Opis:
Cryptosporidium parvum is a coccidian parasite that affects millions of people worldwide. Clinical outcome of human cryptosporidiosis differs between immunocompetent and immunodeficient individuals. C. parvum is responsible for causing protracted and life-threatening diarrhea, biliary, and pulmonary infections in immunocompromised persons, especially in patients with AIDS. Though no effective treatment has been found so far, early diagnosis may be useful in controlling the infection. Thirty-eight stool specimens obtained from 35 HIV-positive patients admitted to the Clinic of Infectious Diseases in Poznań, Poland, were examined for the detection of oocysts, coproantigen and DNA of Cryptosporidium using standard microscopic, immunologic and molecular diagnostic methods. The presence of Cryptosporidium was detected in 10 HIV-positive patients. Oocysts, coproantigen and DNA of this parasite were identified solely in one specimen while Cryptosporidium DNA was detected in 8 specimens. Cryptosporidium coproantigen was found only in one sample. Although, the PCR was the most useful technique in the detection of Cryptosporidium in HIV-positive patients it should be noted that PCR has many pitfalls and needs to be carefully controlled to avoid both false positive and false negative results.

As a result of widespread resistance to chloroquine (CQ) and sulphadoxine-pyrimethamine (SP), artemisinin-based combination therapy (ACT) has been recommended as a first-line anti-malarial regimen in Côte d’Ivoire since 2005. A thorough understanding of the molecular bases of P. falciparum resistance to existing drugs is therefore needed. The aims of this study were to analyze the in vitro sensitivity of P. falciparum field isolates from Abobo to CQ, pyronaridine (PYR) and dihydroartemisinine (DHA), and to investigate the polymorphisms associated with drug resistance. The standard in vitro drug sensitivity microtechnique recommended by the WHO was used to assess the sensitivity of Plasmodium falciparum isolates collected in December 2006. The Pfcrt haplotype 76 was analysed by PCR-RFLP while Pfatpase 6 amplification products were sequenced. Associations between drug sensitivity and parasite gene polymorphisms were evaluated with Cohen’s kappa test. The correlation between the IC50 values for different drugs was assessed by the coefficient of determination (r2). Significance was assumed at p<0.05. Of 128 in vitro tests performed, 112 (87.5%) were successful. Of the isolates, 56.2% were resistant for CQ and 48% for PYR. One isolate (3.6%) demonstrated reduced DHA sensitivity (IC50 higher than 10 nM). The mutant K76T pfcrt codon, present in 90% of DNA fragments analyzed, was associated with CQ-R (ĸ=0.76). The N669Y (16.1%), D734Y (28.6%) and D734H (1.8%) isolates were found to have mutant Pfatpase6, however, these mutations were not associated with diminished DHA sensitivity (k=0.01). These high levels of antimalarial drug resistance in Abobo (Côte d’Ivoire) demand further studies of drug efficacy across the whole country.

Within 1992-2000. a total of 181 Białowieża Forest bison were examined from two winter herds. Twelf parasitic arthropod species were observed, a high infestation being typical of Demodex bisonianus, Chorioptes bovis, Ixodes ricinlls, Dermacentor reticulatus, and Bisonicola sedecimdecembrii. The infestation in section 422 herd being higher for B. sedecimdecembrii, I. ricinus, D, reticulatus, Ch. Bovis, D. bisonianus was slightly more prevalent in the section 391 herd, the intensity being, however, lower than that in the other herd. Among the remaining arthropods found in the Białowieża Forest European bison, some Lipoptena cervi occurred in both herds. Demodex sp. and Sarcoptes scabiei were recorded only in the section 422 herd, Ixodes persulcatus was present only in the section 391 bison and those kept in the reservation, while D. bovis, Psoroptes ovis. and Melophagus ovinus were found in the reserve bison only. In the present study, the largest differences in the extent of infestation involved the hair-dwelling arthropods (B. sedecimdecembrii, I. ricinus).

Within 1994-2000, sections of skin collected from eleven areas on the body of 650 wild boars and 85 pigs living in the northern Poland and in the Great Poland were examined. The D. phylloides infestation in the wild boar was observed to be very heavy (prevalence 32%, mean intensity 74 ind:). In contrast, the domestic pig turned out to be free of the parasite. The wild boar D. phylloides infestations are symptomless. The parasite is relatively rare in the domestic pig, although the high animal density typical of farm stocks should aid in its spreading. Compared to that in the wild boar, the pigs affected show associated skin symptoms. Where demodicosis did occur, most often the entire population was suffering of the condition. Most probably, the wild boar is the original host of D. phylloides. The hair follicle mile in question is very frequent in the wild boar, but produces symptomless infestations. The parasite occurs more seldom in the domestic pig, but - in contrast - the infestations are much more violent.

Demodex flagellurus Bukva, 1985 is one of two known demodecid mites of the house mouse Mus musculus Linnaeus, 1758, in which it is observed in genital area. Skin fragments of 30 house mice from various regions of Poland (residential buildings in Gdynia and Gdańsk, rural region in Wielkopolska-Kujawska Lowland) were examined. The mites were noted in 25.0% of the mice, with mean intensity of 48.0 and intensity range of 2–103. D. flagellurus demonstrated the differentiated occurrence in host populations.
