthesis is a biocompatible and stable method of producing zinc oxide nanoparticles (ZnONPs).In the present study, ZnONPs were biosynthesized using Fumaria parviflora extract by the green method, and the antibacterial and antioxidant properties of these NPs were evaluated. The characteristics of the synthesized ZnONPs were determined by ultraviolet-visible spectrophotometry (UV-VIS), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The antioxidant activity of the NPs was tested by the α-diphenyl-β-picrylhydrazyl (DPPH) method. Antibacterial properties of the synthesized ZnONPs were evaluated against Staphylococcus aureus and Escherichia coli by disc diffusion and microdilution methods. The results of UV-VIS spectroscopy revealed an absorption peak at 370 nm. XRD results showed the formation of a hexagonal wurtzite structure, and SEM analyses demonstrated that ZnONPs had a spherical shape with an average size 42 to 60 nm. Free radical scavenging capacity of ZnONPs was assessed using the DPPH assay with varying concentrations of ZnONPs, and scavenging activity was observed with IC50 of 30.86 μg/ml. In the antibacterial assay, the inhibition zone of the synthesized NPs at 100 μg/ml concentration for S. aureus (24.6 ± 0.72) was greater than that of the antibiotics vancomycin (23 ± 0.51) and a mikacin (13 ± 0.40) and was greater for E. coli (13.2 ± 0.81)than that of vancomycin (12 ± 0.41) (P # 0.05). The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) values of ZnONPs for S. aureus and E. coli were 1.56 and 3.125 μg/ml, and 6.25 and 12.5 μg/ml, respectively. The biosynthesized ZnONPs showed strong antibacterial and antioxidant activities.

Cayratia trifolia Linn. Domin Syn. Vitis trifolia (family: Vitaceae ) contains polyphenols as major bioactive compounds. Plant polyphenolics, such as stilbenes, possess a broad spectrum of pharmacological and therapeutic effects, including antioxidative, anticancerous, antiatherosclerotic, cardioprotective, hepatoprotective, and neuroprotective effects. Elicitation is one of the important strategies to enhance secondary metabolite production in cell suspension cultures. In the present study, the effect of two biotic elicitors, namely one fungal (Fusarium sp., and Helminthosporium sp.) extract and another higher plant-originated gum (Anogeissus latifolia ), was evaluated for enhanced production of stilbenes in cell suspension culture of C. trifolia. For the fungal elicitor, the cell suspension cultures of C. trifolia were treated with varying concentrations of Fusarium extract (FE) and Helminthosporium extract (HE) on the 7th day of culture. Increasing concentration of FE in the medium resulted in slightly decreased cell growth but increased the production of stilbenes, with the optimal concentration at 1000 mg/l. This increase in the yield of stilbenes was -5 times (9.30 mg/l) higher than the control value. HE was less effective than FE in increasing stilbene production at optimal concentrations. The addition of 50 mg/l A. latifolia plant gum sieved through a 150 mesh (0.1 mm) to C. trifolia cell culture enhanced stilbene accumulation and yield by 8- to 9-fold over the control value.

Ephedra alata is an important Egyptian medicinal plant with enormous applications in pharmaceutical industry. Shake-flask suspension cultures of E. alata were established from internodal segment-derived callus. The suspension cultures were treated with two biotic elicitors, namely Aspergillus niger and yeast extract, to enhance the synthesis of ephedrine – the bioactive constituent of E. alata. Ephedrine was quantified using the high performance liquid chromatography technique. Ephedrine concentration was not directly related to biomass production. Aspergillus niger and yeast extract enhanced the production of ephedrine by 2.5- to 7-fold after different durations of incubation as compared to ephedrine content in the mother plant. Treatment with the fungal elicitor A. niger extract induced the maximum accumulation of ephedrine at the concentration of 1.67%; it also increased ephedrine biosynthesis by 7-fold in 24-day-old culture as compared to the ephedrine content in the mother plant and caused higher ephedrine biosynthesis than that in the control treatment without elicitors. The present protocol could be applicable for the large-scale production of ephedrine from E. alata.
